Abstract

Re-constituted rice-premixes made by extrusion, has been successfully formulated for the fortification of market rice with vitamin A (retinyl palmitate and betacarotene) and iron. The statistics of vitamin A deficiency (VAD) and Iron deficiency anaemia (IDA) are alarming in the developing world, particularly in India. In fact, India is second largest producer as well as consumer of rice in the world, so fortification of rice with vitamin A and iron seems to be a successful solution to reduce the VAD and IDA in India. In order to design the final formulation, eight formulations containing target micronutrient profile were tested for vitamin A (both retinyl palmitate and beta-carotene) and iron retention for a period of six months under ambient (24-26 ⁰C) and elevated temperature (40 ⁰C) conditions. Nonsignificant iron loss was observed during six months of storage time under both ambient and elevated conditions. The higher retention of retinyl palmitate and beta-carotene was observed in formulation 5 and 8 respectively. In F5, the retention of retinyl palmitate was 73% and 66% and in F8, retention of beta carotene was 70% and 63% under ambient and elevated temperature conditions respectively after six months of storage. Substituting the original, vegetable shortening with palm shortening resulted in increased retention of retinyl palmitate and beta carotene in formulation 5 and 8. Thus, increased antioxidant effect of natural antioxidants in palm shortenings contributed to stability of micronutrients.

Abstract

This review assesses the nutritional attributes of Amaranth grains and its utilization as food and as formulated foods. Amaranth is a pseudo-cereal and one of the most ancient cultural food plants with excellent source of superior quality protein (rich source of lysine) which is deficient in wheat and other cereals. Amino acid composition of amaranth protein is close to ideal and, as a result, has high food value. Amaranth grain is loaded with phytochemicals including phenolic acids, tannins, anthocyanins, and phytosterols. Amaranth is potentially a good dietary source of the pro-vitamin A carotenoid (f-carotene), antioxidants (nutraceuticals) and canthaxanthin (which has also been reported to be an antitumor agent) and lutein, which is reported to slow down the development of age-related eye diseases. There are also some anti-nutritional factors (Phytic acid, Tannin and Trypsin inhibitor etc.) that may be reduced by certain processing treatments. Several epidemiological studies show Amaranth grains are helpful in reducing several kinds of chronic diseases like cancers, cardiovascular diseases, type II diabetes and various gastrointestinal disorders. Grain amaranth has been tested and recognised by many authorities as a gluten-free foodstuff appropriate for incorporation into the diet for celiac disease patients. Amaranth grain or young leaves could completely or partially substitute common ingredients, so the crop could be potentially exploited as an ingredient in foodformulation in form of flour, flakes, sprouts, grain undergoing fermentation, popping, malting, extrusion cooking, nixtimisation, and special compounds isolation. The interest in this crop is developing due to its several advantageous properties. The nutritionists find that the grain has higher nutritional value than conventional cereals.

Abstract

Sedentary lifestyle coupled with excess calorie intake is partially responsible for increased incidence of lifestyle related diseases like coronary heart disease, obesity, diabetes, cancer and hypertension. Non-nutritive sweeteners can offer consumers a way to enjoy the sweet taste with little or no energy. Further, functional beverages are the drinks which are altered in such a way to provide specific health benefits and disease preventing properties beyond general nutrition. In the present paper, authors have summarized various research works on development of different low calorie functional fruit and vegetable products by replacing sugar with non-nutritive sweeteners like saccharin, cyclamate, sorbitol and stevioside at different levels of sweetness (equivalent used to sucrose). The cyclamate sweetened spiced plum squash at 50 per cent sweetness proportion gave higher taste scores in comparison to saccharin sweetened beverage. In case of sorbitol sweetened beverages, with the increase in the proportion of sorbitol sweetness, the organoleptic score for colour, body and flavour increased compared to saccharin and cyclamate sweetened products. Whereas, the stevia sweetened beverages indicated highest sensory acceptability at 75% stevioside sweetness level. Further, sorbitol sweetened health products were found to have similar calculated energy value as that of sucrose sweetened products (100%), but sorbitol is absorbed slowly and does not raise the insulin levels. The development efforts had successfully reduced calorie value of plum squash up to 42% with cyclamate at 50% sweetness level. The calorie value of the sorbitol and stevioside sweetened products has been reduced by 70% and 60% per serving at 100% and 75% sweetness level, respectively. The developed products were found to have strong antimicrobial activity against E. coli as well as high antioxidant potential. Overall, it was concluded that, the developed products had better taste, palatability and storage stability beside reduced energy value, hence their availability in the market will definitely benefit the at risk group i.e. diabetics, obese and health conscious people.

Abstract

Noodles are widely consumed throughout the world and their global consumption is second only to bread. In the study, response of reducing agents (cysteine hydrochloride and sodium metabisulphite) and oxidizing agents (potassium bromate and benzoyl peroxide) at 10 and 20 ppm levels on the farinographic and amylographic curve characteristics as well as on the noodle making properties of durum and commercial wheat semolina were investigated according to the AACC (1990) procedure. The farinographic curve characteristics revealed that water absorption was enhanced with increasing levels of reducing agents but decreased with addition of oxidizing agents. However there was no significant change in Dough Development Time in case of durum semolina on addition of reducing agents, but commercial semolina recorded significant changes at 20 ppm level of reducing agent. The best reducing agent for durum and commercial semolina was thus sodium metabisulphite and cysteine hydrochloride respectively. Addition of oxidizing agent potassium bromate at 20 ppm level enhanced stability and was thus chosen as the best oxidizing agent for durum semolina and benzoyl peroxide for commercial semolina. Addition of reducing and oxidizing agents improved yield, appearance, elasticity and extensibility characteristics of instant fresh noodles. The best levels were used in the preparation of noodles on the Italian extruder (NAMAD).

Abstract

In the present study, a novel work on comparison of physico-chemical characterization and bacterial diversity of wheat straw compost and vermicomposting has been initiated in the tropical region. Wheat straw compost shows lesser pH, EC, TDS, Organic Carbon, Total Nitrogen and Total Phosphorus than vermicompost whereas there was an increase in Total Nitrogen, Total Phosphorus and Electrical Conductivity and a drop in the pH, TDS and Organic Carbon of the vermicompost as the composting progressed from 0, 3, 10, 17, 24 and 31 days. Strains of Bacillus spp. and Thermoactinomycetes were screened from the wheat straw compost, which are ecologically important and have commercial potential for enzyme production. These strains were able to degrade cellulose, hemicellulose and lignin and exhibited cellulase activity. Enterobacter aerogenes, Micrococcus luteus and E.coli were isolated from the gut of Eudrilis eugeniae which help in the transformation of biodegradable waste materials into valuable compost.

