Therefore, it is essential to establish biosafety regulations and risk assessment frameworks to promote safe and responsible use of biotechnology. This chapter compiled an overview of biotechnology, the importance of biosafety regulations and risk assessment, IPR protection, contaminants, risk analysis, risk management in laboratories and fields, biosafety concerns to human health, environment and ecology, biosafety at international and national levels, bioethics, and general principles.

Overview of Biotechnology and its Potential Benefits and Risks

Biotechnology refers to the use of living organisms or their components to develop products and processes for various fields such as healthcare, agriculture, and environmental remediation. Biotechnology applications include genetic engineering, bioremediation, biopharmaceuticals, and biobased products.

8.1 Steps in Biosafety Risk Assessment
8.1.1 Introduction
Biosafety Risk Assessment is a systematic, science-based process used to identify, evaluate, and mitigate risks associated with handling biological agents, genetically modified organisms (GMOs), and biotechnological products. It is central to ensuring human safety, environmental integrity, and regulatory compliance in both laboratory and field applications.
A well-conducted biosafety risk assessment enables institutions to establish appropriate biosafety levels, implement containment measures, and ensure sustainable and responsible use of biotechnological advancements.
8.1.2 Key Objectives of Biosafety Risk Assessment
• Prevent exposure of personnel to biohazards.
• Avoid environmental contamination and unintended gene flow.
• Determine containment requirements for biological agents.
• Guide institutional and national biosafety decision-making.
• Support regulatory frameworks and compliance obligations.

13.1 Biosafety

Biosafety is the prevention of large-scale loss of biological integrity, focusing both on ecology and human health. These prevention mechanisms include conduction of regular reviews of the biosafety in laboratory settings, as well as strict guidelines to follow. Biosafety is used to protect from harmful incidents. Many laboratories handling biohazards employ an ongoing risk management assessment and enforcement process for biosafety. Failures to follow such protocols can lead to increased risk of exposure to biohazards or pathogens. Human error and poor technique contribute to unnecessary exposure and compromise the best safeguards set into place for protection. The international Cartagena Protocol on Biosafety deals primarily with the agricultural definition but many advocacy groups seek to expand it to include post-genetic threats: new molecules, artificial life forms, and even robots which may compete directly in the natural food chain.

Introduction

Biosecurity includes a set of standard scientific measures, adopted to exclude pathogens from culture environments and hosts and, more broadly, limit the establishment and spread of pathogens” (Mohan et al., 2003; Moss et al., 2012). Biosecurity is an emerging concept, and the phrase is changing as usage differs across nations and among different professional groups. A holistic approach to addressing biological risks associated with food and agriculture is what is widely referred to as "biosecurity.

Biosecurity means protection against infectious biological agents. These agents include bacteria, viruses, protozoa, fungi, parasites, and any other agents capable of introducing infectious diseases into a poultry flock. Biosecurity therefore means maintaining a flock, poultry house or premises in such a way that infectious agents do not have a chance to enter to cause diseases. The biosecurity may be categorized into three: Conceptual biosecurity, Structural biosecurity and operational Biosecurity.

Importance of Biosecurity

In modern poultry production, birds are reared under intensive system where large numbers are concentrated in small area. As a result, when an infectious agent enters a poultry house, there is probability for widespread dissemination if it is not contained. Besides, the broiler and egg industry of today is under increasing consumer and regulatory pressures to guarantee food safety and meet export requirements. As a result, broiler and layer operations around the world require the supply of breeding stock free of salmonellas specific to avian species (such as S. pullorum and S. gallinarum) and those that could cause outbreaks of human food-borne illness. Inadequate biosecurity can contribute to wide epidemics of highly pathogenic or exotic diseases. An infection by a virulent organism within a facility can be devastating, reducing without overt signs of disease. Once contaminated with pathogens, poultry facilities are extremely difficult and expensive to clean, sanitize and disinfect. Prevention of disease is always less expensive than treatment. The cost of implementing biosecurity system is smaller when compared to financial profit that can be made from production. Good biosecurity is therefore vital requirement in the best possible control of disease.

Abstract

The use of biosensors is growing in the food industry as a means to ensure food safety and quality because of its versatility in determining biological compounds. It is defined as analytical technique used for detection of analyte that combines a biological component with a physico-chemical detector. The most important contribution of biosensors in the food sectors is to control production processes and ensure food quality and safety by reliable, fast and cost effective procedures. Biosensors have been useful for the evaluation of food composition, particularly in functional food products fortified with phytochemicals, vitamins and minerals.. Biosensors are replacing conventional analytical tools since they offer advantages in size, cost, specificity, rapid response, precision and sensitivity. Future progress in the biosensor technology involves the development of nutraceutical release technology within packaging material where it can be used to trigger the controllable release of bioactive components and targeted delivery of protective agents into the food product. The growing consumer demands for safe and quality food has led the food industries to search for quick and reliable methods for which the applications of the biosensor technique in the field of food processing and quality control are found to be promising.

Introduction

Biosensor technology is a powerful alternative to conventional analytical techniques, harnessing the specificity and sensitivity of biological systems in small, low cost devices. Despite the promising biosensors developed in research laboratories, there are not many reports of applications in agricultural monitoring. The authors review biosensor technology and discuss the different bio-receptor systems and methods of transduction. The difference between a biosensor and a truly integrated biosensor system are defined and the main reasons for the slow technology transfer of biosensors to the market place are reported. Biosensor research and development has been directed mainly towards health care, environmental applications and the food industry. The most commercially important application is the hand-held glucose meter used by diabetics. The agricultural / veterinary testing market has seen a number of diagnostic tests but no true biosensor systems have made an impact.

