Test of pollen viability (extent of pollen viability) is important in pollination experiments to ensure fertility/seed setting. It is also important in monitoring state of pollens during storage in ecological/ taxonomic studies as well as during research on pollen biochemistry, genetics, stigma interactions, and incompatibility studies. Pollen viability refers to the ability of pollen to accomplish post-pollination events to achieve fertilization. Pollen viability test has gained importance particularly in plant breeding and hybrid seed production experiments/ practices. In this chapter, you will be acquainted with the basics of pollen viability test. However, for more professional knowledge on the topic readers are suggested to refer specialized books.

Pollen Viability Test

It is important to know the extent of viability of pollens used in pollination experiment, in finding fertility of a plant of interest, to monitor state of pollen on storage, in ecological/taxonomic studies, as well as in research on biochemistry, genetics, stigma interaction, incompatibility assay, etc. of pollen. Several pollen viability tests have been developed time-to-time. It is recommended to initially use different tests for a given pollen type to find out the most appropriate method that provides reliable result on pollen viability. It is important to note that every procedure requires standardization of protocol according to the sample used. Following different tests are used for assessing pollen viability.

1. In-vitro Pollen Germination Test

It is a simple, speedy, and quantitative test for assessing pollen viability. In this method, a collection/sample of pollen is germinated in vitro and observed under microscope. The percentage of germinating pollens (producing pollen tube) after appropriate treatment and a given period of time is determined. The percentage provides an index of viability of the pollen sample.

Seed is a living entity (mature ovule) consisting of a tiny plant in embryonic stage along with the stored food protected by an outer layer known as seed coat. Seed is an important biological input in agriculture, which is economically important for successful crop husbandry. Cotyledons serve as the source of nutrients (proteins, carbohydrates, and oils) that not only nurture the plant at the early developmental (seedling) stages of plant but also the vegetarian (heterotroph) organisms. Being one of the important inputs in agriculture, viability of seed needs to be assessed to know the germination percentage of seeds used to raise a crop. Since viability of seeds may deteriorate on storage depending on the storage conditions. Therefore, often it becomes important to test viability/germinability and vigour of seeds before being used for raising a crop or conducting an experiment. In this chapter, we briefly discuss about the different methods which can be used for testing the viability of seeds.

Seed Viability

Seed viability can be defined as the capability of the seed to germinate and produce a normal seedling within a specified period. In other words, seed viability indicates how many seeds in a lot are alive and could develop into plants under appropriate field conditions. It is very important that most of the stored seeds are capable of germinating when sown in a field. They must possess higher viability at the beginning of storage and maintain it over the storage. A seed with high initial viability survives longer in storage; however, the viability deteriorates slowly at first and then rapidly as the storage prolongs. It is important to know the duration of storage after which a considerable decline in viability occurs under given storage conditions so that to take necessary action to revive the seeds. Excessive deterioration in vianility of seeds may lead to loss of the seed/material/ germplasm.

The growing chemicalization of milk presents a great challenge to the dairy industry. A number of chemical compounds get entry into milk, intentionally or un-intentionally, through their direct or indirect usage for crop, animal; or dairy husbandry practices, as environmental contamination. Several of these contaminants may be naturally present on grazing grass or use as pesticides for green fodder or they may get entry through use of compounded animal feeds. These eventually get excreted into milk as residues and pose problems to food safety. The risk of health hazards arises from veterinary drugs, residual pesticides, heavy metals, detergents and micotoxins etc. It is therefore, become necessary to detect these contaminants in order to make food safe for human consumption.

Detection of Aflatoxin M 1 in Milk

Material requirements : Glass column(30 cm x 1 cm), Silica gel (63-200 µg particle size), volumetric flasks, beakers, micro pipette, HPLC equipment fitted with C 18 column(25 cm x 4.6 mm) and fluorescence detector.

Reagents: Sodium chloride solution (400g in 1000ml water), acetonitrile/water mixture (25:75), sodium sulphsate, chloroform, toluene/acetic acid mixture (9:1), acetonitrile/diethyle ether/hexane mixture (1:3:6), chloroform/acetone mixture (8:2), n-hexane and aflatoxin standard ampule.