ABSTRACT

Remote sensing has undergone tremendous development in past decades. Hyperspectral imaging refers to the imaging of a scene over a large number of discrete, narrow contiguous spectral bands in the visible through thermal infrared portion of the spectrum. Thus it is possible to construct a complete reflectance spectrum of every pixel in the scene.

Classification of hyperspectral image requires thorough understanding of surface material properties, sensor characteristics and atmospheric absorption spectra. The reflectance spectra of most materials on the Earth's surface contain diagnostic absorption features. Remote sensors, capable of acquiring complete reflectance spectra over large areas, offer a powerful tool to study of the Earth and the environment. Methods are being developed to extract information from remote sensing data using image-processing approaches. Specific absorption features of reflectance spectrum can be used to identify a number of important rock forming minerals, and have been used by geologists for geologic mapping and studies of volcanoes.

The present discussion includes an application of thermal characteristics of near- IR bands of Landsat TM to estimate the temperature of over and around the crater of Barren Island volcano in 1991. A method using dual band temperature characteristics related to radiant source of sub-pixel size was developed and utilized to prepare temperature contour map of the Island using unsaturated pixels of band 5 and 7.

ABSTRACT

A study was conducted to compare inter-sensor variability by computing vegetation indices, NDVI & SR. The two sensors compared were Hyperion and ALI (Advanced Land Imager) present on EO-1 satellite. Hyperion is a hyperspectral sensor having a spectral range of 400-2500nm with 10nm bandwidth. ALI is a multispectral sensor with 10 bands of 100nm bandwidth. Hyperion and ALI data were acquired at the same time for two different seasons, dry (April) and wet (October) seasons over Shoolpaneshwar Wildlife Sanctuary, Gujarat, India. Vegetation indices such as SR “ Simple Ratio and NDVI -Normalized Differential Vegetation Index were performed. Seasonal variations were observed in both the data sets. For SR index, Hyperion performed well using October month data. In comparison with ALI, Hyperion showed high NDVI for both deciduous and evergreen species using October month data. The highest NDVI value is assumed to represent the maximum vegetation “greenness” during that period. Results clearly indicated the superiority of Hyperion in vegetation analysis using NDVI & SR indices.

ABSTRACT

The Mumbai region of Maharashtra has been witnessing very rapid urbanization development in terms of industrial, commercial and residential establishments. This has resulted in reclamation of tidal flats and estuarine regions. The ‘marine clay', characteristic of this environment has high swelling potential and consequently affects the performance of the structures. This will have a serious negative impact on the urbanization expansion. In this study, an attempt is made to map the spatial distribution of these expansive clays using the state of art Hyperion data. Reflectance spectra of representative samples collected from the field is used to map marine clays using Spectral Angle Mapper (SAM) technique. Laboratory analyses of these foundation soils (reflectance spectroscopy, mineralogy and Atterberg limits) indicate that they have significant amount of montmorrilonite and has good swelling potential (A_C>1.4). The derived expansive clay abundance map will be of use while planning appropriate remedial measure and future development.

ABSTRACT

In the present paper, study of spectral reflectance measurements has been carried out for different type of snow and other ambient objects in snowbound areas of Indian Himalaya using Spectroradiometer (350-2500nm). The observations suggest that there are significant changes in the spectral radiances and reflectance values of snow and other ambient objects, which can be discriminated using satellite multi-spectral bands, however for characterizing the snow using satellite sensor, spectral bands having maximum radiances values should be greater than snow radiances and suitable spectral bandwidth are to be identified. The detail analysis of field collected reflectance suggest that, due to aging and grain size variation in snow, the maximum variations in reflectance was observed in the NIR region i.e. around 1040-1050nm. For contamination and snow depth, the maximum variation was observed in the visible region i.e. around 470nm and 590nm respectively. For the moisture changes, the maximum variations observed around 980 and 1160nm. The study also suggests, normalized difference snow index (NDSI) increases with the increase of aging, grain size and moisture content. This study can be helpful in selection of suitable spectral bands for snow cover applications using satellite

remote sensing and developing the algorithms for seasonal snow cover characteristics using multi-spectral and hyper-spectral data.

ABSTRACT

Hyperspectral data are generally noisier compared to broadband multispectral data. Therefore, it is desirable to smoothen the reflectance spectra. Smoothing needs to preserve an absorption feature while simultaneously removing noise. It is very essential to optimize the smoothing filter to minimize disturbances to the statistical properties of the original data. This study was carried out to see the effect of Fast-Fourier Transform (FFT) based smoothing technique on the statistical properties of vegetation spectral response and to see the effect of the filter size. The data used for the present study is the reflectance spectra data obtained from space borne EO-1 onboard Hyperion sensor. Filter sizes were varied between 3 and 15 with the increment of 2, i.e. 3, 5, 7..........15. Paired t-test was carried out between the original and the smoothed data (for the selective bands) for all the filter sizes in order to see the extent of distortion with changing filter sizes. Our study revealed that the distortion in the spectra increases with the increase in the filter size. Beyond filter size 11, the number of locations within the spectra where the smooth spectra were statistically different from its original counterpart increased to 14 and reaches 21 at the filter size 15.

ABSTRACT

In earth system about 60% of the world's population live within 60 km of coastline and use the coastline for their livelihood. Environmental pollution of the coast, inshore water and deep ocean is one of the important topical issues in the context of human health and global warming. To combat this natural and man made problem, we have to rely on qualitative analysis quantitatively. In the recent advancement in information and technology field, spectral data plays a major dominant role in the environmental studies. This could provide more powerful and accessible means to exploit the enormous wealth of data available in the system. The high-resolution imaging spectrometry (hyperspectral imagery) will help to analyze the data for mapping and other environmental modeling applications. This could attain advanced approaches to deduce surface characteristics and extract features from hyperspectral images. The current capabilities are in which, the prototype system can classify pixels according to surface type. The classification process employs neural network analysis of inputs which include pixel spectra and a variety of processed image metrics, including image “texture spectra” derived from fractal signatures computed for sub image tiles at each wavelength. Thus, this kind of technical analysis can prove to achieve coastal area studies well in advance in the ongoing environmental changes more precisely.

ABSTRACT

A satellite based earth observation system enhances our understanding of the entire earth as an integrated system. Remotely sensed data gives accuracy and complete data. To study the coastal zone or area remote sensing is on of the best technology to monitor the regular changes over the time. Here satellite data used to understand the distribution of prawn farms along the Nagappattinam coast. It has stretched from River Coleroon in the north to Point Calimer in south. Nagappattinam coast rocked by tsunami in 2004 and it damaged the whole life of coastal and fishing people. This paper has attempted to analyse the distribution of prawn farms and impact of tsunami in the Nagappattinam coast using remote sensing.