Introduction

The food that we consume should not only contain all essential nutrients like carbohydrate, protein, fat, vitamins, minerals, fibre etc but also should be hygienic and safe. Safety and quality of food is to be ensured not only during food production but also should be continuously monitored till it reaches the consumer (Coles and Frewer, 2013). The characteristics of food are altered during every processing operations viz., cleaning, sorting, transportation, thermal processing, packaging, storage, distribution etc which may be desirable or undesirable. Persistent physical, chemical and microbiological analyses have to be done at all levels of supply chain to ensure the quality of food. Traditional methods of food safety analysis are spectrophotometry, aerobic plate count method, microscopic plate count, titrations, chromatography etc. Though these techniques provide accurate results, the analysis demand cumbersome extraction steps, trained technical staff and are time consuming (Di-Natale et al., 2000). Hence, steady, sensitive and rapid monitoring of food analysis technique is a need of the hour in the present era. Biosensors can effectively replace the conventional techniques owing to its reliability, rapidity, economic and field applicability (Prasad et al., 2014). Biosensors are an attractive diagnostic alternative owing to their in situ detection ability and comparatively reduced detection time from several days to hours, or even minutes (Ruiz et al., 2017). Rapid food analysis can also minimise spoilage, especially in freshly harvested agricultural commodities, such as fruits and vegetables, meat, fish etc. In addition, the portable detection devices provide in-situ detection capabilities without extensive training and assist in integrating real time analysis in food processing centres.

16.1 Introduction

Food safety is most important issue in today's global environment. To safeguard food supply is a very difficult and challenging responsibility, as a consequence of which the world's largest food companies, retailers, and manufacturers have placed very high food safety standards. Although the safety of food has improved exponentially but progress is uneven and food borne outbreaks due to microbial contamination, pesticides, antibiotics chemicals and toxins are common in many countries. International trade statistics (2007) by World Trade Organization (WTO) reported that Europe has accounted for 46% of world exports of agricultural products, where food represents 80% of agricultural exports. The trading of contaminated food among countries increases the risk for outbreaks and consequently, health risks due to microbial pathogens in food are of major concern to all governments.

Biosensors are an important option in the agricultural and food sectors to control production processes and ensure food quality and safety by dependable, fast and cost effective methods. Biosensors have plenty of advantages over conventional analytical tools since they offer advantages in size, cost, inherent specificity, selectivity, rapid response, accuracy and sensitivity. Food microbiologists are always in search of rapid and reliable automated systems for the detection of biological activity. Biosensors provide sensitive, miniaturized systems that can be used to detect unwanted microbial activity or the presence of a biologically active compound, such as glucose or a pesticide. Immunodiagnostics and enzyme biosensors are two of the leading technologies that have had the greatest impact on the food industry. The use of these two systems has reduced the time for detection of pathogens such as Salmonella to 24 hour and has provided detection of biological compounds such as cholesterol or chymotrypsin. The another useful application for biosensors, is detection of genetically modified organisms (GMO), since several countries have laws regulating the commercialization of GMO products and their derivatives. Biosensors are also useful in the implementation of hazard analysis and critical control points (HACCP) plans by verifying process developments and correcting errors in due time. There is joint application of Biosensor technologies and nanotechnologies in many food and agricultural applications such as the development of nano-scale tools for biosafety, nanoscale compounds for food packaging, and nano-sensors for pathogen detection in animals and plants. These are helpful tools in the detection and control of potential food contaminants by the agricultural and food industries.

Though primarily looked at for coastal regions, under a broader definition Bioshields can play important role in semi-desert areas, hills, etc. also.

• Bio-Shields in coastal areas include raising mangroves, casurina, palms, bamboo & other tree species and halophytes like Salicornia, etc. followed by other regular plants for the land.
• They serve as speed-breakers under conditions of coastal storms, cyclones and tsunami.
• In addition, they serve as carbon sinks, since they help to enhance carbon sequestration and sustainable fisheries and agriculture.
• Integration of Bio-shield with livelihood options and eco-restoration by developing different site specific models is therefore very important.
For example- the case of two villages near Kalpakkam in Tamil Nadu is often taken. During the devastating Tsunami sometime back, the village with plantation- a natural bioshield was saved with hardly any loss of life as against a nearby village with no natural bioshield. Several such examples are available world over. This indicates that if we can have a good, scientifically designed bioshield, it can be of a great help. This concept is bound to get much greater emphasis when we consider the increasing effect and danger due to climate change in the coming years.

Abstract

Soil microorganisms play a major role in the biodissolution of nutrients bound in the soil silicate minerals. Plant nutrients like K, P, Ca, Mg, Fe, Zn, and Mn are bound in them. Silicon is considered as agronomically important as it confers rigidity and strength to crop plants and enhances yield. Biodissolution of these nutrients result in their availability to crops influencing the growth, biomass and yield. The microbial metabolites like organic acids, amino acids, inorganic acids, amines, ammonia, phenolic compounds, extracellular polysaccharides, siderophores, ligands of cations and cation exchange are involved in the biodissolution of K, Zn and Si. Frateuria aurantia, Bacillus mucilaginosus and B. edaphicus and Bacillus sp are recognized as potash mobilizers, silica and zinc solubilizers.

Abstract

Contamination of environment with heavy metals has become a serious global problem due to its threat to animal and human health. Removal of this non-biodegradable, hazardous heavy metals through bio-materials has received paramount interest and intense attention in the last few decades as bio-materials play crucial role in remediating heavy metal polluted environment through a process called “biosorption”. The term biosorption, therefore, has become a popular theme in recent years due to its relatively simple, inexpensive, eco-friendly with rapid mechanism of removing heavy metals from polluted environment. There have been burgeoning studies around the globe for alleviating the problem of heavy metal pollution by selecting and developing an effective bio-sorbent with minimum treatment process for field application. This review aims to provide vast information on types of biosorbent available, their exploitation for better efficiency and their economic attractiveness for the restoration of heavy metal polluted environment.

15.1 Introduction

Statistics is mostly used as a tool for assessing the capability of the system. Statistics refers to facts shown in numbers. Modern statistical methods and data are being found increasingly useful in research in different fields such as hydrology, biology, water quality and forestry etc. Statistics when used effectively becomes so intertwined in the whole fabric of the subjects to which it is applied. Statistics assists in planning the initial observations, organizing them and formulating hypothesis from them and in judging whether the new observations agree sufficiently well with the predictions from the hypothesis. In statistical methods, there is an operation of multiple causes and the investigator has only limited control over there. But statistical methods are found extremely useful in predicting the consequences of a situation. It requires skill for acquiring and handling data. There is a methodology to deal with the data and analysis of data is based on scientific and engineering strategies (Ramakrishnan, 1995).

In a huge rainfall data over a period of 300 years, it is not possible to observe all the items. Statistical methods are developed to estimate the error or deflection from the true or real value when we are taking inference from a sample for a hydrological data.