Preparation of stock solution : Transfer contents from 2.5ml or 0.5ml ampoule, quantitatively, to 25ml or 50ml volumetric flask respectively. Rinse ampoule with small quantities of chloroform and collect rinsing into flask. Makeup the volume with chloroform, mix well and store at 5º C or below. These solutions will contain 1 µg per ml aflatoxin M1.

Working solution : Transfer 0.5ml of stock solution to a 10ml conical flask. Evaporate contents to dryness using inert gas. Dissolve residue in 10ml of acetonitrile/water mixture. Prepare series of dilutions containing 0.0025, 0.005, 0.01 and 0.025 µg/ml aflatoxin. Similarly treat 5ml of stock solution to give a concentration of 0.5 µg/ml.

Preceding chapters discuss the essentials of probiotics, their role in maintaining gut health in combination with prebiotics and significance of systematic classification of probiotic isolates. Further, to draw an idea how probiotics function in the gut, it is essential to know the interaction of probiotics with other microorganisms in the gastrointestinal milieu - the site of their probiotic action. Knowledge in the ecology of probiotic microorganisms in host environment such as gut gives an ideal picture of their functioning manner and the factors (both host and microbial) responsible for it.

Study of microbial ecology in the gastrointestinal tract is very difficult since gut harbors highly diversified microflora that varies not only at species level but also at individual level. The diversity of gut microflora in two healthy individuals of same species may exhibit variations in their composition and it may be influenced by geodemographic factors also. In this regard, the present chapter deals with the study of ecology of Lactobacillus- most commonly used probiotic bacteria in the gastrointestinal tract which in turn helps in framing selection criteria for screening of potential probiotics.

Recent studies on interaction between human and gut microflora discussed that co-evolution between microorganisms and their host have contributed for the development of a very complex microbial diversity in the gut. Highly diversified microflora in the human gut is a consequence of natural selection at two levels, top-down and bottom-up selection pressure. Hierarchy theory expounds that higher levels compel lower levels for possible organizational solutions i. e. organisms at the higher levels govern the structure or population dynamics of an ecosystem while on contrary lower levels control the structure of ecology when bottom-up level is the selection pressure. In case of interaction between human and gut microflora, being host human is at higher level in the hierarchy for selection and as mentioned above, top down selection pressure is driven by higher levels; host selects the beneficial microorganisms for functional redundancy leading to the development of a microbial community with divergent microbial divisions. Chances for lateral gene transfer also thought to influence the selection process and there by structure of the microbial community. On the other hand, competition among the members of host selected microbiota exert bottom up selection pressure resulting in the development of highly specialized genomes with functionally prominent set of genes. Natural selection at host level obviously favors functionally constant microbial communities which benefits the host with functional redundancy encoded in their genomes. Mean while bottom-up cell-level selection pressure aspire to favor niche specialization to avoid competition between the members of microbial community. In order to be a part of specialized niche, to become more potent microorganisms adapt a particular set of abiotic and biotic factors within the habitat by acquiring new sets of genes essential for their survival and thus escape the competition from decrepit microbial species.

It explains freezing-point depression, ice-crystal formation, colloidal behaviour, foam development, and structural changes during processing. Particular attention is given to the relationship between microscopic structure and sensory properties such as smoothness, creaminess, body, and meltdown. For the dairy industry, social responsibility is not an abstract concept; it is a pressing need. Across India, small-scale farmers form the backbone of this sector, especially in rural regions where economic vulnerability is widespread. These farmers often face numerous challenges, including fluctuating milk prices, limited market access, and a lack of resources to improve productivity (Kamath et al., 2019; Sadashive et al., 2017). Businesses in the dairy sector can act as catalysts for change by creating fairer systems, empowering marginalized groups, and ensuring that ethical treatment of livestock becomes standard practice. Beyond its immediate economic impact, the industry has a profound role to play in addressing broader social issues, such as gender inequality and food security, while promoting sustainable development that benefits everyone involved (Jaiswal, 2018; Garai et al., 2019).

