Soil and growing media provide the physical, chemical, and biological foundation for successful horticultural production. This chapter explains soil properties, soil fertility, soil health, growing media, nutrient availability, and principles of plant nutrition. Emphasis is placed on soil testing, pH, electrical conductivity, organic matter, nutrient balance, and crop-specific fertilizer management. The chapter also introduces soilless media used in nurseries and protected cultivation. Understanding root-zone conditions is essential because horticultural crops are highly sensitive to water, aeration, salinity, and nutrient imbalance. Sustainable approaches such as composting, mulching, biofertilizers, and integrated nutrient management are discussed to maintain productivity while improving soil health and resource-use efficiency.

LAND VERSUS SOIL

Land is a part of Earth’s surface that is not covered by water. It is used for agriculture, building houses, of?ce buildings, schools, hospitals, roads, railroads and even water canals. Land is measured by area [length (l) × width (w)] and the units are square meters (m²), square centimeters (cm²) etc. On the other hand, soil is Earth’s crust, which could vary from a few centimeters to a few meters in thickness (depth), needs a measure of volume [length (l) × width (w) × depth (d)] and the units are cubic meters (m³), cubic centimeters (cm³) etc. Soil is thus a three-dimensional natural body on Earth’s surface that provides anchorage to growing plants/ trees and shelter to soil biota and is responsible for the storage of water and plant nutrients, which are essential for crop production. In addition, it helps in the biodegradation of soil pollutants and puri?cation of ground water, the major source of drinking water in the world. Soil feeds all living beings on Earth and its degradation is one of the reasons for inadequate human nutrition

26.1 Plant Available Fraction of Nickel, Cadmium, Lead and Chromium

The DTPA method was originally developed to extract Mn, Cu, Fe and Zn from slightly acidic to alkaline soils (Lindsay and Norvell, 1978), but in some cases it can be used for other metals also, such as Nickel (Ni), Cadmium (Cd), Lead (Pb) and Chromium (Cr). Among them Ni is considered as an essential micronutrient. The DTPA is used to extract soil metals by forming chelates with free metals in solution, which lowers their ionic activities so that additional quantities of metal are released from the soil colloids until equilibrium is attained. Triehanolamine (TEA) is used to buffer the extracting solution at pH 7.3 in an attempt to optimize the extraction of various metals. Calcium chloride is included in the extracting solution to minimize dissolution of CaCO₃.

Interaction of radiant energy, exhibiting the properties of both waves and particles, with matter is dealt in spectroscopy. Matter may be defined as material, existing in solid or liquid or gaseous phase, which is composed of molecules, atoms, or ions. Atoms, molecules, and ions, irrespective of their physical state, remain in constant motion in a matter. Molecules may rotate, vibrate, and move from place to place in space. While radiant energy interacts with matter, these molecular motions are influenced to some extent. It is observed that radiations from ultraviolet and visible range of electromagnetic spectrum lead to the movement of bonding electrons from lowest energy state (ground state) to higher energy state (excited state) in the molecules, while absorption of IR radiation causes the molecule to vibrate more. Such changes, referred to as transition, are responsible for changing the molecular energy level.

Spider lily (Hymenocallis littoralis) is a perennial bulbous plant distinguished by its fragrant white flowers, central corona and spider-like appearance. It is an important commercial loose flower crop in western India, particularly Gujarat, Maharashtra and Karnataka, where flowers are used for garlands, Veni, Gajra and ceremonial decoration. The chapter describes its origin, distribution, genetics and botanical characteristics. Split Plot Design

The split-plot design is a statistical approach widely used in experiments where specific conditions apply:

• Hierarchical arrangement of treatments: This design is ideal for experiments involving two levels of experimental units, typically “whole plots” and “subplots.” The main treatments are applied to the whole plots, while the sub-treatments are applied within these to the subplots.

• Adherence to experimental design principles: The split-plot design follows the principles of randomization, replication, and local control, but with a hierarchical twist. The main treatments are randomized across the whole plots, and the sub-treatments are randomized within these main plots. • Resource management in unequal experimental conditions: This design is particularly useful when some factors require larger experimental units or when changing levels of one factor is more resource-intensive than changing levels of another factor.

