For a researcher, laboratory is another home where he/she spends much time every day. As we take care of cleanliness, safety, and design of our home, we must take care on these aspects of laboratory too. It is the responsibility of the laboratory personnel to ensure cleanliness/safety of the laboratory. Since every laboratory chemical and equipment have the potential to cause harms if not used with appropriate precautions. Hence, as a laboratory user, it is our responsibility to follow the basic safety/biosafety rules and regulations set up by the Principal Investigator of the laboratory, Institutional Biosafety Committee, as well as by other regulatory authorities, if any. We must be aware of the general/personal protective measures, cleanliness and laboratory waste disposal prior to joining a research laboratory. In this chapter we will briefly discuss about personal safety, chemical safety, cleanliness, waste disposal, etc.

A. Personal Safety

1. Personal Protective Equipment (PPE)

Personal protective equipment (PPE) is special arrangement to protect the worker from specific hazards because of using a hazardous substance. PPE does not reduce or eliminate the hazard, but protects the wearer from exposure to hazardous material. PPE should not be reused or brought outside the laboratory as these might increase the risk of spread hazardous material or contamination. Used PPE must be kept inside the laboratory in a designated waste storage area adjacent to the work area.

Buying an Existing Business

In case of Allahabad, it is evident that a major thrust catapulting the city to an invincible position was experienced immediately after 1857. These changes in the character of Allahabad along its journey which witnessed the resurgence of the pilgrim town into a front ranking educational and cultural node also saw the shades of capital center and finally its transition into a low profile city which today has lost its vitality and seem to be comparable to a non functioning muffossil town of Uttar Pradesh. The facts and figures representing the key events and a survey of the city enable us to establish a direct relationship between the functions and performance of the city.

The first part of the chapter is based on the inferences from the study on Allahabad covered earlier. The second part deals with testing these facts on ground by conducting interviews with eminent individuals who had been deeply associated with education and cultural status of Allahabad. The third part consists of the final inferences. Some critical conclusions are also borne out reflecting a clearer understanding of the phenomenon of decline of Allahabad.

Molecular markers may be a short DNA sequence that flanks base pairs like SNP, a Single Nucleotude Polymorphism, small fragment or SSR Single Sequence Repeat or an extended site as in the case of microsatellites. Schematic representation of the SSR assay is shown in Fig. 1. Genetic markers have to be easily identified, linked to a specific locus and polymorphic. Molecular markers are subdivided into codominant monolocus and polyallelic markers and the second group contains dominant, polyallelic and biallelic markers. 

Introduction

Resistance to antibiotics in the bacterial world is a genetically determined phenomenon that was first observed within a few years after the introduction of antibiotics for therapeutic purposes. In Nature, ARGs are believed to have originated about three billion years ago as an intrinsic defense mechanism in antibiotic-producing bacteria (Perry et al., 2016; Iskandar et al., 2022), and subsequently, evolved by complex and quantum jump trajectories to mobilize, diversify and interconnect all ecosystems in the micro-biosphere. Bacterial genomes exhibit plasticity and continue to evolve under the selection pressure of antibiotics and other stressors (Patel, 2016). The ARGs in circulation at present are thus a mixture of ancient and modern counterparts. Recently, the interplay between bacterial metabolism and antibiotic resistance has also been revealed (Zampieri, 2021).

The use of antimicrobials in health management, livestock, and food industry is driving the emergence of resistant microbes at a rapid pace (Wall et al., 2016; Adegoke et al., 2016; Hu et al., 2017). As a result, AR pathogens have spread globally to cause loss of lives, production, productivity, and negatively impact economic growth. Recently, AR pathogens in a large study have been found directly responsible for millions of deaths, and the figures are likely to increase in the coming decades (Murray et al., 2022). The pace of the emergence of AR can however be slowed down by improving human interventions. The global and local AR combat plans implemented during the past few years have enhanced the mass awareness of the pandemic problem (WHO, 2015; FAO, 2016; NAP, GOI, 2017; WHO, 2022; WOAH, 2022). These Action Plans on AR containment encourage avoiding of indiscriminate use of antibiotics for the treatment of sick humans and animals, recommend the ban on the use of antibiotics as growth promoters in food animals, promote AR surveillance and monitoring, and support the ‘One-Health’ strategy and application of the biosecurity principles for containment of the resistant microbes.

