In order to provide for the establishment of an effective system for protection of plant varieties, the rights of farmers and plant breeders and to encourage the development of new varieties of plants it has been considered necessary to recognize and protect the rights of the farmers in respect of their contribution made at any time in conserving, improving and making available plant genetic resources for the development of the new plant varieties. Moreover to accelerate agricultural development, it is necessary to protect plants breeders’ rights to stimulate investment for research and development for the development of new plant varieties.

Principle

Tensile Test (ASTM D638)

Tensile strength is a measurement of ability of plastic material to withstand forces that tend to pull it apart. It is the amount of stress at the break point.

This test also helps to determine the extent the material stretches before breaking known as elongation at break. Percentage elongation is defined as the amount of linear deformation of the sample before breakage. It is measured as strain value at the break point.

Yield Point: It is the point at which an increase in strain occurs on stress-strain curve without the increase in stress. (point C)

Yield Strength: The stress at which a specified limiting deviation from the proportionality of stress-strain curve is observed for a material is known as Yeild strength.

Bhendi is known by many local names in different parts of the World. For example it is called Lady’s finger in England, Gumbo in USA, bhendi in India Okra (Abelmochus esculentus L. 2n= 1 30) belongs to the family Malvaceae. Origin of okra is West Africa. Okra plant is an erect, herbaceous annual, 1-2 meter tall and forms fairly heavy tap root.

One of the objectives of experiments in cultivators’ field is to test technology for adoption in a targeted area i.e. NARP Zone, or a district. In that case interaction between district x treatment within a zone or between blocks x treatment within a district should not be significant. If the targeted area is a district, an experiment in cultivators’ fields must occur in at least two blocks. Analysis of cultivators’ field experiment

The on-farm trials were conducted in 25 villages on kharif rice during 1992-93. The treatments viz. T1-Farmers’ practice with local variety, T2-Farmers’ practice with improved variety, T3-Farmers’ practice with improved variety +recommended fertilizer and T4-Improved practice with improved variety +recommended fertilizer. Farmers level of NPK fertilizer was 40:0:0 and local variety was Suzata while recommended NPK fertilizer was 80:40:20.

In India, the commercial cultivation of oil palm started during 1970. Today about 16 countries of tropics cultivate oil palm. Wilt appears in the fi eld as patches is both seedling stage and developed plants. Symptoms include drooping and wilting of uppermost leaves. Plants become completely yellow and die. Discoloration of vascular bundle are noticed when the stem is spitted

1. General Instructions

Instructions to work in the laboratory are given below. Students are advised to read them carefully before they begin their work in the laboratory.

• Apparatus to be used should be neat and clean.
• Graduated pipette should be used for pipetting.
• The small amount of liquid remaining in the pipette should not be blown out while standardizing the pipettes; allowance is always made for this. However, a frosted ring near the top of recently manufactured pipettes indicates that the last drop is to be blown out.

Plantation crops: Plantation crop are a group of commercial crops that are perennial in nature, cultivated extensively in tropical and subtropical situations that need employment of labour throughout the year and whose products are usually consumed after processing. ex. Principle

ASTM D638 covers the tensile properties of both reinforced and unreinforced plastics in the form of dumbbell-shaped molded plastic test specimens.

Yield strength is measured as the highest stress where a film, when deformed, will resume its original dimensions when the force is removed. The yield stress is expressed as force per unit area (engineering stress) and is calculated as per Equation (1) and the elongation at yield as per equation (2).

Introduction

The hot arid region of the country is spread over nearly 31.7 million hectare land area in different states. The major part of hot arid region of the country lies in western Rajasthan (19.62mha) followed by North-western Gujarat (2.16mha). The major natural constraints in arid vegetable production comprise of very low and variable rainfall (100-450mm), very high potential evapo-transpiration rate, intense solar radiation (320-619/cm²/day), high wind velocity (20km/hr), high infiltration (9cm/hr), extremes of temperature (0 to 48^ºC), low relative humidity, high soil salinity, very deep ground water, etc. These bio-physical constraints reduce the productivity of cucurbits many fold. Despite  several constraints, the hot arid areas of western Rajasthan offers very good opportunity for cultivation of cucurbits like, watermelon, muskmelon, longmelon, bottle gourd, ridge gourd, tinda, summer squash, cucumber, etc.

There are a number of different cell lines derived from human and animals. With the dramatic increase in numbers of cell lines, the risk of intraspecies and interspecies cross contamination rises proportionally. It is particularly a major problem in laboratories where many different cell lines of human and animal origin are handled at a time. The purity of the primary cell or continuous cell lines and species of its origin should be checked on a regular basis to authenticate the cell line identity. In the absence of such monitoring, inter and intra species cell line contamination are likely to occur in the laboratory resulting in loss of investigator’s time effort and resources. The advantages of working with a well defined cell line free from contaminating organisms would appear obvious.

Abstarct

The Photosynthetically Active Radiation (PAR) measure of radiant power is important in evaluating the effect of light on plant growth. The photosynthetic response correlates better with the number of photons than with energy. This is expected because photosynthesis is a photochemical conversion where each molecule is activated by the absorption of one photon in the primary photochemical process. PAR is defined in terms of photon (quantum) flux, specifically, the number of moles of photons in the radiant energy between 400 nm and 700nm. One mole of photons is 6.0222 x 1023 photons ( 6.0222 x 1023 is Avagadro’s Number). The Phtotssynthetic Photon Flux Density (PPFD), i.e. the photon irridiance, is expressed in moles per squre meter and per second (formerly, Einsteins per squre meter and per second). The absorption of PAR by wheat varieties increased from 7.30 h to 11.30 h followed by a decline at 13.30 h at CRI, tillering and milking stages.

Objective: To estimate total protein in the plant sample by Folin-Lowry's method.

Background information:

Protein assay can be done to determine the level of protein in solution by the Folin-Lowry biochemical method. The concentration of protein leads to change in color of the solution and based on different color in different protein concentration solution the change in color may be digitized by colorimeter or spectrophotometer. This technique was developed by Oliver H. Lowry in the year 1940s. The tyrosine and trytophan amino acid residues in a protein sample produces purple or blue color in the absorption maxima at 660 nm by using Folin-Ciocalteau (having phosphate and sodium tungstate molybdate) chemical reagent. The color intensity of protein sample depends on the concentration of above aromatic amino acids (tyrosine and tryptophan), hence every sample will vary for color. For protein estimation the BSA (Bovine serum albumin) is widely used as standard protein molecule due to its cost effectiveness, purity and easy availability. This method of protein estimation is very sensitive and can detect up to 10 μg/ml. This is a relative method of protein estimation and may be interfere by salts, Tris buffer, nonionic & cationic detergents, EDTA, lipids and carbohydrates. For reproducible assay the incubation time, pH (9-10.5) is critical in this method (Lowry, et al. 1951).
