Diagnostic Symptoms

• Dark water soaked large patches appear on the basal portions of the stem near the ground level.

This is a procedure for transforming bacterial cells with circular DNA plasmids containing an origin of autonomous DNA replication and a selective marker (antibiotic resistance). Logarithmically growing cells are made permeable to DNA by incubation in CaCl2 and the DNA is taken up from the surrounding buffer.

Nitrogen, a key component of organic materials like proteins, is analyzed using the Kjeldahl method, which is the global standard for determining protein content in various substances, including human and animal foods, fertilizers, wastewater, and fossil fuels. The Kjeldahl method involves three main steps:

B. Distillation

C. Titration

Nitrogen, making up 1-4% of a plant’s dry weight, is vital for synthesizing chlorophyll, proteins, and other key compounds. Plants with high nitrogen levels turn dark green due to increased chlorophyll. Understanding nitrogen content in plant tissues is crucial for diagnosing deficiencies or toxicity and for effective nutrient management to improve crop production.

Determining the nitrogen content in plants is crucial for understanding plant nutrition, assessing the efficiency of fertilizer use, and making informed decisions in agricultural practices. Various methods can be used to determine nitrogen in plant tissues, with the most common ones being the Kjeldahl method, the Dumas method, and near-infrared reflectance spectroscopy (NIRS).

Introduction
Geranium is also known as cranesbill, scented geranium/rose geranium. It is adaptable over a wide range of soil conditions and drought tolerant. Being perennial aromatic herb, it is cultivated for rose scented essential oil extracted from leaves and tender shoots. The oil thus obtained is also called pelargonium oil or rose-geranium oil. The oil is of varied colour from colourless to pale yellowbrown or greenish. The genus Pelargonium belongs to the family Geraniaceae.
Origin
Geranium is native to sub-tropical southern Africa, and the most popular of bedding and green-house plants. The genus Pelargonium consists of about 600 species, which are annual, biennial, and perennials found in Cape province of South Africa.
Botany
The cultivated geraniums may belong to herbaceous to woody with thick fleshy leaves of round to lobed shape and deep cut. The flowers are present in terminal clusters with varying colours from white to pink, deep red and violet. Cultivated species are hybrids, having double flowers of mixed colours.

Dehydration is a critical condition in animals characterized by an imbalance between fluid intake and loss, leading to a deficit in total body water. It is a common problem across all species and can result from a variety of causes, including disease, environmental factors, and inadequate water intake. Understanding the causes, clinical signs, diagnosis, and treatment of dehydration is vital for ensuring the health and well-being of animals.
Physiology of fluid balance
Water is essential for physiological processes, including nutrient transport, waste elimination, temperature regulation, and cellular metabolism. Animals maintain fluid balance through mechanisms that regulate intake (via drinking and food consumption) and output (via urine, feces, respiration, and sweat in some species). Dehydration occurs when water loss exceeds water intake. Body’s water compartments are divided into Intracellular fluid (ICF): About two-thirds of total body water: One-third of total body water, including plasma and interstitial fluid. Dehydration affects these compartments differently based on severity and cause, potentially leading to hypovolemia (decreased blood volume) and compromised organ function.
Causes of dehydration
Insufficient water intake
Neglect or inaccessibility: Lack of access to clean, fresh water. Behavioural issues: Reluctance to drink due to stress or environmental changes. Illness: Conditions like oral pain or esophageal obstruction can reduce water consumption.

Botany :The cabbage plant, Brassica oleracea, is an herbaceous annual or biennial vegetable in the family Cruciferae grown for its edible head. The head of the cabbage is round and forms on a short thick stem. The leaves are thick and alternating with wavy or lobed edges and the roots are are fibrous and shallow. The plant produces large yellow flowers. The densely leaved heads can range in size from 0.5 to 3.6 kg (1-8 lb) depending on variety. The plant is usually grown as an annual.