1. INTRODUCTION

Nagappattinam coast is located on the eastern part of Nagappattinam. It is a unique immemorial for its agricultural activities and lying in the deltaic region of Cauvery River. Nagappattinam coast (Fig 1) is peninsular delta area bounded by Bay of Bengal on the east, Palk Strait on the south, and Thiruvarur and Thanjavur districts on the west and Cuddalore on the north. The study area is located between 10^o 10’ N to 11^o 50’ N latitudes and 79^o 45’ E to 80^o 00’ E longitudes. Nagappattinam coast has covered an area of 935 sq. km.

Coleus (Solenostemon spp), commonly known ‘Painted Nettles’ / ‘Flame Nettles’ is a genus native to tropical Africa and Asia. They are best known for their stunning foliage in various colours, patterns and texture. Flowers though available in different colours, are unable to create substantial effect being inconspicuous. The group includes herbaceous, semi-herbaceous annuals and perennials. They are grown in beds, pots, hanging baskets and many other ways as per season and climate. In tropical and sub-tropical climate, Coleus is grown during mild period (October to March). However, there are perennial species also which are grown round the year.

The ornamental effects exhibited by Coleus particularly in tropical climate are no way less than other flowering plants grown during that season. The popularity of coleus, therefore, is huge and exclusively used in Indian gardens.

Gardens around the religious places / structures such as temple, mosque, shrine etc. have been developed since very old times. In fact gardening practice actually started with the temple gardens for growing flowers for worship purpose. The main purpose of these gardens is to create scenic beauty and a pleasant atmosphere where one can relax, mediate and spent some time in tranquility besides making prayer and worship.

Design

The garden around such structures depends on its type, requirement and number of visitors of that place. In case, large number of visitors visiting the place daily, garden areas need to be fenced with grill or hedge so that no damage is done to the garden, particularly walking through lawn, flower beds, plucking of flowers, leaves etc. Architectural beauty and position of the structure (temple, mosque, shrine), availability of space (area and allocation - front, side, back) should be taken into account while preparing design. In case of big areas with boundary, trees may be planted along the boundary wall. Remaining area is to be divided into blocks for accommodating other features like shrubbery border, flower beds, water bodies, fountain etc. as per availability of space and suitability. Gradation for making change of elevation by way of earth work or construction of retaining walls must be well planned and completed at the initial stage of garden construction. Other facilities for the public like seats, shelters, public conveniences are also to be taken into account.

Most of the DNA of an living organism is packaged and organized in a chromosome as well as in mitochondria and cell organelles (Feulgen and Rossenbeck 1924). DNA is not found on its own but rather is structured in long strands wrapped around protein complex called nucleosome consisted of a protein called histone. During most of the duration of cell cycle a chromosome consists of long double helix DNA molecule (with associated proteins)During S phase the chromosome gets replicated resulting in X –shaped structure (chromatids) and still attached to a single centromere. The basic role of the chromosome is to provide a frame work which allows each linear segment of the genome to replicate and segregate efficiently. Failure in either of processes cause chromosome imbalance in daughter cells. Three specific cis-acting sites are required for stable chromosome maintenance, i.e., an origin of replication, a centromere and telomeres.

Classical studies of the morphologyof individual chromosomesof the plant species begun by S. Nawaschin 1910-16 and continued by Taylor (1925), Sharp (1929) and Mc Clintock (1929-30). And eventually it was possible to set up working models which account for the internal structure and chemistry of the chromosome and the genetic material as well as genetical facts. The use of aceto-carmine with iron (Belling, 1921) and application of heat greatly improved the staining differentiation of chromosomes. Smears have practically replaced the paraffin techniques of studying chromosome. The gross morphology of chromosomes is generally worked out either at prophase stages of meiosis or metaphase in somatic tissues. The chromosomes may differ in thickness, total length, position of centromere or spindle fiber attachment region, chromomere pattern and number and position of secondary constriction. The total length of chromosomes at metaphase also differs in different tissues of a species and also in certain tissues chromosome present may be very different in appearance. Figure 1 shows various morphological features of a eukaryotic chromosome

Thermal Analysis

Physical and chemical properties of any soil are governed largely by the mineralogical make up of the soil, and more specifically by the clay fraction. Identification, characterization and an understanding of properties of different minerals are important in evaluating soils in relation to classification, agronomic practices and engineering purposes.

Clay fractions of soils are usually composed of mixture of one or more secondary phyllosilicate minerals along with primary minerals inherited from the parent materials. Identification of minerals species as well as their quantitative estimation in such polycomponent system require application of several qualitative and quantitative analysis.

The methods used for identification and quantification have steadily been improved over the last 50 years. Among the several methods often used for this purpose three are very common, e.g. (i) differential thermal analysis (DTA), (ii) x-ray diffraction analysis (XRD), and (iii) infrared spectroscopy (IR).

Thermal analysis is a term covering a group of analyses that determine the changes of some physical parameters, such as weight, energy, dimension and evolved volatile substances with change in temperature. Many soil constituents undergo various thermal reactions upon heating, which can be utilized as a diagnostic tool for qualitative identification and quantitative estimation of the substances. These reactions may be exothermic or endothermic, and phase change or crystal change may take place during reactions. Whatever be the reactions, if properly measured, they give some valuable information to identify the substance under consideration.

Among the several methods available for thermal analysis, three methods are frequently used in soil research: differential thermal analysis (DTA), differential scan calorimetry (DSC), and thermogravimetry (TG). Among these, DTA and TG will be discussed here.

Sulphur (S) occurs in soil in both organic and inorganic forms. Inorganic S fraction may occurs as sulphate (SO₄^2-) and compounds of lower oxidation state such as sulphide (S^2-), polysulphide (S_n^2-), thiosulphate (S₂O₃^2-) and elemental sulphur (S⁰). In well drained soils inorganic S usually present as sulphate. Under anaerobic condition (reduced or waterlogged soil) inorganic S mainly occur as sulphide and under extreme condition as elemental S form. Estimation of S is carried out in two steps:

1. Extraction of different forms of S from soil by suitable extractant.
2. The filtrate is then analysed for S by the turbidimetric method.

The turbidimetric method is the most widely used one because of its simplicity and rapidity.