1. Introduction

Biological systematics (biosystematics) deals with the grouping of biological organisms in discrete taxonomic units based on their character states. These character states are determined through morphological, anatomical, ecological, biochemical and molecular biological analyses of specific parameters. If these character states are converted into numerical parameters, we can apply several statistical and numerical methods to achieve appropriate biosystematic inferences. In fact this particular application of statistical and numerical methods has given rise to what we may describe as Numerical Taxonomy, a concept first developed by Robert R. Sokal and Peter H. A. Sneath nearly a half century ago. In recent years the application of numerical methods to taxonomy has received a great deal of attention with the development of the computer and computer technology to the extent that even classical taxonomists are now resorting to the use of such numerical taxonomy methods with the help of computers. In this article the various methods commonly used, their application and specific limitations or otherwise are discussed in the context of their usefulness for classification and taxonomic disposition of the plants.

Sterols are derivatives of a complex ring system called cyclopentano perhydro phenanthrene. The term sterol means solid alcohol. Sterols, though alcohols, are solids with a M.P. of 100-120^ºC. The 27 carbon atoms impart hydrocarbon character, namely, solubility in non-polar solvents. Free sterols in plants are mono hydroxy  secondary alcohols varying in the constitution of methyl or ethyl group and in double bonds. Cholesterol, ergosterol, bile acids, sex harmones, cortical harmones and D-vitamins are some of the sterol derivatives.
   
The most widely distributed sterols in higher plants are the C₂₉ sterols, stigmasterol, β sitosterol and g sitosterol. Together, they are called steroids. In plants, steroids are present as glycosides. Cholesterol was first isolated from gall stones. The word cholesterol means “solid alcohol from bile”. Bile is a fluid which is formed and stored in the gall bladder. It aids in the digestion of fats in the digestive tract. Sterols form an important group among the steroids. Unsaturated steroids with most of the skeleton of cholestane containing a 3β-hydroxyl group and an aliphatic side chain of 8 or more carbon atoms attached to position 17 form the group of sterols.

Introduction

There are a number of chemicals in the environment, some natural other man-made, some toxic other non-toxic. These are toxic chemicals, whether natural or man-made, which has created problems for the environment and biota on the earth. Of these toxic chemicals, heavy metals are extremely hazardous. Metals are present naturally in the earth’s crust at various levels. However, increasingly widespread heavy metal pollution resulting from anthropogenic activities has caused serious environmental problems and is posing threat to both wild life and human population (Nriagu and Pacyna, 1988). Natural sources include weathering of mineral and metal ions from rocks, displacement of certain contaminants from groundwater or subsurface layers of soil, atmospheric deposition from volcanic activity, and transport of dust. The anthropogenic sources include deposition of industrial effluents, application of sewage sludge, deposition of air-borne industrial wastes, military operations, mining, landfill operations, use of agricultural chemicals, gas exhausts and energy and fuel production (McIntyre, 2003).

The concentration of a true solution refers to the amount of solute present in a given quantity of solvent or solution. It is an important property because it determines the solution’s behavior in chemical reactions, its physical proper ties (such as boiling and freezing points), and its utility in various applications. Concentration can be expressed in several ways, depending on the context and the precision required.

Methods to Express Concentration of a True Solution

1. Molarity (M)

• Definition: Molarity is the number of moles of solute dissolved in one liter (1,000 mL) of solution.

Urban Air Pollution and Its Impact on Roadside Plants
Urban Air Pollution

Modern industrial society produces a large number of gases and particles in quantities to harm human, plant or animal life. The amount of pollutants released to the atmosphere by fixed or mobile anthropogenic sources is generally associated with the levels of economic activity. Deterioration of air quality is a major environmental problem in many urban centers in both developed and developing countries. Automobiles constitute a major source of air pollution in the urban areas. Motor vehicle emissions result from fuel combustion or evaporation. The most common types of transport fuel are gasoline and diesel. CO₂ and water vapors are the main products of the combustion, emitted in vehicle exhaust. The major pollutants emitted from the vehicles are particulate matter (suspended fine and ultra fine), sulphur dioxide, oxides of nitrogen, carbon monoxide and metals.