In the modern world where life sciences is getting more and more importance. To meet our environmental problems and energy requirements due to rapid industrialization and population growth, scop of life science has taken its new shape. Based on statistical analysis, new field like environmental sciences, hydrology, genetic engineering, biotechnology and biosociology have been evolved. Experiments about crop yields with different types of fertilizers and different types of soils or the growth of animal life and plant life under different environmental conditions and diets are very often designed and analyzed according to statistical method. Even in the field of medicine and public health, statistical methods are used for testing the efficacy of new medicine and method of treatments. As a matter of fact, for a research worker in any field, which is concern with numerical results, a study of statistical method is not only useful but also necessary. Hence it can be rightly by remarked “Science without statistics bears no fruit, statistics without science have no root”.

A primary issue for modern horticulture is facing two contradictory objectives, such as the need to produce food for the increasing world population and to minimize damage to the environment, which can in turn negatively impact horticulture. Fertilizer and chemical use and manufacture are hazardous to the environment and humans. As a result, new and effective environmentally friendly compounds are required. Plant biostimulants, which are harmless for humans and the environment, are one answer. They are particularly useful for lowering pesticides in agriculture. Plant biostimulants have become popular in farming and cultivation in recent years.

Agriculture worldwide is undergoing a profound shift toward sustainability, driven by the need for increased productivity and environmental stewardship. Among the many innovations shaping this transition, biostimulants have gained prominence as a vital tool for enhancing crop performance, improving soil health, and promoting sustainable farming. Unlike conventional agrochemicals, biostimulants work by activating natural plant processes, enhancing nutrient uptake, and increasing resilience to environmental stress. The global agrichemical industry now distinctly categorizes biopesticides, biofertilizers and biostimulants.
WHAT ARE BIOSTIMULANTS?
Biostimulants are natural synthetic substances that enhance plant growth and development without being fertilizer or pesticides. They work by improving nutrient uptake, increasing stress resistance, and stimulating beneficial microbial activity in the soil. Unlike chemical fertilizers, which directly supply nutrients, biostimulants enhance the plant’s ability to utilize nutrients more efficiently.
TYPES OF BIOSTIMULANTS
Biostimulants are categorized based on their composition and mode of action:
Humic and Fulvic Acids – Improve soil structure, enhance nutrient availability and increase root growth.
Seaweed Extracts – Rich in phytohormones and trace elements that promote plant growth and stress resistance.
Amino Acids – Play a crucial role in protein synthesis and help plants recover from environmental stress.
Microbial Biostimulants – Beneficial bacteria and fungi that enhance nutrient uptake and improve soil health.
Brassinoloilds & Tricontanols – Help in vegetative growth and fruit development.
Alkaloids – Improve plant defense mechanism.

13.1 Introduction

A surfactant or surface-active agents are the compounds that lower the surface tension between the two liquids or between a liquid and a solid. Surfactants may act as detergents, wetting agents, emulsifiers, foaming agents and dispersants. Surfactants are normally organic compounds that are amphiphilic, meaning they contain both hydrophobic groups or hydrophilic groups. Therefore a surfactant contains both a water insoluble component and a water soluble component. Surfactant will diffuse in water and adsorb at interfaces.

The interfacial free energy is the minimum amount of work required to create that interface and the interfacial tension between two phases is determined as the interfacial free energy per unit area. The interfacial (or surface) tension is also a measure of the variance in nature of the two phases meeting at the interface (or surface). The greater the dissimilarity in their natures, the greater the interfacial (or surface) tension between them. A surfactant is therefore a substance that changes the amount of work required to expand those interfaces. Surfactants usually act to reduce interfacial free energy.

Surface active agents in the form of emulsifiers have a long history of use and are widely used in many products being used in everyday life. An extensive basic research is required to understand the reason of instability and methods to prevent the breakdown of emulsions. Emulsifiers can function as dough conditioners by improving tolerance to variations in flour quality, better gas retention resulting in lesser yeast requirements, increased uniformity in cell size, a finer grain, more resilient texture and improved slicing. Also, the characteristics of freshly baked bread like soft crumb can be retained longer, if the appropriate emulsifiers are added. Commonly used surface active agents in bread production are propylene glycol mono- and diesters, polyoxyethylene sorbitan monostearate, ethoxylated mono- and diglycerides, succinylated monoglycerides and Diacetyl tartaric acid, esters of monoglycerides. Cookie characteristics like top grain, volume and spread ratio can also be improved. Here surfactants used are sodium stearoyl fumarate, sucrose fatty acid esters, sodium stearoyl lactylate, sorbitan fatty acid esters, polysorbates and ethoxylated monoglycerides. In dairy products like Ice cream surface active agents play some important functions like to improve fat dispersion and control fat agglomeration, facilitation of fat– protein interactions, facilitate air incorporation, give smoother texture due to smaller ice crystals and air cells, increase resistance to shrinkage, improve melt-down and reduce whipping time. In candy products, the elimination of “bloom,’’ i.e., the transition of fat crystals is a key reason for the addition of surface active agents. In mixtures of triglycerides, surfactants like sorbitan esters (Spans) and ethoxylated sorbitan esters (Tweens) can control product viscosity in chocolates and in cream fillings and modify the crystal structure. Surface active agents have been utilized in the production of meat analog products also. In margarine, Emulsifiers fulfil several functions like modification of crystal structure in the vegetable fat, assistance in emulsion formation and antispattering. Here, a mixture of citric acid monoglycerides and monoglycerides or lecithin can be used. The properties and mode of action of these chemical surfactants is discussed in the following sections.

ABSTRACT

The use of engineered nano-materials has been increased as a result of their positive impact on many sectors of the economy including agriculture. Silver nanoparticles (AgNPs) are now used to enhance seed germination, plant growth and as anti-microbial agents to control plant diseases. This study investigated the synthesis of silver nanoparticles using the aqueous solution of three medicinally important plants (such as Spondius mombin L., Stachytarpheta jamaicensis (L.) Vahl and Syzygium samarangense (Blume) Merr & Perry leaf extract in room temperature (350C). AgNPs present in the leaf confirmed by colour change, UV-VIS spectrum analysis, average size of AgNPs confirmed by PAS. FTIR measurement were carried out to identify the other possible biomolecule groups such as alkenes, esters, phenols, alcohols and aromatic. Leaf extract AgNPs were shown significantly higher antimicrobial activities against four species of bacteria. This findings suggest that the seed germination percentage, relative seed germination rate, relative shoot and root growth and germination index of the tested plant depends upon concentrate gradient of AgNPs.