Agroforestry is a new name for a set of old practices. The farmers have been practicing agroforestry since ancient times. The general concept of agroforestry is to integrate trees and agriculture so as to create a more diversified landscape, while providing the producers with new environmental and economic benefits. In other words, agroforestry is a method of farming that allows trees and shrubs to grow along with crops and/or livestock, therefore blending agriculture and forestry in the same production system. In fact, man’s association with forest is much older than with agriculture. First man was a food gatherer and hunter in forests. Then he realized that the seeds of the fruits he collected germinated, grew into plants and bore the fruits again and thus man started to cultivate foods. Man’s desire to live in a community created settled agriculture.

Since then, the pressure on the agricultural lands has increased manifolds due to the increasing population, expansion of urban area and the industrialization process. The environment has also been disturbed. Soil is losing its productivity and the biodiversity is threatened. Farming community is trying all means to increase the land productivity. Chemical fertilizers and pesticides are applied in higher proportion, causing environmental pollution hazards. Under all these circumstances agroforestry has shown that besides sustainable agriculture it can also help promote a better environment. This relatively young science known as agroforestry was brought from the realm of indigenous knowledge into the forefront of agricultural research four decades ago and was promoted widely as a sustainability-enhancing practice that combines the best attributes of forestry and agriculture.

Farmers have been growing trees for different purposes for thousands of years. Tree species that are grown to provide more than one significant function are called multipurpose trees. These functions may be productive such as producing fuelwood, timber, fibre, fodder, food, medicine, etc. and/ or protective such as soil conservation, shade, shelterbelt, microclimate amelioration, land sustainability, biodiversity preservation, etc. All trees are multipurpose; some, however, are more multipurpose than others. Tree species can be multipurpose in two ways.

1. A single tree can provide more than one function. For example, Gliricidia sepium is grown as living fences that provide fuel, fodder and green manure for agricultural crops - all at the same time.
2. Trees of the same species, when managed differently, can provide different functions. For example, Leucaena leucocephala is managed so that some trees will mainly yield wood while others mainly produce leaf fodder.

Farmers can grow multipurpose trees (MPT) in various combinations with other crops, as in agroforestry, in block plantations of trees or in naturally regenerating tree farms. In certain cases, multipurpose trees are grown and managed for only one purpose. For example, Gliricida sepium is grown only to provide shade in coffee plantations. The same species may be planted in some other places and is managed differently for a very different use.

Introduction

Agriculture occupies a prominent place in Indian economy and rural livelihoods. But, it is faced with the challenge of producing food for a rapidly growing population whilst maintaining the nation’s fragile resources.Efforts through green revolution brought out significant increase in food grain production by concentrating on the potential farms in terms of irrigation and other basic resources.Nationally and globally, it is necessary that the growth rate of food production must remain higher than the population growth rate to avoid hunger and overcome famines. But, food security one of the basic human rights is in jeopardy, today. Agriculture is crucial for ensuring the food and livelihood security of the country and hence it is important that this sector with diversified farming systems becomes resilient to increasing climatic variabilities and global economic changes. This concept encourages the farmers towards efficient utilization of natural resources and also proper channelization of by-products of one enterprise to the other enterprise in farming. The integrated farming systems, therefore, assume prime place for sound management of farm resources to enhance the farm productivity, reduce the environmental degradation, and improve the quality of life of resource poor farmers and to maintain the sustainability to meet the challenges of food security. In order to meet the soaring demand for food there is a need to go for integrated faming system.This approach facilitates in strengthening the socio-economic status of Indian farmers besides strengthening ecological balance. Integrated farming system is common to Indian farmers simply because of the preponderance of small farm systems which form the backbone of traditional agriculture. For small farmers, these systems enable a means of diversifying the use of meagre resources in the context of a rational means of reducing risk. Additionally, it also enables increased efficiency in the use of these same resources in a manner that there will be livelihood security and stability to farming systems. There is no scope for increasing the farm size. Yet steady increase in population with shrinkage of cultivated land as a result of industrialization and urbanization would pose serious hurdle for agricultural development. Only vertical expansion is possible by integrating appropriate farming components requiring lesser space and time ensuring continuous income to the farmers.

Fruit production is gaining momentum in the country. With the production about 90.18 million tonnes of fruits from 6.30 million ha in 2015-16, India is the second largest producer of the fruits in the world after China. During the past three decades, international trade in fruits has seen a rapid expansion.The newer markets are now opening for Indian produce e.g. Japan and USA have opened the route for Indian mangoes. At domestic level, the increase in demand of fruits for consumption both in fresh and the processed form has been evident. The income and livelihood security from this sector are increasing. The government has also supported the horticulture sector particularly area expansion and productivity enhancement of fruits through technological interventions like drip irrigation, fertigation, emphasis on integrated nutrient management, production of high quality planting material etc. Selection of an ideal variety and availability of plant material from a reliable source is the basis for achieving high yield and quality uniform produce. Lack of clonal uniformity not only leads to the problems in crop management but also the differences in maturity may cause delay in commercial picking. A number of improved fruit crops’ varieties have been developed during past few years, which ranged from the high yielding to stress resistant to high quality produce. In fruits and plantation crops more than 150 high yielding varieties suitable for various purposes have been developed. A list of some popular cultivars and new cultivars in major fruit crops has been given here (Table 1).

Indian horticulture has made a rapid stride, converting the country from food scarce to self sufficiency. The horticulture, which includes fruits, vegetables, spices, flowers and medicinal and aromatic plants, has proved beyond doubt its potentiality for gainful diversification. One of the significant developments is that horticulture has moved from rural confine to commercial production, and this changing scenario has encouraged private sector investment in production system management. Since the growing of fruit crops is highly rewarding to the farmers in terms of returns per unit area, the horticulture sector is expected to contribute significantly for the food and nutritional security, employment opportunity and poverty alleviation.

In recent past, several innovations and new initiatives have been made both in research and development for improving production, productivity, quality and marketing and to identify critical gaps in various emerging areas in fruits & horticulture technology. Evolution of the Internet

The Internet, as we know it today had a very humble beginning. J C R Licklider of the Massachusetts Institute of Technology envisioned the Internet as far back as 1962 in a series of memos written by him that talked about social interactions that could be enabled through networking, a concept that he termed his “Galactic Network”. He joined DARPA (Defense Advanced Research Projects Agency) in October 1962 and was its first research head. There, in collaboration with Lawrence G Roberts and other scientists in 1965, the ARPANET (Advanced Research Projects Agency Network) was conceived. With other developments, ARPANET was transformed into the Internet as we know it today. The technology involved in this was a very basic packet switching network that later moved to include packet satellite networks, ground-based packet radio networks and other networks. In packet switching mode, the hub router, which acts as the intermediary, decides the best path for sending or receiving data packets, unlike circuit switching mode (used in telephony) in which there is a dedicated path by which the data travels from the source to the destination.