History of Agriculture
Cultivation of crops is one of the oldest occupations of mankind and probably began when it was discovered that certain seeds spilled on disturbed ground, grew in some mysterious way into new plants. The seeds were then collected and spread into different regions. This is how origin and introduction of crop plants started. Development of agriculture about 12,000 to13,000 years ago changed the way humans lived. They switched from nomadic hunter-gatherer lifestyles to permanent settlements and farming. However, scientists are unable to explain why man took so long to discover agriculture. Agriculture began independently in different parts of the globe and included a diverse range of taxa. At least eleven separate regions of the Old and New world were involved as independent centers of origin however, there was no single factor or combination of factors that led people to try farming in different parts of the world. Farming could have been brought on by climate changes at the end of the last ice age that favoured different annual plants like wild cereals to grow. But whatever the reasons for its independent origins, farming sowed the seeds for the modern age. Archeological evidence for the origin of crops points to mountainous areas of both Old World (Asia, Europe and Africa) and New World (North and South America), which have pronounced wet and dry seasons. Under such climatic conditions seeds would need to germinate, grow quickly with the onset of rains, complete their reproductive cycles and go to seed formation before summer sets in such as we see in annual plants.

Animals are exhibited in various ways and events like in fairs, judging shows, in circuses, in zoos and as performing animals. Some famous animal fairs in northern part of the country are Sonepur cattle fair of Bihar. It is the largest cattle fair of Asia, held on the confluence of holy river Ganges and Gandak annually on KartikPoornima in the month of November for the trade of animals such as Cattle, horses,goats,donkeys,camels,birds and elephants. Haathi Bazaar is the major attraction of SonepurMela. Nagaur cattle fair held during the months of January and February. It is the second biggest cattle fair in India. Pushkar Cattle Fair,popularly known as camel fair held for five-days between the months of October and November. Cattle shows are organized time to time where animals are judged for their different characteristics, selecting for their best type, possessing true breed characteristics, dairy conformation, good temperament, proper growth and development according to its age, with optimum condition of fleshing and free from defects. To take part in such shows animals are transported to different places. So, apart from knowledge of restraining methods, concept related to ageing, animal handling (flight zone, point of balance) animal behavioural consideration and welfare indicators are of much importance for transporting and presenting animals in any show. This chapter explains processing operations, quality preservation and the commercial importance of value-added onion products. It also addresses the large quantities of peels, outer scales and trimmings generated during processing. These residues contain quercetin, polyphenols, dietary fibre and other useful compounds that can be recovered for functional foods, nutraceuticals, natural antioxidants, biofuels and biodegradable materials, supporting waste reduction and a circular bioeconomy.

Abstract

Rice being the major food crop in the world requires a serious attention to protect the crop from variable pest attack under change in climatic scenario. More then 100 species of insects have been recored as rice pest affecting the productivity as well as maintaining sustainability in rice production. Bio-control agents for the control of rice pests are to be systematically evaluted. More than 1500 species of pathogens are formulated as microbiological insecticides. So our current awareness in pest- pathogen-abiotic interactions should be made use in designing the crop protection strategy.