In 1907, the Russian Nobel laureate, Dr. Elie Metchnikoff, fascinated by the unusual longevity of Bulgarian farmers, consuming beneficial lactic acid bacteria by way of traditional fermented milk products, suggested that regular intake of friendly microbes could benefit human health. Although Dr. Metchnikoff ’s recommended bacteria for the purpose proved largely ineffective, the theory was carried forward with moderate success until the war on bacteria began because of the advent of sulfa drugs and antibiotics (Scott, 2012). However soon after their rampant use and the realization of the rapid spread of antimicrobial resistance, researchers turned yet again to ecological notions and the time was fertile for the advent of probiotics. By the 1980’s the term had stabilized both in medical and veterinary literature. The mid 1990’s witnessed a surge of exponential studies on probiotics, the definition of Probiotics emerged in 2001 and the years thereafter saw tremendous progress in the science of probiotics.

Almost simultaneous was the study on the study of the intestinal microbiota when Alfred Nissile demonstrated that transferring members of the human gut microbiota to healthy typhoid carriers helped in Salmonella being cleansed from their system (Nissle, 1916). It was then, that the importance of the gut microbes in prevention of infection became evident. What followed was the advent of culture independent, modern biotechnological tools for whole genome sequencing and the successful launch of two most ambitious projects - Human Microbiome project (Turnbaugh et al., 2007) and the Met Hit project (Metahit, 2009; Qin et al., 2010). They provided fascinating insights into the highly complex microbial ecosystem harboured by the human body. The term Microbiome (refers to the collective genome of our microbial inhabitants) had entered literature in 2001 and the concept of gut microbiota in maintaining overall health gained wider attention.

Introduction

Nanocomposites are materials that incorporate nanosized particles into a standard material matrix. The addition of nanoparticles can significantly improve properties such as mechanical strength, toughness, electrical or thermal conductivity. Typically, only a small amount of material (0.5-5% by weight) is required due to the effectiveness of the nanoparticles. A nanocomposite is a solid material that consists of multiple phases, where at least one of the phases has one, two, or three dimensions in the nanometer size range. The goal of the nanoscale phase process is to achieve synergy between different constituents. Nanocomposites represent a promising alternative to overcome current limitations of microcomposites and monolithics, and are becoming the materials of the future (Schmidt et al., 2002).

Asiatic Elephant (Elephas maximus) is the national heritage animal of India. This particular species now exists in 13 Asian countries including India. As per 2017 census, India is home to an estimated figure of 29,964 elephants that is >60% wild population of the globe. Once distributed across the country, now they are squeezed to 14 states grouped in four regions ie., southern (14,612), north-eastern (10,139), east-central (3,128) and northern regions (2,085). There are 2675 captive elephants across different states under both private and government custody. This largest terrestrial mammal is protected under Seclude I species of Indian Wild Life Protection Act, 1972.

Both central and state Government are implementing short- and long-term programmes for health and well-being of captive as well as free-range elephants. The Central Government has established an ‘Elephant Cell’ at the Wildlife Institute of India (WII), Dehradun to provide technical inputs to the ministry for strengthening the efforts towards conservation of elephants. Add-on, the ‘Project Elephant’ was launched in 1991-92 in the Ministry of Environment, Forests and Climate Change to restore the elephant habitats and reduce their sufferings. Elephant habitats are notified as Elephant Reserves that are basic management units for elephant conservation and managements. A total of 33 Elephant Reserves has been notified in the country. A book titled, ‘Caring for elephants: Managing health and welfare in captivity’ is edited for field officers, veterinary professionals and elephant handlers.

Sample Digestion

Dry ashing and wet oxidation are two widely adopted methods for releasing mineral elements from plant tissues. Dry ashing, typically conducted at temperatures between 550 and 600 °C, can lead to the loss of phosphorus (P) and potassium (K) through volatilization. This method is more time consuming and has several drawbacks, such as the volatilization of sulfur (S) and chlorine (Cl), which can be mitigated by adding sodium carbonate (Na2 CO3 ). Additionally, some phosphorus and micronutrients may become occluded during the process, making dry ashing less commonly used.