ABSTRACT

Recognizing the need to apply plant tissue culture tools for generating planting materials of important spice crops especially vanilla, the Spices Board started Biotech Production Unit at its head quarters building at Kochi with an annual production potential of 8 lakhs plantlets. Complete tissue culture protocols were developed and scaled up for large scale production of vanilla and other spices viz. large cardamom, small cardamom and tree spices. The laboratory is equipped with all modern facilities and is established in an area of about 2000 sft. Department of Biotechnology, Govt. of India also participated in the venture by sanctioning a project entitled “Regional centre for large scale production of tissue culture plantlets of vanilla and other spices” with a financial out lay of Rs 95 lakhs equally shared by DBT, New Delhi and Spices Board, Cochin.

At present there is a stock of over 3.00 lakh vanilla cultures in the lab and already 1.25 lakh plant lets were sent for hardening. The Spices Board, in coll aboration with “Kudumbashree”, a poverty eradication mission of Govt. of Kerala, extend short term training in tissue culture techniques to undergraduates from ‘Below Poverty Line’ (BPL) families and engage them as long term trainees in the Biotech Production Unit. The Board also coordinates and extends support to Kudumbashree members for, establishing and maintaining hardening facilities for TC plantlets thus providing means for empowering women from BPL families.

Tissue culture consists of growing plants cells as relatively on organized masses of cells on an agar medium (callus culture) or as a suspension of free cells and small cell masses in a liquid medium (suspension culture). In other words, In plant cell culture, plant tissues and organs are developed in vitro on artificial media, under aseptic and controlled environment. The procedure depends mainly on the concept of totipotentiality of plant cells (Haberlandt, 1902) which refers to the ability of a single cell to express the full genome by cell division.

A. Tissue culture and micro propagation
Tissue culture and micro propagation are essential techniques for the propagation of various spices and medicinal plants, allowing the rapid production of diseasefree and genetically identical plants. The overview of techniques apply are as follows:
1. Black Pepper (Piper nigrum)
1a. Tissue culture in Black Pepper
Callus induction from leaf and stem explants, followed by somatic embryogenesis. The protocol for black pepper (Piper nigrum L.) tissue culture involves several stages, including callus induction, shoot regeneration, root initiation, hardening, and implementation of tissue culture techniques. The general outline of the process based on recent advancements are as follows.
a. Callus induction
• Explants: Typically, leaves, stems, or nodes are used as explants.
• Medium: Murashige and Skoog (MS) medium supplemented with auxins like 2,4-Dichlorophenoxyacetic acid (2,4-D) @ 0.5-2.0mg/l or Naphthalene acetic acid (NAA)@ 0.5-1.0mg/l is commonly used to induce callus
formation.
• Conditions: Cultures are usually maintained in dark conditions at 25°C to 28°C to promote callus induction.

The Convention on Biotechnology has defined biotechnology as “any technological application that uses biological systems, living organisms or derivatives thereof, to make or modify products or process for specific use”. The global food requirement is increasing with growing population, so the productivity gap could be filled by improving genetic health. Animal breeding focuses on genetic improvement through biotechnological and genomic approaches. Genetic progress is influenced by the type of biotechnology such as selection accuracy, genetic interval, intensity of selection, genetic variance. It helps to improve animal production, treatment of genetic disorders, production of transgenic animals resistant to diseases, vaccine production and also to maintain genetic diversity for endangered species. Genomic selection is a type of marker assisted selection. It improves the genetic gain by decreasing genetic interval and improving reliability. These novel approaches have high potential in animal breeding, if executed judiciously may provide practical benefits to mankind. The magnitude of genetic improvement of livestock species

Summary

Stem cells are undifferentiated cells which have the potential to divide and multiply indefinitely and can be differentiated to any type of cell of the body of an organism. Due to its peculiar characteristics stem cells are being experimentally used to cure certain dreaded diseases of the humans including different haemoglobinopathies. The stem cells isolated from bone marrow have been cultured, modified and differentiated to get various blood cells like erythrocytes, leukocytes, and thrombocytes etc. Recently haematopoietic stem cells have been transplanted to the sickle cell and beta thalassaemia patients successfully. Gene therapy using the embryonic and cord blood stem cells have been experimentally tried successfully to cure transgenic sickle as well as thalassaemic mice. Induced Pluripotent stem cells (iPS) generated from autologous skin has also been used successfully for treatment of transgenic sickle cell anaemia mouse.

Abstract

Anaerobic microorganisms occur in oxygen-free environments as they possess biomolecules and metabolic processes that have adapted to sustain them in the stressed environments. These biomolecules and metabolic processes are harnessed and employed in various biotechnological applications. Anaerobes are abundant in various ecological niches such as freshwater or marine sediments, subsurface aquifers, deep thermal vents etc. where, in the absence or limitation of oxygen, they utilise reduced ions such as nitrate, sulphate, ferric, carbonate and certain organic compounds like degrading organic matter, fatty acids or alcohols as electron acceptors during respiration/fermentation to produce more or fewer reduced products such as, alcohols, ammonia, organic acids, hydrogen and carbon dioxide as the oxidised product. Diversity of substrates that can be fermented and the wide range of products that can be formed make anaerobic processes useful in biotechnological applications. Their substrate versatility and flexibility, for example, they can utilize simple sugars as well as many of them can utilise complex renewable biomass make their biotechnological applications sustainable and environment friendly. Anaerobes have been employed in food, chemical and material industries for the production of food items, preservative, additives, chemical intermediates, solvents, catalysts and polymers. They find widespread usage in energy sector for generation of various kinds of biofuels that can mitigate our reliance on non-renewable sources for fuel production. An emerging aspect of anaerobic biotechnology is waste valorization that involves treatment of any kind of waste (industrial, municipal, agricultural, pharmaceutical), where organic fraction of the waste is converted into valuable products. Anaerobes play key role in maintaining the global cycles of carbon, nitrogen, and sulfur, and also facilitate bioremediation by breakdown of persistent compounds. Lately, role of anaerobes in medicine and health care sector is widely explored as they are found to produce several bacteriocins, antibiotics, immunomodulators, vitamins and anti-tumor compounds. Some of the anaerobes have also been employed as probiotics. In the present chapter, we discussed these and many more biotechnological applications of anaerobes in white, green, red and blue biotechnology.