Therapeutic foods are products, which are multiutility food products that may be used as nutrition as well as medicine. A nutraceutical may be defined as a substance, which has physiological benefit or provides protection against chronic disease. Nutraceuticals are used to improve health, delay the aging process, prevent chronic diseases, increase life expectancy, or support the structure and/or function of the body. Oflate nutraceuticals are receiving considerable interest due to potential nutritional, safety and therapeutic effects. The examples of therapeutic foods are the foods used for feeding of malnourished children on an emergency basis or to supplement the diets of persons with special nutrition requirements, such as the elderly. Therapeutic foods can also form a part of treatment/prevention/reduction of other health conditions such as diabetes, cardiovascular disease, obesity, anaemia and any other lifestyle disorders.

The incidences of diet-related diseases are progressively increasing due to greater availability of calorie-rich foods and a sedentary lifestyle. Obesity, diabetes, atherosclerosis, and neurodegeneration are major diet-related diseases. Nutraceuticals and functional foods represent a novel therapeutic approach to prevent or attenuate diet-related diseases.

There is always a requirement for cheaper formulations using locally available ingredients that are processed in a safe, reliable, and financially sustainable methods. Some of the literature available on health foods for various diseases is summarized below. However, since the burden of diabetes in India is staggering. In 2017, 72 million cases of people suffering from diabetes were reported and the figures are expected to reach 134 million by 2025, this chapter more emphasis is given on the safe, natural treatment/reduction of diabetes by herbal means.

Abstract

Application of solar energy for drying of agricultural food products is one of the most attractive and cost-effective drying techniques. However, the intermittent nature of the solar energy is one of the major shortcomings of the solar drying system that can be alleviated by storing excess energy during the peak time and using it in off sun hours or when the direct availability of solar energy is inadequate. The use of sensible and latent heat storage systems in solar dryer is an effective way of storing thermal energy for drying purpose. This chapter will present information on different thermal energy storage (TES) materials, their advantages and disadvantages. The information available in the literature regarding the studies conducted on TES assisted solar drying systems is also covered in this chapter. In addition, some of the models that have been used by different researchers to test the performance of different TES materials are also reviewed.

Heat treatment is the most commonly used method in the dairy industry for milk processing. Its primary objective is to eliminate pathogens and reduce spoilage-causing organisms, ensuring milk is safe for consumption and has a longer shelf life. While advanced methods like high-pressure processing, pulsed electric fields, ultrasound processing, cold plasma treatment, and ultraviolet sterilization have emerged, heat treatment remains the preferred choice due to its simplicity, reliability, and proven effectiveness. All dairy products undergo some form of heating during processing, whether through pasteurization, sub-pasteurization (e.g., for milk used in aged cheeses), or sterilization. To facilitate this, liquid heating systems such as continuous heat exchangers or jacketed tanks are widely used, typically powered by steam or steam-heated water. Consequently, steam generation boilers are a standard feature in almost all dairy processing facilities.

1. Elimination of harmful pathogens in milk, ensuring it is safe for consumption and protecting public health.
2. Increasing the keeping quality of milk by reducing the number of spoilage organisms.
3. Inactivating enzymes naturally present in milk or secreted by microorganisms to prevent enzymatic degradation.
4. Preventing the autoxidation of lipids, which can lead to off-flavors and spoilage.
5. Inactivating agglutinin to reduce rapid creaming and maintain uniform consistency.
6. Enhancing the growth of starter cultures by eliminating natural inhibitors and dissolved oxygen in milk, essential for producing cheese and fermented products.

The definition of quality is described from a very simpler one “fitness for purpose or use” to a comprehensive “the totality of features and characteristics of a product or service that bear on its ability to satisfy stated or implied needs”. The concept of total quality management in food industry is adopted recently and it is the “integrated organization approach in delighting customer (both external and internal) by meeting their expectations on a continuous basis through everyone involved with the organization working on continuous improvement in all products/processes along with proper problem solving methodology”. TQM is concerned with the integration of all the efforts in organization towards quality improvement, quality development and quality maintenance to meet full customer satisfaction at economic levels. TQM enhances the work culture and employee’s satisfaction through participation and involvement and consequently the image of the organization.

Cheese, butter, whole milk powder, and skimmed milk powder are a major part of the world’s dairy trade. The demand for worldwide dairy exports is highly concentrated. With a share of 12 per cent, New Zealand is the largest exporter of dairy products, followed by Germany, Netherlands, France and the US. Germany has been the world’s major dairy importer as well with a share of 10 per cent. Other large dairy product importers are China, Netherlands, France, Italy and Belgium.