Sulphur (S) occurs in soil in both organic and inorganic forms. Inorganic S fraction may occurs as sulphate (SO₄^2-) and compounds of lower oxidation state such as sulphide (S^2-), polysulphide (S_n^2-), thiosulphate (S₂O₃^2-) and elemental sulphur (S⁰). In well drained soils inorganic S usually present as sulphate. Under anaerobic condition (reduced or waterlogged soil) inorganic S mainly occur as sulphide and under extreme condition as elemental S form. Estimation of S is carried out in two steps:

1. Extraction of different forms of S from soil by suitable extractant.
2. The filtrate is then analysed for S by the turbidimetric method.

The turbidimetric method is the most widely used one because of its simplicity and rapidity.

4.1 Determination of Specific Surface Area of Soil

Specific surface area of a soil is defined as surface area per unit weight of soil and is expressed as m² g^-1. The specific surface of soil greatly influences the physical and chemical properties such as retention of water at high suction, swelling, plasticity, soil strength, cation exchange capacity and availability of nutrients. The basic principle of determination of specific surface is based on the amount of gas or polar liquid required to form a monomolecular layer on the soild. Knowledge of molecular size (diameter) and the mass of the adsorbate enables one to calculate specific surface of the adsorbent.

Principle

Dyal and Hendricks (1950) introduced a method for estimating specific area based upon the adsorption of ethylene glycol (EG) to form a monomolecular layer over the entire soil surface. The method involves adding EG to pretreated soil or clay sample and evaporating the excess EG in an evacuated system. The quantity of EG retained at the moment the evaporating rate decreases is proportional to the surface area. Later on Carter et al. (1965) suggested the use of ethylene glycol monoethyl ether (EGME) having high vapour pressure at room temperature than EG for determining surface area of the layer silicate minerals and soils.

The concept of sustainability in agricultural system

All forms of production systems exist today, from nomadic herding to continuous, intensive monocrop systems. Their distribution is determined by soil and climate conditions, and by social and economic factors. In very general terms, as one moves from drier to wetter areas, pastures and animals become less important, and trees more so. Population density is greatest where soils are most fertile, and management systems the most intense. Most production systems have evolved so that they are sustainable in terms of the environmental conditions prevailing at the time—including the level of demographic pressure.

1. Introduction

Seeds have complex and remarkable diversity in shape, size, colour and morphological and anatomical sculpturing details of seed coat, which are valuable for taxonomic significance and can be used as an additional parameter combined with other parameters during taxa identification and classification. Distinctive seed coat morphology usually allowing identification of species even solely on the basis of seed characters. All members of a particular taxon possess variation within acceptable limits of their description and can be used for identification purposes. High structural diversity provides most valuable criteria for classification between species and family level. Takhtajan (1959) pointed out that even for the phylogenetic correlation between families and genera, the structure of the seed coat might be important. Most taxonomists agree that data concerning the structure and microstructure are of great significance mainly for the classification of angiosperm taxa. Consequently, in addition to vegetative and reproductive characters, seed characters are being used in most of the studies for the analysis of taxonomic relationship in wide variety of plant families. It was observed that seed morphology and anatomical features are rather conservatives, which makes them taxonomically important (Esau 1977; Barthlott 1984).