The future of networking was based on this skeleton. Further down the line, the TCP/IP (Transmission Control Protocol/Internet Protocol) was developed, which had the ability to address networks and machines further downstream. Other technologies like telnet, e-mail, internet telephony etc, were also developed subsequently. By 1990, the ARPANET was decommissioned and TCP/IP had displaced most other Wide Area Networking (WAN) protocols and was fast becoming the accepted protocol for internetworking. After a while, the World Wide Web (WWW) came into existence. A consortium called the World Wide Web Consortium (W3C) was formed in 1992 and it had taken on the responsibility for evolving the various protocols and standards associated with the web. Soon after this, commercialization of the internet started taking place. There was a marked increase in the number of communities across the internet. Bulletin Board Services and Usenet groups were now home to more computer users than ever before. With the advent of such information, corporations and businesses also started looking at the internet in terms of business viability. In 1994, Pizza Hut started offering pizza ordering on its Web page while First Virtual, the first ‘cyberbank’, launched its online presence. By this time, there were more than 38,64,000 hosts on the Internet. Since then, the Net has grown rapidly and the process continues to this day.

Though the Internet hit India in the late 1980s, it was deemed fit only for researchers and scientists. Commercial internet access was first introduced by VSNL in August 1995. Today internet in India has ballooned and India has become a giant in the IT Enabled Services (ITES) field. With falling internet access costs and increasing users, India’s ranking in terms of internet users is rising fast.

The Internet is growing at an amazing rate. The main reason for this is that the Internet has now become an accepted part of the mainstream urban life. It is used for almost everything imaginable, from recreation and entertainment to learning about rocket science. However, not everything about is good. There is an information overload and not all the data that is available is true and should not be accepted at face value. The internet is not limited to the PC anymore and has percolated down to various devices thanks to advancement in technologies like GPRS, EDGE, CDMA, WiFi etc. through which it can be accessed with a mobile device.

Dirying in India has traditionally been the preserve of landless agricultural labourers and small and marginal farmers, who have maintained very small herd sizes. However, of late, with increasing consumer awareness regarding milk quality, there has been a marked increase in the number of large, well-organized dairy farms which cater to predominantly urban markets. Terms like clean milk production, organic milk, microbial load of milk etc. are being used more frequently while referring to milk quality. Larger, intensive dairy farms not only profit from the economies of scale by way of optimum utilization of land, labour and resources, but also benefit from the fact that it is easier and cost-effective to implement more efficient technologies like machine milking, manufacture of by products, chilling, packaging, bulk transport etc. Further, most of these farms are able to realize a premium price for their products on account of high quality and brand identity, thus leading to greater profitability.

1. INTRODUCTION

The numbers of plant products that influence the nervous system are many and particular group that act on central nervous system by influencing neurotransmitters or endorphins are treated as psychoactive plants. Psychoactive drugs that act on the central nervous system are very much involved in psychiatry. The range of effect is wide for these plants and they can be divided into three categories viz.,

• Stimulants, which excite and enhance psycho motor activity.
• Depressants which reduce mental and physical performance and
• Hallucinogens which are capable of inducing dream like state.

A wide range of plant derivatives are involved in these mind altering drugs.

Water

• Water constitutes about 35–95% of the different portion of the plants. The leaves of the plant contain 35–95% water, Roots 60–90% and Fleshy fruit contain 70–90% water.
• Water is one of the most important inputs essential for the production of crops.
• Plants need it continuously during their life and in huge quantities.
• It profoundly influences photosynthesis, respiration, absorption, translocation and utilization of mineral nutrients etc.
• Soil needs the application of water to:
• Remove stress condition.
• Release nutrients in the soil solution for absorption by plants.
• Preparation of land for raising crops.
• To maintain the temperature and humidity of the soil micro–climate and the activity of soil microbes at optimum level.

Introduction

Farming system conceptually is an integrated set of activities that farmers perform in their farm with available resources and circumstances to maximise productivity and net farm income on a sustainable basis. The system approach suggest a judicious mix of enterprises and activities based on the cardinal principle of minimising the competition and maximising complimentarity taking into account the components of soil, water, crops, livestock, labour, capital, energy and other resources. With all the resources, enterprises and external forces interacting in the system, the farmer operate at the centre of the farm household to take decisions observing variability, profitability, stability and sustainability. It is not the technology alone, but a set of social, political, institutional and economical forces influence the decision making process in a farming system leading to a model or acceptable combination of enterprises for overall development of the farm household.

Though science and technology have played a vital role in finding out innovations in the farming system approach through addition of new enterprise leading to profitability; new technologies ensuring stability and sustainability, the basic philosophy of the approach based on set principles is being pursued since initiation of agriculture in the civilisation.

In general farming system approach is more of a thought, a movement, a philosophy than simply choosing enterprise or combination of so to improve upon the standard of living. The interaction of bio-physical and socio-economic values plays an important role in up-scaling farming system approach and the adoption behaviour largely depend on the human values rather than material gain.

With the changing scenario, several factors like increasing instability in production, market price, high cost of production inputs, labour and negative impact of climate change, degradation of natural resources directly or indirectly warrants the use of sustainable rather regenerating technologies, compatible and noncompetitive farm enterprises to bring in sustainability and enhancement in production and income. For more than a century, programmes and activities have been designed to address the vulnerability of rural poor and are often targeted towards improving agricultural practices as means of increasing productivity, efficiency and ultimately income. In majority of the TOT cases technology has taken the centre stage even though the basic fundamental question like ‘whether the farmer adopt the offered technology and, if they do, whether they do it in ideal combination and for a considerable length of time needed to produce designed result remains unanswered. More importantly, in general the technology in isolation can not be recommended to all categories of farmers operating under varying situations. On the other hand acceptance and continuity of the technology is a dynamic process and varies both with the quality of the technology being promoted and the adoption behaviour of the individual farmer.

Ruminants have evolved to consume and subsist on roughage - grasses and shrubs built predominantly of cellulose.The rumen is a fermentation vat par excellance, providing an anaerobic environment, constant temperature and pH, and good mixing. Well-masticated substrates are delivered through the esophagus on a regular schedule, and fermentation products are either absorbed in the rumen itself or flow out for further digestion and absorption downstream. Rumen gases, particularly methane, are increasingly in the news because of their contribution to greenhouse gas and climate change.