Transgenic organisms are defined as being introduced artificially with a transgene and incorporated stably into their genomes. There are various aquatic organisms in which the trans-genesis process has been applied like fish, crustaceans, molluscs, etc. Presently scientists can transfer a particular gene for a desirable and beneficial trait from one organism to another with more precision and in less time than traditional breeding. To date, over thirty-five species have been genetically engineered in laboratories. The transgenic fish are efficient feed converters. Salmon is the first transgenic fish that was used as food. Various techniques are involved in aquatransgenesis. They include – microinjection, electroporation, particle gun bombardment, liposome-mediated gene transfer and so on (setac. onlinelibrary.wiley.com). Through all these techniques a fish is genetically transformed so that it has desirable and beneficial trait. Numerous significant biotechnological uses for fish transgenesis are anticipated to be beneficial across a wide range of industries. The majority of genetically engineered fish serve purposes in fundamental studies of genetics and development. Various research teams have been creating genetically modified zebrafis to identify aquatic pollution. The lab that created GloFish initially aimed for them to alter their colour in response to pollutants, acting as environmental indicators. Transgenic fish have an improved growth rate than their nontransgenic counterparts. Moreover, they are disease-resistant and can be used as models for performing various scientific experiments in the field of environmental toxicology and biotechnology. All these beneficial traits are making transgenic fish more valuable in the biotechnological field. To fully realize the potential of transgenic fish technology in aquaculture, certain ethical and ecological issues must be scientifically resolved.

In our fore-going chapters, we have studied variety of plant products such as alkaloids, glycosides, volatile or essential oils, gums and mucilage and oleoresins with varied role to play to the plants as well as to human.Knowledge of plants and knowledge of healing have been closely linked. It is well established that industries have many direct and indirect effects on human population. Increased stress is most evident at the same time health awareness goes parallel in the recent times. Most of the natural plant products are compounds biosynthetically derived from primary metabolites such as amino acids, carbohydrate metabolic intermediates and fatty acids. These secondary metabolites are the major sources of pharmaceuticals, food additives, fragrance and pesticides.

Application of biotechnology know how helps to synthesize the novel molecules and also could be beneficial to enhance the production of secondary metabolites. Classical definition of biotechnology states that it is “The use of living organisms or their products to enhance our lives and our environment.” The word Pharming is commonly use to describe genetically engineered plants or livestock to produce medically useful products –predominantly protein drugs. Plants or animals are genetically engineered to contain genes capable of producing a biologic or drug compound. Therapeutic proteins are designed to treat illness includes serum proteins, cytokines, growth regulators, anti coagulants, antbiotics, lysosomal enzymes and plant secondary metabolites.

Biotechnology offers an opportunity to exploit the cell, tissue, organ or entire organism by growing them in vitro and to genetically manipulate them to obtain desired compounds. It plays a pivotal role in search for alternatives to production of desirable biomedical compounds. There are several approaches have been used to increase the yield of metabolites from cell cultures such as manipulation of nutrient medium, application of exogenous growth stimulators, chemical treatment, precursor feeding, elicitation, transformed cultures, metabolic engineering, micropropagation, bioreactor cultures and etc.

India with population of 1.30 billion in which 47 per cent is engaged in agriculture earnestly needs efficiently use of biotechnology and genetic engineering for transforming agriculture sector. Biotechnological tools offer great opportunities for increasing agricultural production and can play an important role towards the attainment of production sustainability of crops. Biotechnological tools have the potential to release burgeoning pressure on agriculture and allied sectors; ecology and environment; plant, animal and microbial biodiversity; and allied resources such as land and water with improved and sustainable agricultural productivity which enhances food security. Agricultural biotechnology, including tissue culture, DNA-marker-assisted breeding and genetic engineering (GE) technology have potential to increase stress tolerant, selection efficiency, crop adaptation, crop productivity and food security in more efficiently manner through the development of improved varieties with increased yields, nutritional content and storage characteristics. Genetic engineering tools should continue to build capacity in this technology, giving priority to traits and crops that are of economic importance such as drought tolerance in maize, rice, beans; cold tolerance in rice; increased nutritional value and improved shelf life in fruits and vegetables. Crop varieties and livestock with improved characteristics have been developed using tools such as marker-assisted breeding and genetic engineering. Molecular pharming is one of the applications of genetically modified crops and livestock for the production of pharmaceutical proteins. Reproductive technologies are alternative ways for genetic improvement of livestock. The development of molecular diagnostics and vaccine technology as a tool improved the ability to detect and treat several diseases of livestock. The advancements in biotechnological tools such as protein engineering and metabolic engineering play a significant role in production of a large number of valuable and high quality industrial products.

Introduction Worldwide, 40% of a crop is lost to insect damage, particularly in developing countries. Some of the major insect pests belong to the Lepidoptera and Coleoptera, and their larval or grub stages cause major damage to economically important crops including tobacco, cotton, corn, sorghum, tomato, sunflower, pulses, wheat, and lucerne. Recently, some insecticides have been primarily used for agricultural pests. Over the years, most pesticides have led to insecticide resistance in major lepidopterans, a reduction in natural enemy populations, and harm to mammals and the environment.

‘Tissue culture’ is commonly used as a blanket term to describe all types of plant cultures, viz., callus, cell, protoplast, anther, meristem, embryo and organ cultures (George, 1993). It relies on the phenomenon of cell totipotency. The different techniques of culturing plant tissues may offer certain advantages over traditional methods of propagation.

Growing plants in vitro in a controlled environment, with in-depth knowledge of the culture conditions and the nature of the plant material, ensures effective clonal propagation of genetically superior genotypes of economically important plants. Micropropagation has become an important part of the commercial propagation of many plants (George and Sherrington, 1983; and Zimmerman et al., 1986) because of its advantages as a multiplication system (Pierik, 1997 and Razdan, 2003). Several techniques for in vitro plant propagation have been devised, including the induction of axillary and adventitious shoots, the culture of isolated meristems and plant regeneration by organogenesis and/or somatic embryogenesis (Williams and Maheswaran, 1986; and Gautheret, 1985).