Gaertner (1788-1805) was the first botanist to use seeds and fruit morphology for comparison and identification of different taxa. Later on, Harz (1885) presented a comprehensive account of seed structures and their individual characters of different taxa. In the 20^th century interest in legume seed morphology was renewed. Capitaine (1912) studied the seed morphology of entire family and concluded that seed morphological characters are helpful in legume classification and identification at the tribal, generic and specific levels. After 40 years of Capitaine, Jensen (1998) documented 27 publications on seed morphology. Gunn (1981) summerized the seed characteristics for 510 legume genera. Seed characters of seventeen families were compared by Isley (1947). Duke (1961) indicated that seed shape, sculpturing and colour provide a critical indication of the systematic position of species of Drymaria (Caryophyllaceae). Obermeyer (1962) pointed out that the only reliable distinction between the two genera Chlorophytum and Anthericum of Liliaceae appears the number and shape of seeds. Hairy outgrowth on the testa, their length and colour provide useful characters in the distinction of genera/species in Malvaceae, Convolvulaceae, Asclepiadaceae and Acanthaceae. Gunn (1970, 1971) divided one hundred species of Vicia into major and minor groups on the basis of seed size, shape and length of the hilum, lens position and seed coat pattern and sculpturing. He successfully employed these characters up to specific level. Gunn and Gaffney (1974) studied the seeds of 42 economically important taxa of Solanaceae, including hilum and surface pattern as key character. Later on Gunn and Barnes (1977) had also used seed characters in the delimitation of Erythrina species. Seavey et al. (1977) studied the seeds of 210 species of Onagraceae, where seven seed groups were recognized and their evolutionary implications were also discussed. Matthews and Levinus (1986) demonstrated fairly constant seed characters in those species of Portulacaceae which have restricted geographical distribution, whereas the seeds of species with wider geographical distribution show varied morphological structures.

Tannins: Group of plant phenolic compounds found in higher plants as secondary metabolites. Their main function is to defense against microorganisms, insects, and herbivores and in allelopathy.

Taxis (tactic orientation; formerly topotaxis): Movement of motile organisms, cells or organelles, the prevailing direction of which is determined by the direction of the stimulus.

TATA box (Hogness box): A sequence of nucleotides which serves as the main recognition site for the attachment of RNA polymerase in the promoter region of eukaryotic genes. Located at around 25 nucleotides upstream of the transcriptional start site. It consists of seven base consensus sequence, TATAAAA.

TCA cycle: Tricarboxylic acid cycle. It is also called as citric acid cycle or Krebs cycle, It is a step in aerobic degradation of glucose to convert into energy. During this cycle several intermediates are produced, which leads to several metabolic pathways. During this cycle, there is production of reductancts such as NADH, FADH and GTP.

Telomere: Sequences at both termini of each eukaryotic chromosome that facilitate replication.

Tabernaemontana, commonly known as Chandni, Crape Jasmine or Pinwheel Flower, is an evergreen flowering shrub valued for its pure white flowers. Its flowers are extensively used for garlands, decorations, weddings, hair adornment and religious offerings. Unopened buds can also serve as a substitute for jasmine during its off-season. The chapter covers origin, distribution, genetics, botanical description and important cultivars, including single and double forms. Production practices include climate and soil management, propagation, planting, nutrient management, irrigation, pruning and other cultural operations. The chapter also addresses pests and diseases, harvesting, postharvest treatments, storage and yield, with emphasis on maintaining flower freshness and commercial quality.

1. Introduction

Plant taxonomy and the cultivated plants have a close relationship of great antiquity. This ancient relationship can be traced back to the invention of incipient agriculture (neolithic revolution), about 10-13000 years BCE (Stearn 1965; Balter 2007). The early foundation of plant taxonomy was thus laid largely on humans’ knowledge on cultivated plants. This body of rudimentary knowledge on identifying the properties of different types of plants for food, medicine, clothing, shelter, and other uses led to the development of plant taxonomy- the first aspect of plant science to emerge as botany (Constance 1957). The fundamental principles of modern plant taxonomy with regard to the natural vegetation and flora were originally derived from cultivated plants (Li 1974).