The ruminant stomach is composed of four parts; the rumen, reticulum, omasum and abomasum. These compartments gives the cow and other ruminants a decided advantage in digesting and utilizing those parts of the plants and other compounds which are practically useless to animals with simple stomach. Substances such as cellulose, a major constituent of plant tissues and urea, a non protein nitrogen compound, are of limited use of non ruminants. However ruminants through fermenting and synthesizing action of microorganisms in their complex stomach, can efficiently utilize these substances for productive purposes. The dairy cow converts this feed into milk and meat, two highly nutritious and palatable food for humans. This ability to make use of feed which otherwise would go waste because it is not directly consumable for humans is one of the most compelling justifications for the existence of cattle and buffaloes, in a world which must make use of all possible food resources.

Reproduction in the males culminates in the production of semen containing fertile spermatozoa and deposition of these in the female reproductive tract at an opportune time.

Male Reproductive Organs

The reproductive organs in male domestic animals comprise the testes, epididymides, and accessory glands. The testes produce the spermatozoa and the male sex hormones (androgens). The spermatozoa pass from the testis into the epididymis where they acquire motility and attributes essential for fertilization and are then stored there. At ejaculation they are squeezed along the ductus deferens into the urethra and mixed with the secretions of the accessory glands to constitute the ejaculated semen.

Testis

The testis are composed of long tubes called seminiferous tubules and interstitial tissue. The spermatozoa are produced within the seminiferous tubules. This process is known as spermatogenesis. The interstitial tissue contains Leydig cells which are responsible for the production of the male hormone called androgens. The androgens are produced under the influence of hormones from the anterior pituitary namely ICSH (LH) and FSH. The principal androgens produced by the testis are testosterone and dihydrotestosterone.

The testis are suspended from the body in the loose sacs called scrotum, present in the inguinal region. The scrotum in addition to its container function, also acts as a regulator of temperature required for spermatogenesis.

Bone

Bones provide the rigid framework of the body of animals and man. The skeletal system serves three main functions. (1) It supports and protects the soft tissues and organs of the body. (2) It helps to maintain, through homeostatic regulation, the constancy of the ionic environment within the organism. (3) It helps in locomotion with the help of muscles and tendons.

The ends of long bone is called the epiphysis: the long, compact shaft, the diaphysis where it joins the epiphysis, the metaphysis. The diaphysis is composed of a hollow cylinder which contained the medullary cavity and bone marrow. During postnatal life, erythropoiesis occurs mainly in the bone marrow. In the adult, bone contains on wet weight basis approximately 25% water, 45 Percent ash and 30 percent organic matter. With increasing age, the ash content progressively increases as the water content decreases; the organic fraction remains relatively constant. Calcium constitutes about 37% of the ash content, and phosphorus about 18.5%.

Domestic animals including chicken and mammals, belong to the category of homeotherms, as opposed to poikilotherms. Homeotherms or warm blooded animals are those which are able to maintain constant body temperature in the face of varying environmental temperature. The poikilotherms or the cold blooded animals change their body temperature according to the temperature of the surroundings. Most invertebrates and lower vertebrates belong to the category of poikilotherms.

Why do animals have to maintain constant body temperature? As we are aware that enzymatic reactions in the body are temperature sensitive. In fact, a 10 degree centigrade increase in temperature can result in doubling of the reaction rates. In order to maintain optimum functioning of the enzymatic reactions in the body a constant temperature is required. This constant body temperature is not very constant in the sense that it varies somewhat during exercise and also when the animal is exposed to extremes of temperature of the surroundings. That means that the temperature regulation mechanisms in the body are not 100 percent perfect.

At this stage it is desirable to clarify the distinction between surface temperature and core temperature. The surface temperature, in contrast to the core temperature rises and falls with the temperature of the surroundings. It is the temperature of the deep tissues or internal organs which remains constant. The temperature of the rectum is usually taken as a useful indicator of core temperature because of the ease of measurement. The rectal temperature of several domestic animals is given in Table 1.

Since, designing, producing, and maintaining a sophisticated Website is very expensive, protecting content ownership is extremely important. Electronic commerce will continue to be highly competitive. As Websites become more and more interactive with consumers, their creation, design, and maintenance place enormous demands on innovative marketing techniques that should be legally protected.

Never before has it been so easy to violate a copyright owner’s exclusive right to copy the material. Everyone with a computer and an Internet connection creates his own Web pages and thus become a publisher. Hence the rules that once applied to only a few companies bind million of people now. Imagine having paid a Website developer hundreds of thousands of rupees to launch a killer application and acquiring millions of Rupees from a venture capitalist firm, only to discover that another e—business is using your online material. Or consider a scenario where your Website is linked to an e-business that displayed copyrighted material without the owner’s consent. A farmer has to make judicious decisions on the use of scarce resources, having alternative uses to obtain the maximum profit and family satisfaction on a continuous basis from the farm as a whole. In other words, management seeks to help the producer in deciding problems like what to produce, how much to produce and when to buy and sell and organization and managerial problems relating to these decisions.

Management – Definition

According to Lawrence A. Appley, management is an art of getting things done through the efforts of other people.

Every small business manager is a personnel manager in the sense that work is done through people, with people and for the people. Consequently, the owner-manager should be personally capable of handling employee relations until the company becomes large enough to afford a personnel manager. Planning personnel requirement, developing sources from which new employees can be recruited, choosing (recruiting) the needed people, training and developing them into productive workers, evaluating their performance, compensating them and dealing with various personnel relationship, including industrial relation are included under personnel management.

Staffing

Staffing is a critical function of organizing and managing a successful business. All companies, whether large or small, are involved in the staffing process. All businesses run the same risk every time they hire a new employee. The staffing function is generally divided into four major categories (1) staffing needs, (2) acquisition, (3) motivation, and (4) retention.

The science of Meteorology is defined as the science that deals with physical and chemical processes of the lower atmosphere. The physical and chemical changes in the lower atmosphere produce weather and popularly known as the weather science. The weather science concerns the study of structure, state and behaviour of the lower atmosphere. It is essentially an observational science dealing with different atmospheric variables such as atmospheric pressure, temperature, wind, humidity, density, cloud amount, radiation, precipitation and related variables at a given moment and attempts to predict the behaviour of atmosphere over a period of few hours to few days ahead and disseminate the same in time to the users? community. The statistical approach of weather phenomena deals with the mean state of the physical properties of atmosphere. This branch of science is popularly known as Climatology. The weather science can be categorized into several branches based on method of approach, purpose and function, scale and region. Based on the purpose and function, it can be divided into several branches like Livestock Meteorology, Agro Meteorology, Forest Meteorology, Hydro Meteorology and Bio-Meteorology (Fig 1.1).

