Operation theatre (OT) is a place where surgical operations are performed under the principle of aseptic surgery. An ideal operation theatre :
    Should be located near the work and intensive care areas of the hospital.

    Should be independent of general traffic. One way traffic flow is maintained to ensure sterility. 

    If possible, air within the OT should be under mild positive pressure so that when the surgery room opened, air flows out of rather than into the room.
 
    Floors, walls and ceilings should be constructed of impervious material that can easily be cleaned and disinfected.
 
    The lighting of the OT should entirely be dependent on artificial light.
 
    Room temperature should be about 70^ºF with a relative humidity of about 50% provides a better environment for the surgeon and patient.

Ionization Chambers for Gases

Generally these type of instruments are known as dose-rate meter (e.g., the so called “cutie-pie”). Radiation intensity (i.e., a constant stream of particles or photons) gives rise to a continuous series of pulses, and if these are allowed to merge, they form a weak electric current, which may be amplified and registered by an electronic circuit. The final scale reading will then be a measure of the energy dissipated in the following chamber per unit of time by the ionizing particles or photons. In gas-flow counters, the radioactive samples are placed inside the detector which will be transfused by a gas at atmospheric pressure. This instrument counts particles of low energy, such as the b particles from 14C, effectively (Window-less counting).

Principle of DNA Ligase Mechanism

The process of connecting two pieces of DNA together is called ligation, and is catalyzed by an enzyme called ligase. The ligase used most often in molecular biology is derived from the T4 bacteriophage, and uses ATP to supply the energy necessary for the reaction. In addition to ATP, T4 ligase requires DNA with a 5´-phosphate group and free 3´-hydroxyl groups.

ABSTRACT

Bacopa monnieri (L.) or Herpestis monniera, commonly known as brahmi, purifies the blood, and is useful in scabies, leucoderma, syphilis, diarrhoea and pyresis. As Brahmi is a major ingredient in several ayurvedic preparations, there is growing demand for steady supply of planting materials. Conventional propagation methods are less efficient than in vitro propagation. However, there are only a few reports on the in vitro propagation of this plant. Therefore, an attempt was made to evolve an improved protocol for the in vitro propagation of Bacopa. Shoot tips and nodal buds along with a portion of the stem excised from field grown disease free plants were taken as the explants. After three weeks of establishment on MS basal medium, the sterile cultures were sub-cultured on MS medium supplemented with different concentrations of BA (0.5,1.0 and 1.5mg l^-1). Rooting was induced on in vitro proliferated shoots by culturing on MS basal medium with NAA 1.0 mg l^-1 and IBA 0.5 mg l^-1. Treatments with BA 1.0 mg l^-1 and IAA 1.5 mg l^-1 recorded the maximum number of shoots (40.9 and 35.2, respectively). The height of the shoots was also significantly influenced by the concentrations of IAA (1.5 or 1.0 mg l^-1) in combination with BA (1.0 mg l^-1), recording 5.4 and 5.5 cm. The shoots with an average length of 3.0 to 4.0 cm were kept for rooting. NAA (1.0 mg l^-1) in combination with IBA (0.1, 0.5, 1.0 or 1.5 mg l^-1) produced significantly greater number of roots. The length of the roots was also greater in this treatment. The plantlets were transferred to pots in a greenhouse and later to the field with ninety per cent survival.

Mysore raspberry is a shrub growing to 1-2.5 m tall. The stems are whitish velvety at first, becoming smooth green to purple later. Leaves are compound with 5-11 leaflets. The leaflets are dark green above, densely pale grey to white velvety beneath. The flowers are five dark pink to red petals. The fruit is densely grey velvety, dark red at first, ripening to black. The fruits are commonly harvested from forest for local use as a food and is also used as a medicine. Mysore raspberry is grows wild throughout the temperate Himalayan region. This species is successfully introduced North America. It resembles the raspberry.

In spite of late introduction, different forms of chilli (Capsicum spp.) did spread throughout the length and breadth of India. Capsicum chinense commonly known as Ney Mulaku, Karanampotti Mulaku is gaining popularity in the state due to increasing demand from Maldives during Ramzan period. Hence the crop is also known in yet another name Mali Mulaku. This species is characterized by perennial and spreading nature with medium sized oblong to round, fleshy and highly pungent fruits. When the crops are under moisture stress, the principles involved are conservation of soil moisture and reduce the evapotranspiration from crops. The moisture conservation can be achieved by suppressing soil evaporation using mulches (Fig. 14.1). The evapotranspiration can be reduced using anti transpirants. These are chemicals sprayed on crops. Some antitranspirents close the stomata of the leaves to reduce evapotranspiration. Some chemicals increase the reflectance of radiation from the crops, thereby reducing heat load on crops. When crops are grown under irrigation in isolated patches, micro shelter belts can be provided across the predominant wind direction to reduce the increased evapotranspiration from advection. Advection is phenomena of horizontal transport of heat due to strong hot and dry winds.