The active process of cultivation of a plant, primarily through the intentional activity of humans has been considered the main criterion for categorising cultivated plants (Mansfeld 1959). According to Mansfeld’s approach, the number of cultivated plant species of the world was estimated to be about 7000 species, excluding partly the forestry plants, ornamentals, and plants used in amenity horticulture (Henelt and IPK 2001). Khoshbakht and Hammer (2008) expanded this list to include a total of 35000 species of cultivated plants, including 28000 ornamental plant species connected to gardening and landscaping. About 2500 plant species have undergone domestication worldwide, with over 160 families contributing one or more crop species (Zeven and de Wet 1982; Dirzo and Raven 2003).The growing numbers of cultivated plant species and the enormous diversity found in each of these species offer plenty of challenges as well as opportunities to the taxonomists to take up cultivated plant taxonomy as an active scientific pursuit. Despite this, there has been a lack of interest among taxonomists towards cultivated plant taxonomy.

Soil organisms play vital role in agri-ecosystem processes such as nutrient cycling, soil detoxification and organic matter decomposition. They also influence water infiltration by improving soil porosity through production of polysaccharides. Rhizosphere microorganisms help plant to acquire macro and micronutrients, produce phytohormones for better crop establishment. Soil microbes process organic matter to provide a balance of minerals and nutrients which are utilized by plants to achieve healthy and vigorous growth. Microbes also colonize on plants parts and offers protection by excluding many plant pathogens. They are the basic indicators of soil fertility and are responsible for maintaining a hospitable environment for crop growth.

Primary metabolism in a plant comprises all metabolic pathways that are essential to the plant’s survival. Primary metabolites, namely, carbohydrates, lipids and proteins are compounds that are directly involved in the growth and development of a plant whereas the secondary metabolites are compounds produced in other metabolic pathways that, although important, are not essential to the functioning of the plant. However, secondary plant metabolites are useful in the long term, often for defense purposes, and give plants characteristics such as color.
 
Secondary plant metabolites are used in signalling and regulation of primary metabolic pathways. Plant hormones, which are secondary metabolites, are often used to regulate the metabolic activity within cells. Secondary plant metabolites help the plant maintain an intricate balance with the environment, often adapting to match the environmental needs. Plant metabolites that color the plant are a good example of this, as the coloring of a plant can attract pollinators and also defend against attack by animals. Terpenes, alkaloids, sterols, pigments, growth regulators, and phenolics are the important secondary plant metabolites.

Plants are exposed to various stresses such as temperature, water, oxygen, salts, nutrition etc, during its growth under dry land condition. Various simple tests have been developed by plant scientist for evaluation of different cultivars against these stresses. These tests are simple to perform in the laboratory with fewer instruments that are commonly available in most of the laboratories in developing countries. These tests have significance in plant water relations and cultivars reliably screened for various stresses. The crops upon which these tests have been performed here are used by the proponents of the tests. These tests are not advised to be performed on crops other than those used by the authors. It probably needs standardization for other crops. The following test has been explained for the benefit of the readers.

Introduction

Test of pollen viability (extent of pollen viability) is important in pollination experiments to ensure fertility/seed setting. It is also important in monitoring state of pollens during storage in ecological/ taxonomic studies as well as during research on pollen biochemistry, genetics, stigma interactions, and incompatibility studies. Pollen viability refers to the ability of pollen to accomplish post-pollination events to achieve fertilization. Pollen viability test has gained importance particularly in plant breeding and hybrid seed production experiments/ practices. In this chapter, you will be acquainted with the basics of pollen viability test. However, for more professional knowledge on the topic readers are suggested to refer specialized books.

Pollen Viability Test

It is important to know the extent of viability of pollens used in pollination experiment, in finding fertility of a plant of interest, to monitor state of pollen on storage, in ecological/taxonomic studies, as well as in research on biochemistry, genetics, stigma interaction, incompatibility assay, etc. of pollen. Several pollen viability tests have been developed time-to-time. It is recommended to initially use different tests for a given pollen type to find out the most appropriate method that provides reliable result on pollen viability. It is important to note that every procedure requires standardization of protocol according to the sample used. Following different tests are used for assessing pollen viability.