Selye, H (1936), the pioneer in stress physiology, defined stressors as noxious stimuli that increase adrenocorticotropic hormone and clubbed these stimuli under the term stress. Rivier and Rivest (1991) and Chrousos and Gold (1992) defined stress as “a state of disharmony, or threatened homeostasis.” Stress challenges homeostasis, but does not disrupt it, in an adapting organism. McEwen (2000) defined stress as a “real or interpreted threat to physiological or psychological integrity that results in physiological and/or behavioral responses.” The following are the various stressors:

Air, water and land constitute our environment and these are technically known as atmosphere, hydrosphere and lithosphere respectively. All these spheres together constitute the biosphere. In the biosphere, apart from human beings, plants, animals, birds, fishes, insects and microorganisms also exist. All these living beings get affected whenever a change, physical or chemical occurs in the biosphere. This change is termed as pollution and the agents that institute these changes are called pollutants. A kind of pollution that has come in to prominence more recently is radiation, causing environmental and health problems of immense magnitude. The radiation may either be natural like cosmic radiation from outer space and sun?s radiations or produced by disintegration of naturally occurring and manmade elements. Such radiation is not visible to our eyes, but can penetrate the body and do immense harm. Man and animals have been continually exposed to different sources of radiation. But the radiation levels were well within the tolerable limits in the past. Yet, since the mid 1970?s human activities have been changing the chemistry of atmosphere in a way that reduces the amount of ozone in the stratosphere which is responsible for protecting the living things from excessive amounts of ultraviolet radiation. Further, it is only after the advent of nuclear power stations that radiations increased to reach harmful levels. Though mankind has benefitted by harnessing nuclear energy for peaceful purposes, one cannot deny the inherent dangers of radiation. Nuclear power plants no doubt help in the production of controlled electricity. However, a worldwide controversy is ragging over the existing nuclear power plants and setting up of new plants particularly since 1986 disaster of Chernobyl nuclear plant in Russia. A fire in the plant allowed massive amounts of radiation to be released into the atmosphere. This fall out drifted into the Scandinavia and Eastern Europe causing enormous damage to crop and livestock.

21.1 Automatic Weather Station

Automatic Weather Station, popularly known as AWS, is an electronic device and an automated version of the traditional weather station. Numerous weather sensors are connected to the system which automatically records and stores weather data. It is quite useful in remote areas where no frequent access is possible. The system requires hardware as well as software to log in, store, display, retrieve and analyze the data. The digital data recorded from AWS can be downloaded to the laptop through the memory device. The same digital data can be converted to the actual values indicating various weather parameters through the software provided along with the AWS. It should be also maintained in the desktop of PC/AT so that the data are doubly assured. Every AWS has its own software depending upon the manufactures. Time interval can be fixed for recording weather data depending upon the purpose since it is a continuous recorder. Continuous AWS data are integrated at hourly or half-hourly interval for monitoring weather data, otherwise enormous data are continuously stored and no way are used. Of course, continuous weather display is possible for monitoring weather conditions spatially and temporarily.

Automatic Weather Station consists of a datalogger, rechargeable battery, telemetry (optional) and the meteorological sensors with an attached solar panel. The system may report in near real time via GPRS or save the data for later recovery. Majority of automatic weather stations possesses normally Thermometer Sensor for measuring temperature; Anemometer for measuring wind speed; Wind vane for measuring wind direction; Hygrometer for measuring relative humidity; Barometer for measuring atmospheric pressure; Rain gauge for measuring rainfall and Pyranometer for measuring solar radiation. Automatic weather stations are based around a programmable datalogger that measures the sensors, then processes, stores, and transmits the data. Dataloggers have wide operating temperature ranges, on-board instructions, programmable execution intervals and input channels for commonly used sensors. Wind vector, wet bulb, histogram and sample on maxima or minima are standard in the datalogger instruction sets. Most sensors can be measured directly without external signal conditioning. Data are typically viewed and stored in the units of your choice(e.g., wind speed in mph, m/s, knots). Measurement rates and data recording intervals are independently programmable, allowing calculation of 15 minute, hourly, and daily data values from 1 minute or 1 second measurements, for example. Conditional outputs, such as rainfall intensity and wind gusts, can also be recorded. The program can be modified at any time to accommodate different sensor configurations or new data processing requirements.

Rearing poultry industrially is a necessity in the present condition where the demand is high and the land holdings for extensive rearing is diminishing. All over the world the poultry industry is growing rapidly. The poultry industry in India is growing at around 8 per cent per annum. The poultry population of the country mainly consists of chicken, ducks, quails, ornamental birds and ratites. Chicken form is the major component of the industry. They are mainly reared as broilers for meat purpose and as layers for eggs. The climatic variations and their effects on profitable poultry rearing are an important topic of study in the current situation, where intensive rearing and global warming are being discussed with equal importance. The welfare aspects of the birds are also a matter of concern all over the world. Thus the knowledge of normal physiology of poultry, the effects of the tropical climate on them and the measures to alleviate detrimental effects produced by adverse climatic conditions have been discussed in detail here.

Abstract

Geological conditions in the Tertiary mountainous areas of the western and eastern hill ranges of Manipur often cause landslide movements. These repeatedly occur mostly in old landslide regions during rainy seasons on a large scale, causing considerable economic loss and injuries and deaths of humans. In order to improve such landslide hazard assessment of the region for effective mitigation, Electrical resistivity surveys were conducted using Schlumberger array technique on the very unstable and problematic frontal part of the Kotland landslide, NH-37, Manipur along three profiles. Electrical resistivities aided the characterization and quantification of the subsurface rock units, structure and tectonic conditions of the Landslided area which can contribute for the better planning of mitigation strategies. The results lead to the elucidation of depth of the different litho-units and the geometry of failure surface. The probable failure surface was observed between 10 and 22m depth, characterised by low resistivity (< 150 Ωm) and geometry of the sliding surface was rotational. In addition, a fault was also inferred in the resistivity pseudosection model and hence the area may be highly vulnerable to landslides.

