ABSTRACT

Role of cytokinins in the regulation of sink development in potato was studied by transgenic approach. Transgenic potato plants over-expressing Agrobacterium native ipt (gene for isopentenyl transferase, rate limiting enzyme in cytokinin biosynthesis) under the transcriptiona l regulation of tuber specific patatin promoter were developed. Transformants obtained showed normal phenotype, while the cytokinin levels showed significant increase over the control plants for the various cytokinins tested, viz., trans zeatin riboside (tZR), dihydrozeatin riboside (DHZR) and isopentenyl adenosine (iPA). The primary transformants were hardened and tested under green house conditions. Green house studies showed a significant increase in the number of tubers in ipt transformants when compared to the wild plant as well as GUS transformant. However, the total weight of tubers did not show appreciable difference between transformants and control plants. Reduction in the weight of individual tubers appears to be due to source limitation.

Introduction

Shock is a complex physiological syndrome marked by widespread circulatory irregularities that result in inadequate tissue perfusion. In other words, it is a clinical condition caused by an insufficient delivery of oxygen to tissues or the tissues' inability to correctly use oxygen. It is a complication of trauma and diseases typically seen in emergency rooms, such as heart failure, inflammatory conditions (e.g., pancreatitis), and sepsis. In shock, tissue perfusion and oxygen delivery are insufficient to support the tissues' baseline metabolic demands. Whatever the cause, shock is distinguished by reduced blood flow, which is typically accompanied by low blood pressure (hypotension).

In India, majority of the people live in rural areas and are engaged in agriculture, earning a subsistence wage. Development which has been focused on them seems to have just passed by them. In India out of the 147.90 million rural households around 60 per cent are cultivation households. Eighty-eight per cent are headed by small/marginal farmers with holding less than 2 hectares (Throat, 2005). The tube rating chart provides important information on the maximum safe exposure time that can be used with specific mA and kV setting. The fruit of pome class referred as Cydonia oblonga (also Pyrus cydonia) is an ancient fruit and yet its significance is not fully recognized and crop has received very less attention. There is hardly any mention of this old fruit in respect of its merit, its cultivation or in the form of some standard literature. One reason could be that amongst pome fruits where quince is placed, better fruit form, raw eating quality persisted, which markedly restricted quince improvement. Quince fruit lacks dessert class except when cooked but is commercially used for flavouring, in preparation of jellies marmalade and preserves. However in near East and Persia, quince cultivation is highly improved through the selection of better varieties which are eaten raw.

Biotechnology provides significant opportunities and greater effects in horticulture, where even minor changes such as quality, colour, aroma, flavour, structure, disease-pest resistance and postharvest behaviour bring tremendous achievements commercially. Tissue culture, genetic engineering, molecular breeding, biofertilizers, biopesticides and postharvest biotechnology are important aspects of biotechnology in horticulture such as fruit, vegetable, ornamental, spice, medicinal, aromatic and plantation crops.

Tissue Culture

Tissue culture is the true reality of the plant biotechnology. The annual demand of tissue cultured production constitutes nearly 10 per cent of the total world biotech business, amounting to 15 billion US dollars. The global production of tissue culture industry is expanding @ 15% per annum (Chadha, 2000). In India, more than 130 tissue culture units are presently producing > 300 million plantlets per annum (Choudhury and Tejaswini, 2000).

India is the second most populous country in the world with a population of 1.3 billion and is expected to reach 2 billion by 2050. Though the country transformed itself from a food deficit nation to surplus donor in agricultural commodities in a short span of 70 years, it also encountered several stumbling block along the way. In the process, what went unnoticed was the hidden hunger resulting in malnourished population with different kinds of ailments. ‘Hidden hunger’ manifested due to an imbalanced diet, lacking in micronutrients–vitamins, minerals and other essential diet components. Serious among them, which plague the country even today are iron deficiency causing anaemia, Vitamin A deficiency (VAD) causinig blindness in children and increased risk of other illnesses and death. Anaemia and calcium deficiency among women and stuntend growth in children are the other diet related diseases. About 10% of population are also suffering due to over feeding or mal-noursihment. Economic burden and loss of human productivity due to lifestyle diseases is taking a toll in India during the last couple of decades.