1. In-vitro Pollen Germination Test

It is a simple, speedy, and quantitative test for assessing pollen viability. In this method, a collection/sample of pollen is germinated in vitro and observed under microscope. The percentage of germinating pollens (producing pollen tube) after appropriate treatment and a given period of time is determined. The percentage provides an index of viability of the pollen sample.

Introduction

Seed is a living entity (mature ovule) consisting of a tiny plant in embryonic stage along with the stored food protected by an outer layer known as seed coat. Seed is an important biological input in agriculture, which is economically important for successful crop husbandry. Cotyledons serve as the source of nutrients (proteins, carbohydrates, and oils) that not only nurture the plant at the early developmental (seedling) stages of plant but also the vegetarian (heterotroph) organisms. Being one of the important inputs in agriculture, viability of seed needs to be assessed to know the germination percentage of seeds used to raise a crop. Since viability of seeds may deteriorate on storage depending on the storage conditions. Therefore, often it becomes important to test viability/germinability and vigour of seeds before being used for raising a crop or conducting an experiment. In this chapter, we briefly discuss about the different methods which can be used for testing the viability of seeds.

Seed Viability

Seed viability can be defined as the capability of the seed to germinate and produce a normal seedling within a specified period. In other words, seed viability indicates how many seeds in a lot are alive and could develop into plants under appropriate field conditions. It is very important that most of the stored seeds are capable of germinating when sown in a field. They must possess higher viability at the beginning of storage and maintain it over the storage. A seed with high initial viability survives longer in storage; however, the viability deteriorates slowly at first and then rapidly as the storage prolongs. It is important to know the duration of storage after which a considerable decline in viability occurs under given storage conditions so that to take necessary action to revive the seeds. Excessive deterioration in vianility of seeds may lead to loss of the seed/material/ germplasm.

Free radicals, such as singlet oxygen (¹_O2), superoxide (O₂ ^.–) and some active derivatives of oxygen, like H₂O₂ are although generated in plant cells during the course of normal metabolic processes, but their production is notably enhanced when the plants are exposed to any kind of stress environment. The reactive oxygen species damage important cellular components, including cell membrane by peroxidation of lipid and de-esterification of fatty acids.

Plants have evolved a wide range of mechanisms to contend with this problem. The antioxidant defence system of a plant comprises of a variety of antioxidant molecules and enzymes which take care of free radicals produced in the cells. These enzymes are popularly termed as the ‘Scavenging Enzymes’ or ‘Antioxidative Enzymes’.

Conversion of oxygen molecules into its reactive form leads to induction of a sort of stress in plants, to which we call as ‘Oxidative Stress’. It is the equilibrium between oxidative stress and antioxidant enzymes, which determines the fate of a plant.

Plants possess scavenging enzyme system to protect them from destructive oxidative reactions. These enzymes are not restricted to the intercellular compartment but are also found in the apoplast to a limited extent. Some of the enzymes are as follows:-

Abstract

Drought and salinity are two major abiotic stresses that affect various aspects of human lives of one third world population including human health and agricultural productivity. Abiotic stresses such as salinity, drought, nutrient deficiency or toxicity, and flooding limit crop productivity world-wide. A living organism is considered resistant if it tolerates a given physicochemical stress. The presence of microorganisms within the soil, specially a main group known as arbuscular mycorrhizal fungi (AMF), is a key factor in the adaptation of plants to the different ecosystems. The AMF colonizing within the plant roots can determine the success of plants in salt- and/or drought-affected areas. The symbiosis with AMF has been proposed as one of the mechanisms of salinity and water stress avoidance. Studies have shown greater drought and salinity tolerance in AMF-colonized plants by a number of mechanisms. These mechanisms include enhanced water and nutrient uptake directly by extraradical hyphae, higher leaf stomatal conductance and/or better root system architecture, higher capacity of osmotic adjustment and antioxidant defense systems, better soil structure due to higher glomalin and over expression of genes encoding antioxidant enzymes.

Thermal Analysis

Physical and chemical properties of any soil are governed largely by the mineralogical make up of the soil, and more specifically by the clay fraction. Identification, characterization and an understanding of properties of different minerals are important in evaluating soils in relation to classification, agronomic practices and engineering purposes.