Abstract

Women Technology Park is a technology modulation and a training centre to show case livelihood technologies for women. The aim of Women Technology Park is to create awareness among rural farming community and give trainings of proven technology to reap maximum benefits from their existing resources. Its objective is to make them aware about “processing” that can enhance their income in multiple times. In this direction, the first ever Women Technology Park (WTP) of Jammu and Kashmir was sponsored by SEED division of Department of Science and Technology, Government of India and endorsed by Livestock Products Division FVSc, SKUAST-Jammu. It was inaugurated by Vice chancellor of SKUAST- Jammu at Deoli Village of Bishnah Block, Jammu and Kashmir. 12 groups of women were screened out and trainings were imparted to them for a week. More than 75 training cum awareness programs have been organized with more than 1500 beneficiaries. There are more than 50 beneficiaries who received start up grant in the form of goods essential for preparing livestock products and started up their own micro-enterprises and became self-reliant and set an example for other women to come up and take up processing of livestock products as profession to earn their livelihood.

Livestock plays a pivotal role in sustaining the livelihood of millions of people worldwide, particularly in rural and semi-urban communities. Beyond providing food products such as milk, meat and eggs, livestock also contribute to income generation, draught power, manure for agriculture and socio-cultural functions in many regions. The health and productive performance of livestock animals are central to achieving the economic, nutritional and ecological goals of animal husbandry. Any compromise in animal health or productivity directly impacts farmers’ income, food security, and national agricultural output. Animal health is the foundation of productive and sustainable livestock farming. Good health ensures animals can grow, reproduce and perform at their genetic potential. Conversely, disease, poor nutrition and environmental stress can severely reduce productivity and increase mortality. Common health challenges in livestock include infectious diseases (bacterial, viral, and parasitic), metabolic disorders and management- related stress conditions. Moreover, zoonotic diseases—those transmissible to humans— are of growing concern in the context of public health and food

Rajasthan is the largest state covering about 34.2 million hectare geographical area of the country, of which 27.1 million ha (79 %) is under arid and semi-arid sub-tropic hot climate and spread over 21 districts. The land-scape of Aravali range provides boundary for arid western, semi-arid eastern and sub-humid to humid areas of the state. In-spite of several environmental restrictions and bio-physical constraints in most of the parts, the state can be classified into three distinct agro-climatic region i.e. hot arid, semi-arid and sum-humid from horticultural perspective. On the basis of research experiences and SWOT analysis over last two decades for R&D road-map of Rajasthan Horticulture, the classified zones have excellent vegetable development potential with wide and varied opportunities of crops and utilization of resources.

Abstract

The conventional methods of pathogen identification have often depended on identification of disease symptoms, isolation and culturing of the organisms, and identification by morphology and biochemical tests. The major limitations of these culture based morphological approaches, however, are the reliance on the ability of the organism to be cultured, the time consuming nature and requirement of extensive taxonomic expertise. Furthermore, diagnosis of plant diseases can be even more difficult with asymptomatically infected propagative materials such as tree grafting stocks or potato tubers. The use of molecular methods can circumvent many of these shortcomings. Accordingly, there have been significant developments in the area of molecular detection of plant pathogens in the last three decades. The polymerase chain reaction (PCR) which revolutionised molecular diagnostics and biological sciences. In the last decade the range of targets that can be diagnosed using diagnostic PCR have grown tremendously. The very flexibility and application specific variations in the basic theme of the system have allowed the development of many PCR variants adapted to wide range of applications. Furthermore, diagnostic PCR has been greatly improved by the introduction of the second generation PCR, known as the Real time PCR where closed-tube fluorescence detection and quantification during PCR amplification (in real time) is possible eliminating the need for laborious post-PCR sample processing steps which greatly reduces the risk of carryover contamination. Using Real Time PCR, it is possible not only to detect the presence or absence ofthe target pathogen, but it is also possible to quantify the amount present in the sample allowing the quantitative assessment of the number of the pathogen in the sample. Enumerating the pathogen upon detection is crucial to estimate the potential risks with respect to disease development and provides a useful basis for diseases management decisions. Crops can be attacked by many pathogens which, in addition, often occur in complexes. Therefore, many disease diagnostic applications require simultaneous detection and quantification of several targets. Methodological limitations, however, are in many cases the reason for developing simplex or assays designed for only few targets. The DNA Microarray technology, originally designed to study gene expression and generate single nucleotide polymorphism (SNP) profiles, is currently a new and emerging pathogen diagnostic technology, which in theory, offers a platform for unlimited multiplexing capability. It is viewed as a technology that fundamentally alters molecular diagnostics. The fast growing databases generated by genomics and biosystematics research provides unique opportunity for the design of more versatile, high-throughput, sensitive and specific molecular assays which will address the major limitations of the current technologies and benefit plant pathology.