At present, the world can very broadly be divided into two main categories. (i) Industrialized countries where food is plentiful and the diet is varied, and the most prevalent diet-related disorders are cardiovascular disease (CVD), cancer, obesity, diabetes and osteoporosis. (ii) Developing countries, where food is scarce and malnutrition is a major problem or where traditional foods and cultural practices are being replaced by western-style diets resulting in lifestyle diseases/disorder. India falls in the second category. Diversified food habit is one of the direct impacts of increasing disposable income in India. Global epidemiological studies with reference to food habits have shown that increase in lifestyle diseases are directly related to reduced consumption of fruits and vegetables and increased consumption of animal food (meat and animal fat).

Introduction

The impacts on the various facets of life – agriculture and food processing – need no longer be addressed in a way that is well established. Nearly every aspect of the planet has shown its effect at different rates, both positively and negatively. As a subtropical region, India is also witnessing different variations in weather and an increase in temperature that affect farming. Only in recent years has the potential to reduce transpiration from plants by the use of chemicals been further explored. These antitranspirants can either control perspiration by filming the leaf surface or by manipulating the aperture of the stomach. Antitranspirants are the materials or chemicals that reduce the water loss from plant leaves by reducing stomata size and number. During transpiration, almost 99 per cent of the water consumed by the plant is lost.

Introduction

Economic Importance
The family is of no specific economic importance except that of some well known decorative plants it contains. Some species of Primula may cause contact dermatitis. Anagallis arvensis is used for curing snakebite, leprosy, gout and even hydrophobia. Cyclamen purpurascence contains glucoside cyclamen which is poisonous. Some of the commonly cultivated ornamental plants of Primulaceae are the species of Primula (primrose), Androsace (rock jasmine), Cyclamen (Fig. 46.1), Dodecatheon (shooting star), Douglasia, Lysimachia (Loosestrile) and Omphalogramma.

A Humble Beginning

Guava (Psidium guajava L.) is one of the most important tropical fruit crops, valued for its vitamin C content, nutraceutical compounds, and adaptability to diverse agro-climatic conditions. However, guava production faces problems of seedling variability, prolonged juvenile phase, susceptibility to wilt and anthracnose, and poor success rates of vegetative propagation (Singh et al., 2022).
Biotechnological tools, ranging from micropropagation to molecular diagnostics, provide solutions to these challenges. These methods complement conventional horticultural practices by ensuring clonal fidelity, enabling rapid multiplication of elite genotypes, characterizing genetic diversity, and offering prospects for disease resistance and quality improvement (Kumari et al., 2021; Mishra et al., 2020).
16.2 Micropropagation: Commercial-scale Clonal Propagation
Micropropagation is the most widely used biotechnology tool in guava. Nodal segments and shoot tips cultured on Murashige and Skoog (MS) medium with cytokinins such as benzylaminopurine (BAP) can produce multiple shoots, while rooting is induced using auxins like indole-3-butyric acid (IBA) (Singh et al., 2022).
Juvenile explants generally perform better, and the addition of antioxidants (ascorbic acid, polyvinylpyrrolidone) or activated charcoal reduces tissue browning. Plants require careful acclimatization under mist chambers before field planting. Micropropagation ensures large-scale, disease-free, uniform planting material for high-density orchards (Singh et al., 2022).

X-ray photons are produced as a result of electrons hitting metal while extremely rapidly moving at high speed. This is achieved through the application of an electric current to a cathode (negative electrode) via a high voltage power source (electricity) which enables electrons to be released from the cathode into the X-ray tube. These electrons, being negatively charged, are attracted to the anode (positive electrode or target). When the electrons strike the metallic target (anode) in the tube, X-rays and heat are produced. Basically the process of producing an X-ray involves a source that produces X-rays and radiating them through a body or object, where they are absorbed at different rates until they are detected by a sensitive film or cassette. It should be mentioned at this point that most of the energy of the electrons is not converted into X-rays but is dissipated as heat. In fact 99% of the energy dissipated in the target is lost as heat, and only 1% is converted to X-ray energy.

In the perspective of the accelerating national growth rate of 8% recorded at the end of the 10th Plan to a projected growth rate of 10% by the end of the 11th Plan, yielding an average GDP growth rate of about 9% it was appearing that the nation is going to have a strong footing of development. It can be made possible only through working with them by appreciating their struggle for survival using all round innovative measures coming out of their daily life experiences and traditional knowledge. Technopreneur Promotion Programme (TePP) is a unique scheme of Government of India for development of society especially by promoting the intellectual properties of grass- root level innovators coming out from the society with a source of inspiration arising out of their daily life struggle for existence.