Tissue Culture is becoming as an alternative means to vegetative propagation of plants. In vitro growing plants are usually free from bacterial and fungal diseases. Virus eradication and maintenance of plants in virus free stage can also be rapidly achieved in cultures (Sharma, 2016).

Introduction

Plants are often subjected to unfavorable environmental conditions – abiotic factors, causing abiotic stresses - that play a major role in determining productivity of crop yields but also the differential distribution of the plants species across different types of environment. Some examples of abiotic stresses that a plant may face include decreased water availability, extreme temperatures (heating or freezing), decreased availability of soil nutrients and/or excess of toxic ions, excess of light and increased hardiness of drying soil that hamper roots growth. The ability of plants to adapt and/or acclimate to different environments is directly or indirectly related with the plasticity and resilience of photosynthesis, in combination with other processes, determining plant growth and development, namely reproduction. A remarkable feature of plant adaptation to abiotic stresses is the activation of multiple responses involving complex gene interactions and crosstalk with many molecular pathways.

Pineapple is a short-stemmed herbaceous plant that necessitates a significant amount of planting material. For example, depending on the intensity of planting and the cultivar used, one hectare of cultivation needs 35,000 to 70,000 planting parts. Realizing the need for quick multiplication, Ma (1968) devised a method for quick pineapple multiplication using the decapitation technique. Later a technique for rapid in vitro propagation has been developed. Since then, numerous reports on crop micro-propagation using tissue culture techniques have been published. On supplemented MS medium, different types of explants, such as leaf base, shoot base, excised lateral buds and tips from crowns, young-leaf segments, and excised leaf buds, have been used to regenerate plants. Plantlets raised in this manner thrived in the field. In vitro mutation may also be induced using tissue culture techniques. Shoot tips from in vitro grown plantlets may be treated with chemical mutagens (EMS) or irradiated with gamma rays to produce mutants with superior traits such as spineless leaves, larger fruits, improved pulp consistency, or disease resistance. Similarly, somaclonal variants can be screened out early to improve f ield results.

The primary problem facing humanity at the start of the new millennium is to provide sufficient sustenance for every person living on this continent, given the rapidly growing populations of the majority of Asian countries. As the amount of land per person continues to decline, several strategies are being used to boost agricultural output from these finite resources in order to satisfy the growing demand of the population. But maintaining adequate soil health levels to support healthy plant development is the fundamental requirement for maintaining food production from these shrinking land areas. Questions concerning the over-reliance on mineral fertilizers to maintain the health and, consequently, the productivity of this continent’s arable soils are frequently raised by the recent trend of consistently decreasing the use efficiency of mineral fertilizers under highly productive systems coupled with issues of gradual deterioration of soil health due to indiscriminate use of fertilizers. It is widely acknowledged that mineral fertilizers are limited to providing plant nutrients to soils; they are not able to address other physical, chemical, or biological aspects of soil health. Conversely, organic materials are far more beneficial in this regard. In light of this, the idea of an integrated plant nutrition system that incorporates both organic and inorganic nutrient sources has been developed and is gaining traction (Chattopadhyay, 2005). Because most tropical and subtropical nations’ arable soils have low levels of organic matter due to high temperatures and increased microbial activity, the integration of mineral fertilizers and organic manures is particularly important for many Asian countries (Gaur, 2006).

Wool which obtained primarily from sheep is an essential textile fiber that is used across the world. Whether for clothing, canvas work, carpets, or even upholstery, wool is a vital resource supporting many industries and occupations. This natural fiber has many benefits, such as being biodegradable, easy to clean and even having a natural structure that traps air which is excellent for heat retention. The annual wool production is 2 million tones. China ranks first in terms of global wool production. With a production of more than 333,624 tonnes of wool in 2020, China accounts for approximately 19% of the global wool clip. Australia produces 16% of global wool and is by far the largest wool exporter in the world. There has been a constant increase in the demand of wool products worldwide. This has made scientists to try to improve livestock and livestock associated derivatives. The conventional system along with the recent biotechnological interventions will be helpful for enhancing wool quality and production.

1. Introduction

 Banana is the developing world’s fourth most important food crop (after rice, wheat and maize) accounting for 70 million metric tons per annum (FAO stat, 2007 faostat.fao.org/site/567/Desktop Default.aspx?). Banana is also the second highest fruit crop in the world, representing a wide range of varieties of cooking and dessert types, providing a staple, nutritious inexpensive food for millions of people. The word “banana” is a general term embracing a number of species or hybrids in the genus Musa of the family Musaceae known simply as the banana family. Two Musa species namely Musa acuminata and Musa balbisiana, out of the 28 described were first noted in 1865 by Kurz as being the origin of most of the commercial clones (Stover and Simmonds 1987). Cultivated clones are parthenocarpic, mostly sterile with starchy fruits. Indeed, wild types are very fertile; their fruits are full of seeds with little starch, and are seldom fit for human consumption. Its abundance, varietal diversity, availability round-the-year and low cost make banana an important commodity for the religious occasions in many tropical countries. The vegetable bananas, generally grouped under ‘Plantains’ are larger, more angular than dessert type and are generally intended for cooking unripe, although ripened ones are quite like dissert bananas.

Introduction

The horticulture sector has been a driving force in stimulating a healthy growth trend in Indian agriculture. This sector contributes around 28 per cent of the GDP from about 13.08 per cent of the area and 37 percent of the total exports of the agricultural commodities (Rather et al., 2013)  India is currently producing 257.2 million tonnes of horticulture produce from an area of 23 million ha. Over the last decade, the area under horticulture grew by about 3.8% per annum but production rose by 7.6% per annum. The higher growth rate in horticulture was brought about by improvement in productivity of horticulture crops, which increased by about 28% between 2001-02 and 2011-12. (Anonymous, 2012)

Food processing involves the use of wide range of unit operations and technologies to transform the raw, bulky and perishable food into more shelf-stable, palatable and marketable food products. Food processing is one of the most effective solution to reduce the wastages, improves the quality and safety aspects of food products, increase the food availability and marketability. Fruits and vegetables are considered as an integral part of any dietary system as these are rich sources of vitamins, minerals and dietary fibres. India is the second largest producer of fruits and vegetables after Brazil and China which contributes 10 % of world fruit production and 14 % of world vegetable production. Fruits and vegetables are more prone to spoilage as compared to cereals due to their nature and composition. The spoilage can be occurr at the time of harvesting, handling transportation, storage, marketing and processing that results in lot of wastage in the form of seeds, pulps, peels etc. Efficient management of these wastes can help in preserving vital nutrients of fruits and vegetables, and bringing down the cost of production of processed foods, besides minimizing pollution hazards. In India, processing of fruits and vegetables is extremely low as compared to the developed countries.