Different crops are grown over a wide range of agroecosystems. Though the crops are raised in varied climates, ranging from very hot to very cold, and very wet to very dry, across temperate, tropical and semiarid zones, they are very sensitive to climate. The crop production is dependent on climatic parameters like rainfall, light, temperature, relative humidity and carbon dioxide concentration. Each kind of cultivated crop plants has its own optimum growth requirements. Breeding and selection of new cultivars have allowed for a greater adaptability to less favourable growing conditions than was possible in the past, but the inherent climatic requirements of a specific kind of crop have not changed materially. Crop management is therefore very much about managing climate risk so as to have financially viable and sustainable agricultural systems. Professor Henry Nix of the Australian National University posed three questions that need to be addressed when considering what crop can be grown where (Nix, 1981).

Introduction

Most glands of the body deliver their secretions by means of ducts. These are called exocrine glands. There are few glands that produce chemical substance that they directly secrete into the blood stream for transmission to various target tissues. These are ductless or endocrine glands. The secretions of endocrine glands are called as hormones. Therefore, it is a chemical substance which is produced in one part of the body, enters the circulation and is carried to distant target organs and tissues to modify their structures and functions. Hormones are stimulating substances and act as body catalysts. The hormones catalyze and control diverse metabolic processes. Despite their varying actions and different specificity’s depending on the target organ, the hormones have several characteristics in common with enzymes. They act as body catalysts resembling enzymes in some aspect. They are required only in small quantities. They are not used up during the reaction. They differ from enzymes because they are produced in an organ other than that in which they ultimately perform their action. The major hormone secreting glands are as follows— Pituitary, Thyroid, Parathyroid, Adrenal, Pancreas, Ovaries and Testes. The hormones are classified into following main classes according to the chemical structures:

Male-sterility
Normal development of the male reproductive organ (stamen) and male gametophyte (pollen grain) is essential for successful completion of sexual reproduction in angiosperms. Abnormalities at any stage of stamen and pollen development can result in male sterility. Male sterility in plants implies the inability to produce or to release functional pollen. Male sterility is caused due to failure in the formation or development of functional stamens, microspores or gametes and /or release of pollens from the anther. Male-sterility can result from mutations in nuclear genes alone or mitochondrial genes or mitochondrial genes coupled with nuclear genes. The resulting conditions are known as genic male sterility (GMS), cytoplasmic male sterility (CMS) and cytoplasmic-genic male sterility (CGMS), respectively.
Classification of male-sterility
1. Genetic male-sterility
This male-sterility is caused due to mutations in nuclear genes alone or mitochondrial genes or mitochondrial genes coupled with nuclear genes which results abnormalities at any stage of stamen and pollen development.

Abnormal combustion: Combustion that does not proceed in the normal way, in which knock, pre-ignition, run-on or surface ignition occurs. Refer knock.

Abrasion: It is the surface discontinuity brought to any solid by the process of removal of surface material through friction of another solid, liquid or gas or combined thereof.

Absolute or specific humidity: It is the mass of water vapour present in a unit mass of dry air. It is also called as humidity ratio.

Absolute instrument: It is an instrument, which measures a quantity (such as pressure, temperature etc.) in absolute units by means of simple physical measurements on the instrument.

Absolute pressure: It is the actual pressure at a given position and it is measured relative to absolute vacuum (i.e., absolute zero pressure). Absolute pressure = pressure shown on the pressure gauge + atmospheric pressure. At sea level, atmospheric pressure is 14.7 psi (1.033kg/cm²).