In contrast, wet oxidation employs oxidizing acids, such as a mixture of nitric acid and perchloric acid (HNO3-HClO4 ) or a tri-acid mixture of nitric acid, sulfuric acid, and perchloric acid (HNO3-H2 SO4-HClO4 ). The use of perchloric acid (HClO4 ) prevents the volatilization loss of potassium (K) and provides a clear solution, while sulfuric acid (H2 SO4 ) aids in completing the oxidation. Upon heating, perchloric acid dissociates into nascent chlorine and oxygen, enhancing the oxidation efficiency at high temperatures. However, direct contact between perchloric acid and plant samples can lead to explosions and f ires; thus, pre-digestion of samples is preferred.

There are thousands of diseases, which attack crop plants. Classification can be made based on several criteria. The various ways of classifying diseases of plants are given below.
A. Based on type of infection
1. Localized disease: Affecting only a part of the plant; leaf spots and anthracnoses caused by different fungi.
2. Systemic disease: affecting the entire plant.
B. Based on symptoms: Rusts smuts, wilts, blights, cankers, mildews, rots, damping-off, die-back, scab etc.
C. Based on the host plant
1. On the basis of host e.g. Diseases of apple, diseases of wheat, diseases of rose, diseases of coconut, diseases of coffee, diseases of cotton.
2. On the basis of host group- e.g. cereal crop disease, pulse crop diseases, oilseed crop diseases, root crop disease, forage crop disease, plantation crop disease etc.

Introduction
Genus Curcuma belongs to the Zingiberaceae family, and containing of about 122 species. These are grown profusely in Asian countries including, India, China, and Southeast Asia, Northern Australia and New Guinea. Like ginger, it is also the popular herb of this family for the utilization of its underground rhizomatous stem by human beings. Botanically it is named as Curcuma longa L. Its bright yellow spicy powder is got from the dried rhizome is termed turmeric powder, which is the final product of this herb. In addition to this curcumin bio-active compound is also extracted by cursing rhizomes of turmeric. The plants turmeric are considered to be a hybrid and sterile leaving vegetative propagation from rhizome as the single mode of propagation. India is recognized as the power house of turmeric production. Along with us, our neighbours in South East and Eastern region, including Malaysia, Thailand, Bangladesh, Cambodia, Philippines, Indonesia and even China are considered major producers.
Floral Biology
All species of Curcuma genus have almost alike flowering spikes that mainly originate from the topmost region of the rhizome (pseudo-stem) or sometimes on a separate stem directly from the rhizome. These two stems differ onlyin the inner portion of rhizome, which differ in colour, i.e., creamy, white, orange, yellow, bluish-green, blue, and sometime black also. The rhizomes exhibit commercial value as a medicine as well as food spice. Curcuma longa L. (Syn. Curcuma domestica Val.), is the common turmeric. It is the top ranking herb based on its economic potential among all the members of this genus and cultivated in India about 150,000 hectares.

Hemorrhagic disorders refer to conditions that impair the ability of an animal’s blood to clot properly, resulting in prolonged or excessive bleeding. These disorders can be broadly categorized into congenital coagulation defects and acquired coagulation defects, each with distinct etiologies, pathophysiologies, and clinical manifestations.
a) Congenital coagulation defects
Congenital coagulation defects are inherited conditions caused by genetic mutations affecting one or more components of the coagulation cascade. These defects are often breed-specific and can be observed in a variety of domestic animals.
Common types of congenital coagulation defects
Hemophilia- A
Cause: Deficiency of factor VIII.
Affected animals: Common in dogs, especially German Shepherds, and less common in cats and horses,
Pathophysiology: Factor VIII is essential for forming the intrinsic tenase complex, which amplifies thrombin generation. Its deficiency results in inadequate fibrin formation and poor clot stability.
Clinical signs: Spontaneous bleeding, hematomas, hemarthrosis (bleeding into joints), and prolonged bleeding after trauma or surgery.
Diagnosis: Prolonged activated partial thromboplastin time (aPTT), normal prothrombin time (PT), and reduced Factor VIII activity.