Abstract

The Nilgiri Mountains of South India is one of the Landslide prone hill ranges in India. The landslides, in general, are due to the independent, interdependent and the combined effects of natural earth system processes, triggering phenomenon and anthropogenic activities. So the scientists, technocrats and the academicians from all over the world have started studying the landslides using various techniques and advanced tools in mapping the morphology, preparing the vulnerability maps and carve out mitigation strategies. But the discrimination of active slides is a difficult task and also needs continuous monitoring and further a time consuming process too. So, in this context, an attempt has been made to study the signatures of active sliding in one of old slides namely the “LINGASLIDE” in which frequent minor sliding events are often reported. In the study, Total station based large scale topographic survey was carried out in LINGASLIDE and from the same 50 cm topographic contours were generated. From such large scale contours, Digital elevation Model (DEM) was generated and from the DEM, shaded relief map was generated.Uutilizing both the contours and shaded relief maps, small slides were mapped in the LINGASLIDE from the well exhibited subsidiary crowns, toes and the traverse cracks. Resistivity tomography surveys were also carried out along two longitudinal profiles and one traverse profile to identify the subsurface geology, the slip surface and the zones of moisture accumulations, if any. The subsidiary crowns, toes, the conjugate fracture systems and the resistivity tomography showed that the LINGASLIDE is active and the sliding is going on towards easterly because of the easterly compression,resulted from the higher slopes caused by the settlements raised over Lingaslide and the obstruction provided by the retaining walls at the toes in the east. Such compressive forces are clear from the NW-SE and NE-SW fractures seen in Lingaslide and the sliding is clear from the cracks seen and monitored in the buildings constructed over Lingaslide.

11.1 Introduction

Economic analysis is the most important parameter for determining the optimum size of farm pond. It is, no doubt that a big size farm pond can meet the irrigation demand of the crops grown in the command areas in addition to meeting the other demand in the command areas. But a big size farm pond is an oversize structure which necessitates large investment to be incurred which may not be economically viable. On the other hand, a small size structure may not be able to meet the crop water demand and also other demands in the command areas and so is called as under-designed structure. It has been observed that as the size of the farm pond gradually increases from a certain minimum size, the total investment cost increases. At the same time, the benefits from the increasing storage capacity of the farm pond also get increased. In such cases, from a purely business point of view, the most economical capacity is that which provides the highest rate of return on investment. As the size of the farm pond increases, the proportion of the benefit/profit to the cost also increases from certain minimum size, attains maximum value at a particular size and then declines as the size further increases. This particular size of the farm pond at which the net profit is maximum, is called as the optimum size. It is to be noted that at the optimum size, the other economic indicators like the benefit-cost ratio and internal rate of return are also high. The variation of net profit with size of the farm pond is a concave curve. The size of the farm pond corresponding to the peak of the curve which represents the highest net profit is the optimum size of the pond.

The principle of obtaining the optimum size of farm pond is to first assume a certain minimum size say five percent of the farm area and conduct the economic analysis by studying the costs and net profits of the assumed size. Then the size is gradually increased and the economic analysis is conducted for each increased size. Then a curve relating the size of the farm pond and net profit is drawn. Such a curve is generally concave shaped. The size of the structure corresponding to the maximum net profit gives the optimum size. It is to be noted that any size chosen more or less than the optimum size would give less net profit and benefit-cost ratio than that of the optimum size.

Sustainable development and efficient management of water is an increasingly complex challenge in India. Increasing population, growing urbanization, and rapid industrialization combined with the need for raising agricultural production generates competing claims for water. There is a growing perception of a sense of an impending water crisis in the country. Some manifestations of this crisis are:

(a) Increasing costs of developing new water resource – Many major and medium irrigation projects seem to remain under execution forever as they slip from one plan to the other with escalating cost and time overruns.

(b) Siltation of reservoirs and owing to lack of maintenance, the capacity of the older irrigation systems seems to be going down.

(c) Declining groundwater table due to over-exploitation imposing an increasing financial burden on farmers who need to deepen their wells and replace their pump sets and on State Governments whose subsidy burden for electricity supplies rises.

(d) Water pollution and degradation of water-related ecosystems - Water in most parts of rivers is not fit for bathing, let alone drinking. Untreated or partially treated sewage from towns and cities is being dumped into the rivers. Untreated or inadequately treated industrial effluents pollute water bodies and also contaminate groundwater,

(e) Wasteful use of already developed water supplies, often encouraged by the subsidies and distorted incentives that influence water use,

(f) Besides in many rural habitations there are pockets where arsenic, nitrate, and fluoride concentration in drinking water are posing a serious health hazard

(g) Rise in water-logging and salinity resulting in degradation of soils in irrigated areas,

(h) Increasing water conflicts about water rights between upper and lower riparian states in a river, conflicts about quality of water, people’s right for rainwater harvesting in a watershed against downstream users, industrial use of groundwater and its impact on water tables and conflicts between urban and rural users etc

(i) The gross irrigated area does not seem to be rising in a manner that it should be, given the investment in irrigation. The difference between potential created and area actually irrigated remains large. Unless we bridge the gap, significant increase in agricultural production will be difficult to realize.

Microorganisms, being ubiquitous, are found in soil, air, water, food, sewage, on body surfaces as well as on degrading and decomposing substances. Their survival and continued growth mostly depend on an adequate supply of nutrients and favourable growth environments. The culture media (nutrients) consisting of chemicals support the growth of microorganisms in a given condition. They are devised in such a way that the organism should get all the nutritional requirements. These are usually prepared in laboratory by weighing and dispensing the individual ingredients. Generally, the common media contain both inorganic and organic nutrients; however, for the cultivation of large number of microorganisms, specialized media are required.

Basically the culture media are of three types: (1) Natural, (2) Semi synthetic and (3) Synthetic.

10.1 Transformation of E. coli by electroporation Principle

Preparation of electro-competent bacteria is considerably easier than preparing cells for transformation by chemical methods. Bacteria are simply grown to mid –log phase, chilled, centrifuged, washed extensively with ice cold buffer containing 10% glycerol. DNA may be introduced immediately into the bacteria by exposing them to a short high voltage electrical discharge. Alternatively, the cell suspension may be snap frozen and stored at-70^oC for 6 months before electroporation, without loss of transforming efficiency.

Since E.coli cells are small, they require very high strengths (12.5- 15KV cm^-1) for electroporation as compared to those used to introduce DNA into eukaryotic cells. Electroporation is temperature dependent and is best carried out at 0-4°C.

The highest efficiency of transformation is obtained when the concentration of input DNA is high (1-10 μg/ml) and when the length and intensity of the electrical pulse are such that only 30-50% of the cells survive in the procedure. Under these conditions, as many as 80% of the surviving cells may be transformed. Higher frequencies of transformation (colonies/ molecules input DNA) are obtained when the DNA concentration is low (?10pg/ml).

The method outlined in this protocol is followed with most strains of E.coli and with plasmids <15kb in size. However, substantial variation in the efficiency of transformation between the strains of E. coli. has been reported. As in case with chemical transformation, linear plasmid DNAs introduced into E.coli by a pulsed electrical discharge transform very efficiently from 10 to 1000 fold but less efficiently than the corresponding closed circular DNA – perhaps because the exposed terminal of linear DNA are susceptible to attack by intracellular nucleases.