Biotechnology can help in the bio-processing of fruits and vegetables for the production of novel products having better sensory and nutritional qualities, improved shelf life. Bio-processes involve the use of different micro-organisms and enzymes along with modern biotechnological techniques like genetic engineering.

Small size grains namely millets are produced by extraneous grasses and the word millet is derived from ‘‘mille’ a French word synonymous to thousand indicating handful of millet grains in a handful measure contains thousand grains (Taylor and Emmambux, 2008). Small millets and pseudo cereals belong to Gramineae family and were major ancient foods of Asian and African continents while Western countries used them as only bird feed.

9.1 INTRODUCTION

India is the second largest producer of fruit crops with annual production of 68.4 million metric tonnes from an area of 6.1million ha (Anon 2010). The national horticulture mission commenced in 2005 had fixed a target to double the horticultural production by 2011-12. According to the available production data of horticultural crops, it seems that we are very close to achieve our target. But, if the loss caused by biotic and abiotic stresses is accounted, a sound conclusion can be drawn that still there is a huge scope for productivity improvement in fruit crops.

Among biotic factors bacteria, fungi, nematodes, viruses and insect pests, diseases and other pests are widely recognized as the biggest threat to the sustainable production of fruit crops. Every fruit crop is infected with one or other pathogen/pest, which causes a significant reduction in yield. The disease alone both in field and storage accounts for 25-30 per cent losses (Rangaswami and Mahadevan 2002). Along with diseases, insect-pests also cause reduction in the yield. About 600 insect and mite pests have been reported in temperate fruits alone in India (Sharma and Singh 2006).

Beside biotic factors, abiotic factors (salt, drought, low and high temperature) also limit the potential yield of fruit crops. It is estimated that nearly 10 million ha is affected by salinity/alkalinity in India (Singh 2006). Moreover, fruit cultivars are also to be modified for yield and traits of consumers preference e.g. seedlessness in grapes and citrus.

Genetic improvement can be achieved by following the conventional breeding approaches, when the gene of interest is present in related cultivars/species and the crop is sexually crossable. But due to long juvenility and genetically prevalent problems like incompatibility, sterility in most of the fruit crops and apomixis in citrus, mango and apple, conventional techniques seem to have limited role in the improvement of fruit crops.

Various biotechnological approaches either alone or in combination with conventional breeding approaches can provide a direct solution for the genetic improvement of fruit crops.

Biotechnology is one of the most exciting and revolutionary sciences of 21st century. According to Convention on Biological Diversity, biotechnology is defined as “any technological application that uses biological systems, living organisms, or derivatives thereof, to make or modify products or processes for specific use”. Biotechnology has proven its immense applications at every step of human life such as, health care, crop improvement, development of valuable products and abatement of environmental problems. Agricultural biotechnology is the area of biotechnology involving its applications to agriculture. Agricultural biotechnology has been practiced for a long time, as people have sought to improve agriculturally important plants/ organisms by selection and breeding.

Biochemical and biotechnological methods present eco-friendly and efficient pathways for converting biomass and organic waste into valuable resources, contributing to environmental protection and sustainable development. These methods rely on microbial and enzymatic processes to break down waste materials, resulting in the production of biofuels, chemicals, and other beneficial byproducts. In today’s pursuit of sustainability and circular economy principles, the concept of waste valorization—turning waste into valuable products—has gained significant traction. Biochemical and biotechnological approaches stand out due to their specificity, lower energy requirements, and minimal environmental impact. These techniques transform organic waste into bio-based products like biofuels, bioplastics, organic acids, enzymes, fertilizers, and animal feed. In contrast to thermochemical processes that operate at high temperatures and generate less selective outputs, biochemical processes function under milder conditions, using microbes and enzymes to achieve targeted conversions. These approaches reduce reliance on landfills and fossil resources while creating high-value products from underutilized biomass. A core aspect of biochemical valorization is the microbial breakdown of complex organic compounds such as carbohydrates, proteins, and fats commonly found in agricultural waste, food residues, and industrial effluents. This is accomplished by natural microbial communities or genetically enhanced strains with tailored metabolic capabilities. Anaerobic digestion exemplifies this process, where organic waste is decomposed without oxygen to yield biogas (mainly methane and carbon dioxide) and nutrient-rich digestate. The biogas can be harnessed for energy generation or as a transport fuel, while the digestate serves as a sustainable fertilizer, thereby supporting circular agricultural practices.

Summary

Idealistic theories of property are projected to offer general and explicit statements of the justification for deciding legal and moral questions about the status property claims. Debates over property rights in biotechnology were occasioned by specific legislative proposals such as the US Animal Patent Act of 1986, and by filing of patent applications for DNA sequences and processes in the early 1990s. While these debates make sporadic petition to philosophical theories of property, moral claims were entwined with questions about filing requirements, tests for worth, and the rules for licensing and defending patents. The comparative scarcity of discussion on intellectual property at that time dictated the general approach of the chapter: Review basic philosophical approaches to property rights, and speculate on how one might use these approaches in constructing an argument pertinent to agricultural biotechnology. This chapter deals with different approaches to intellectual property in the field of Plant Sciences especially the Biotechnology. Origin, History and Definition