Clay fractions of soils are usually composed of mixture of one or more secondary phyllosilicate minerals along with primary minerals inherited from the parent materials. Identification of minerals species as well as their quantitative estimation in such polycomponent system require application of several qualitative and quantitative analysis.

The methods used for identification and quantification have steadily been improved over the last 50 years. Among the several methods often used for this purpose three are very common, e.g. (i) differential thermal analysis (DTA), (ii) x-ray diffraction analysis (XRD), and (iii) infrared spectroscopy (IR).

Thermal analysis is a term covering a group of analyses that determine the changes of some physical parameters, such as weight, energy, dimension and evolved volatile substances with change in temperature. Many soil constituents undergo various thermal reactions upon heating, which can be utilized as a diagnostic tool for qualitative identification and quantitative estimation of the substances. These reactions may be exothermic or endothermic, and phase change or crystal change may take place during reactions. Whatever be the reactions, if properly measured, they give some valuable information to identify the substance under consideration.

Among the several methods available for thermal analysis, three methods are frequently used in soil research: differential thermal analysis (DTA), differential scan calorimetry (DSC), and thermogravimetry (TG). Among these, DTA and TG will be discussed here.

Crop health valuation plays a vital part in modern agriculture as it positively stimulates food security, economic constancy, and ecological sustainability. This process involves monitoring and evaluating the growth and development of crops, finding possible stressors or diseases, and executing suitable management practices. Judicious and precise evaluation of crop health permits early exposure of diseases, pests, and other irregularities. Detecting these problems at their early growth stages can aid prevent extensive occurrences and decrease the need for unnecessary pesticide use, indorsing justifiable crop management practices. Assessing crop health on a regular basis help in enhancing yields by detecting factors that could inhibit crop productivity. Agricultural systems can be made resource-efficient by incorporating tools, modern technologies and information management systems that come under the precision agriculture concept. Modern scientific improvements, comprising Global Positioning System (GPS), grain yield monitoring system, Variable Rate Technologies (VRT), sensor networks, and Femote Sensing (RS), have assisted farmers to detect in-field heterogeneity of crop and soil and adopt site-specific farm management practices. Varying the application of inputs with VRT can decrease the cost of inputs and labor, increase productivity, and decrease the potential environmental influences from the overuse of farm inputs. However, the success rate of VRT needs precise and consistent information on the location and extent of crop and soil health conditions, such as crop nutrient shortages, soil and crop water deficiency, and injury caused by insect pests, weeds, or pathogens. So far, the dominant remote sensing technologies that the farming sector has used are visible (VIS), Near-Infrared (NIR), and short-wave infrared (SWIR) sensors. The use of thermal sensors in agriculture is to some extent narrow despite its wide use in the areas of medical science (Ring and Ammer, 2012), intelligence/military (Hinz and Stilla, 2006), and the food processing sector (Vadivambal and Jayas, 2011). Temperature regulates germination, plant growth, flowering, maturity and many insect and disease processes. Thermal indices integrate temperature over time to quantify biological development more effectively than simple temperature averages. This chapter introduces growing degree days, heat units, photothermal units, heliothermal units and related measures. It explains base, optimum and maximum temperatures and connects accumulated thermal time with crop phenology. These indices help predict development stages, adjust sowing dates, estimate harvest periods, schedule farm operations and assess varietal suitability. Thermal-index analysis is particularly valuable under climate variability because it enables researchers and farmers to anticipate shifts in crop development and improve climate-responsive agricultural management.

Tissue culture refers to the collection of techniques used to maintain or grow of cells, tissues and organs under controlled environment. The German botanist G. Haberlandt (1854-1945) originated the concept of cell culture in which he described his pioneering experiments with isolated plants cells and developed a concept of totipotency (potential of a cell to reproduce entire plant and all of its characteristics). He is considered as father of tissue culture.

Object: To demonstrate anaerobic respiration. 