There are many benefits to having journals added to the Scopus database. In addition to its extensive content coverage encompassing virtually all subjects, Scopus is known for its high level of academic rigor, including an annual journal re-evaluation process, which it requires as “an incentive for journals to maintain their high content quality.”
Scopus also provides a wide range of metrics for indexed content, including
• Journal-level metrics such as Citescore, which both publishes an annual impact score and gives monthly updates on how things are currently shaping up for each of the indexed titles via the CiteScore Tracker
• Article-level metrics such as PlumX, which analyzes how readers are responding to content in the broader online context (i.e., via tweets, blogs, Wikipedia references, etc.)
• Author-level metrics such as the h-index, which aims to provide an objective measure of the overall impact of individual authors’ publication histories.

Tropical plum (Spondias mombin) also known as yellow mombin, hog plum, and java plum, is a deciduous fruit species widely spread in the tropics. This species is evaluated for its fruit as an unconventional source of vitamins A and C, which are used for making juices, ice creams and jellies. This plant also has medical importance.
 
It is a graceful tree of the family Anacardiaceae, a native of tropical America. It was introduced into India for edible fruit and also to serve as fencing. The fruit flesh (48% on  weight of fruit) contains: total solids 11.47%, protein 1.38%, fat 0.57%, acids (astartaric) 1.50%, reducing sugars 9.41%, sucrose nil, fibre 1.17%, and mineral matter 0.66%. The seeds are eaten when tender. The fruit is a good source of minerals and vitamin B1 and C.

The Metric System

At present metric system is followed to calculate final results in analysis work. This system is quite simple in nature as well as in principle. The units of the metric system are: (1) the  metre (linear measure), (2) the litre (measure of capacity), and (3) the gram (measure of mass.)

One litre is the volume of pure water at 4ºC and 760 mm pressure which weigh 1 kilogram (kg). One litre equals 1000.027 cubic centimetres (Cm3) for most work, the units “millilitre”, “cubic centimetre” and used interchangeably.

At present there are three different temperature scales in common use. These are: the Fahrenheit (F), used chiefly in English-speaking countries. the centigrade (C), used universally in scientific work and the Absolute (T) or Kelvin (K) scale, differing from the centigrade only in the position of the zero point, its divisions being of the same size. its zero point is -273ºC. The kelvin scale is used especially in dealing with very high or very low temperature and in computations involving the laws of gases. To know the method of immobilization of the limb after repair of the transected gastrocnemius tendon (tendo achiles).

Procedure

The immobilization procedure of limb after gastrocnemius tendon repair is done in 2 stages.

1^ST Stage

Immobilization of limb for avoiding movement at tendon suture site.

It is done by placing the sutured area within a pre- fabricated metal frame so as to keep the limb undisturbed at the suture site.

The frame is given in Figure 1.

The purpose of addition of the metal frame is to immobilize the limb keeping the fetlock and foot away from in a flexed manner to avoid weight bearing during healing period.

The limb is immobilized with netted bamboo splint (Fig. 2).

An understanding of normal values of electrocardiographic indices of healthy dogs and cats is utmost necessary for interpreting an abnormal electrocardiogram. The criteria for normal configuration and measurements of the waves, segments and intervals in an electrocardiogram of healthy subjects provides basis for detecting abnormalities in the electrocardiogram of the ailing animals. The electrocardiogram, like other diagnostic aids, is not an absolute indicator of health and disease and therefore it is required to be interpreted in conjunction with patient’s history, clinical manifestations, and the results of other investigations.