The term biotechnology was coined by Karl Ereky, a Hungarain engineer, in 1919. The origin of biotechnology can be traced back to prehistoric time when armers and gardeners have been making use of hybridization and grafting among plants with a view to bring about an improvement in existing attributes. Using these approaches, several hundred varieties in ornamental plants were produced. Microorganisms were already used for processes like fermentation, formation of yoghurt and cheese from milk, vinegar from molasses, production of butanol and acetone from starch by Clostridium acetobutylicum or the production of antibiotics like penicillin from Penicillium notatum. Four defferent phases of biotechnological research were witnessed during the last century. In 1920, Chain Weizmann used Clostridium acetobutylicum to convert starch into butanol and acetone (during world war I, acetone was used as an essential component of explosives). Alexander Fleming in 1929 discovered penicillin from Penicillium notatum and wounded soldiers were given treatment using this antibiotic during world was II. In 1970s with the discovery of restriction enzymes, which led to the development of recombinant DNA, technology brought a revolution in the field of biology. The latest phase in the history of biotechnology is the study of structure and function of whole genome (DNA) and whole proteome (protein). This area of research is described as genomics and proteomics, respectively. Some more important activities in the field of biotechnology are given in Table1

In fact, biotechnology is the product of interaction between science of biology and technology. It is the technological exploitation and control of biological system. Biotechnology has wide range of its uses and accordingly Office of Technology Assessment described biotechnology as: (i) old biotechnology that includes fermentation, i.e. formation of vinegar from molasses and that of cheese and yoghurt from milk, production of antibiotics from certain fungi and process of baking and brewing and (ii) new biotechnology that includes the techniques of recombinant DNA and PCR polymerase chain reaction, cell culture and fusion and bioprocessing which became possible only through the researches in molecular biology. However, no efforts have been made to distinguish between old and new biotechnology. Keeping in view the importance of biotechnology, attempts have been made to define it. Following are some of the definitions of biotechnology, however, none of the definitions of biotechnology seems to be complete. The first definition seems to be comprehensive and wide.

Introduction

As per the reports of the United Nations Organization (UNO), the world population is expected to increase to about 9.7 billion by 2050 and to 10.4 billion by the mid-2080s. The increasing population will result in food scarcity and hunger across the world. To meet the demand for food to feed the people and meet the present aim of WHO of “Zero Hunger by 2030”, agriculture and livestock production needs to be increased. However, according to the ICAR Vision 2050 document, growth of the livestock sector is expected to be higher than the agriculture due to different factors like urbanization and changing food habits among the population residing in the urban area, lack of technology among the small and marginal farmers, reducing operational land, water scarcity, reduced irrigation infrastructure, climate change, etc. to name a few. But to achieve food security through the livestock sector, animal health and production of the animals need to be addressed.

Over the years, global climate change has negatively impacted the resources of western Himalayan region and the diversity of floral wealth has been endangered. The endangered livelihoods of agriculture based sector call for incorporating stress resistance by sustained effort that includes resort to infusion of new technologies. Biotechnology offers possible solutions to many of the intractable problems. Development and commercialization of transgenic crops have witnessed phenomenal growth in recent years. An array of traits viz. insect pest resistance, herbicide tolerance, abiotic stress tolerance, augmentation of nutritional qualities etc. have been targeted and successfully modified through plant genetic engineering. The global area of transgenic crops has seen a tremendous increase to attain 148 million hectares in 2010. Progress in understanding plant genome structure and function by high throughput characterization of genome, transcriptome, proteome and metabolome is providing new opportunities to deal with complex traits that result from interactions of a large number of genes. The other significant application of biotechnology in crop improvement has been “Marker Assisted Selection” (MAS). Development and integration of DNA based markers in the selection process has empowered the breeder to identify the desired genotypes without any interference of environmental effect or tissue specificity of expression. Cultivation of MAS derived crop varieties in the country bears testimony to the success of this technology.

Any technique that uses living organisms or substances from these organisms to make or modify a product or to improve plant or animal or to develop microorganism for specific uses, can be termed biotechnology. Biotechnology involves microbes or cells derived from plants and animals for efficient production of useful products for mankind. The use of microbes for production of alcohol and penicillin, to name a few, is well known. However, in the last 25 years, several novel organisms, yielding useful products have been created by genetic engineering. Today, there is a wide application of biotechnology in human medicine, agriculture and animal husbandry in the area of diagnosis prophylaxis and therapy.

Aquaculture industry has undergone a sea change world over in the last three decades. The important needs of the industry are development of fast growing disease resistant fish and shellfish varieties, development of cheap and effective vaccines, disease diagnostics, cell lines sand probiotics. Efforts on these lines have already begun since 3-4 decades and results are highly promising. Some of the biotechnologies employed in aquaculture health management (Plate 15) are fish cell lines, monoclonal antibodies and nucleic acids for diagnostics sub unit and nucleic acid vaccines for prophylaxis, transgenes for disease resistance, gene editing in fishes and their pathogens and probiotics for bioremedial measures.

A. Cell Lines for Health Management

Cell lines are transformed cells that are adapted well to grow and multiply outside the host in in vitro culture systems. Cells lines can be developed from fast growing tissues of young fish. There are two methods of developing cell lines (1) Explant method, (2) Enzymatic method. Cell lines are grown in sterile plastic or glass containers containing appropriate medium having nutrients salts, antibiotics, buffers, serum etc. Minimum essential medium (MEM) is the common medium employed for fish cell line. Cell lines are developed, characterised and deposited in national and international repositories, which can be procured for use in research and industry.

Biotechnology can be defined as utilization of materials and methods for benefit of mankind by the utilization of microorganisms and biological agents. It is an area with a tremendous potential in solving basic problems of food, fibre, fuel and medicines in developing countries. According to the office of the Technology Assessment of the US Congress, biotechnology can be defined as any technique that uses living organisms to make or modify a product, to improve plants or animals or to develop micro organisms for specific uses. Flower industry has always increasing demand for new forms, colours and types of flowers. Biotechnology has made significant advances in the improvement of ornamental crops and development of new varieties has great potential to be utilized in future. Utilization of this technology can be done in floriculture under two broad subheads viz., tissue culture and genetic engineering.

TISSUE CULTURE

The major commercial utilization of tissue culture has been micropropagation, which is an alternative to vegetative propagation. Micropropagation permits true to type multiplication of a desired genotype using in vitro culture techniques. Micropropagation process can produce a large number of propagules in a relatively short time and space under controlled conditions. It has already revolutionized production process of several nurseries. The dream of multiplying disease free plants in required quantity on the specified time schedule in an industrial way has become true due to micropropagation.