Material required: Soaked pea or gram seeds, mercury, test tube stand, forceps and KOH pellets. 

Theory: Generally, plants at certain stage have the capacity to respire in absence of oxygen i.e. anaerobic respiration. In this process, oxidation of carbohydrate takes places, ethyl alcohol and CO2 being end products and only a little amount of energy is liberated-

Object: To demonstrate chlorophyll fluorescence.

Material required: Spectrophotometer, fresh green spinach, mortar, pestle, acid washed sand, acetone, funnel, filter paper and test tubes.

Principle: Chlorophyll molecule absorbs light in the blue (450nm) and red (660nm) region and get into excited state and emits an electron. The electron drops back to the ground state within 10-9 seconds and some energy (which is not used in biochemical reaction) are given out as the observed fluorescent far red light.

Procedure and observation: Take some soft green spinach leaves. Grind these with a pestle -mortar in acetone adding a pinch of acid washed sand. Make a fine paste. Filter to get a clear solution in a test tube. Take the test tube in bright sun light.

Object: To demonstrate that light and carbon dioxide are essential for photosynthesis (Moll’s half leaf experiment). 

Material Required: A potted plant, a wide mouthed bottle, caustic potash, splits of rubber Cork and vaseline etc. 

Object: To demonstrate that O₂ is evolved during photosynthesis. 
 

Material required: Hydrilla plant, beaker, test tube, funnel, pond water and pyrogallol. 
 

Theory: Oxygen is a byproduct of photosynthesis, which is evolved in presence of light and chlorophyll during photosynthesis.

Object: To demonstrate that oxygen is used up during respiration. 

Material required: A conical flask, some germinating seeds, glass tube having two bends at right angles, mercury, beaker, small tube and caustic potash (KOH).

Theory: Plants at some stage have a capacity to respire more rapidly in presence of oxygen i.e. aerobic respiration. In this process, oxidation of carbohydrate occurs resulting in conversion of carbohydrate into carbon dioxide (CO₂) and water with release of large amount of energy.

Object: To demonstrate that water is lost from a living plant during transpiration. 

Material Required: A potted healthy plant, a bell jar, a rubber sheet or oil cloth, a glass sheet and grease.

Theory: Transpiration: Loss of water in the form of water vapour from the internal tissues of living plants through aerial parts of the plant is known as transpiration.

Object: To demonstrate the measurement of growth in plants with the help of Arc auxanometer. 

Material required: Potted plant, Arc auxanometer, rope and weight.

Theory: Growth is a permanent change in plants with respect to its size, form, weight and volume.

Object: To demonstrate the optimum conditions for seed germination. 

Material required: Beaker, glass slide, bean seeds, thread and water.

Theory: Germination is a process by which the embryo in the seed becomes activated and begins to grow into a new seedling under favorable conditions. Food is stored in the seeds in dry conditions but the developing embryo cannot utilize this dry food. Food can be utilized in liquid form only and seeds can utilize only dissolved oxygen. Excess water stop germination because once all the dissolves oxygen is utilized by the seed, further germination is not possible as the life supporting oxygen is lacking. Suitable temperature is necessary because low temperature retards the embryo activity and high temperature destroys the delicate embryo tissue. Seed usually germinate between 0oC to 50oC temperature and the optimum temperature is lies between 25 to 30oC. During germination rapid cell division takes place. Energy is required for cell division. This energy is obtained from oxidation. The oxygen is required for oxidation is supplied by the air.

Object: To demonstrate the phenomenon of endosmosis and exosmosis by using grapes and raisins.

Material required: Grapes, raisins, beaker or petridish, water and sugar solution.

Object: To demonstrate the phenomenon of osmosis using potato osmoscope. 

Material required: A potato tuber, sugar solution, beaker, water, petridish, knife, measuring cylinder, and stand.

Theory: Osmosis is a phenomenon in which water molecules diffuse through the semi permeable membrane from region of its higher concentration to the region of lower concentration.

Object: To demonstrate unequal transpiration by the two leaf surfaces from four leaf method.