Before 1999, India was a net importer of dairy products. The successful implementation of the ‘Operation Flood’ program and trade reforms caused the direction of India’s dairy trade to change marginally. India has been a net exporter of dairy products since 2000. Statistics show that India’s export share of dairy products over world dairy exports increased marginally from 0.1 per cent in 2001 to 0.5 per cent in 2019, while its import share of dairy products over world dairy imports increased marginally from 0.018 per cent in 2001 to 0.04 per cent in 2019. The Middle East, South Asia, South East Asia, and the USA are the major markets for India’s dairy products. Conversely, India is a favorable market for the European Union, New Zealand, Australia, UAE, and US dairy products.

Importance of Traditional Foods

Traditional convenience food, or tertiary processed food, is commercially prepared food designed for ease of consumption. Products designated as convenience foods are often prepared food stuffs that can be sold as hot, ready-to-eat dishes; as room-temperature, shelf-stable products; or as refrigerated or frozen products that require minimal preparation (typically just heating).

These products are often sold in portion controlled, single serve packaging designed for portability for “on-the-go” eating. Convenience food can include products such as candy; beverages such as soft drinks, juices and milk; fast food; nuts, fruits and vegetables in fresh or preserved states; processed meats and cheeses; and canned products such as soups and pasta dishes.

Vadagam is a traditional product mostly prepared from rice flour and also sago. It is an deep fat fried product and consumed as a side dish in the meals. Papad also known as appalam or papadam in South India, is essentially a thin wafer like product, usually circular in shape rolled from a dough made out of pulse, cereals, edible vegetable oil, alkaline and mucilaginous additives. It is normally consumed in toasted or fried form. In India, papad has been associated and deeply entwined with social customs/ rituals and has been developed into culinary art.

Traditional knowledge is a community based functional knowledge system which is developed, preserved and refined by generations through continuous interaction, observation and experimentation with their surrounding environment. It includes beliefs, values, and practices gathered from the practical experience of older generation, and its whole function is survival and development of culture of people. Traditional foods, originated from ancestral kitchens are developed through ages, invented, modified, utilised and evolved to improve nutritional and social wellbeing of the people around the world. Most of them are culture specific, region specific, environment specific, community specific and season specific. These foods are socially, culturally and economically important and provide food security, enhance livelihood and improve nutritional and social wellbeing of people. Food culture arises out of the place of a people’s origin and so traditional local foods hold the potential to bind and stabilise communities and enable a cultural continuity through conserving their histories. Indian cuisine is represented as a wide spectrum of food cultures with distinctive regional differences and preferences (Achaya, 1998).

Training is a major manpower management function. No organization can afford not to train its employees, regardless of his previous education, training, and experience. TRANSFER

A transfer is a change in job assignment. It may involve a promotion or demotion or no change at all in responsibility and status. It is a change in assignment in which the employee moves to another job at approximately the same level of responsibility, the same skill, the level of pay. Transfer is to be distinguished from promotion and demotion which imply an ascending or descending change respectively, in the hierarchy of positions. A transfer may be either temporary or permanent, depending upon the need, and may occur within a department, between departments and divisions, or between plants or officers within a company/organization.

Introduction

Commonly available food materials especially dairy products, cereals and fruits are perishable and require processing for long term storage. The methods of food storage has been known from the pre historic times and was mostly based on processing the food into a more stable form with a longer shelf life. Such processes increases the time range of food availability and in many cases improves the taste thereby, cumulatively improving the food value. One of documented method of food preservation is by fermentation and is age old. Other commonly used methods include drying, refrigeration, salting and fermentation (Luck 1985, Paulus, 1985, Christensen et al., 2003, Martindale and Schiebel, 2017).

Flavour and appeal of food has been considered as one of the important factors contributing to the demand of the food. Originally the flavours were largely plant derived, current industrial practices largely utilise synthesis by chemical reactions and subsequent extraction to achieve a uniform quality (Weckerle et al., 2006, Davidovich-Rikanati et al., 2008, Sethupathy et al., 2018). These chemicals referred to as “flavourings” are defined as compounds that enhance the taste, appeal and palatability of food. They are naturally derived or artificially synthesised compounds. Considering that many artificially synthesised chemical flavours might affect the health adversely. Industrial food research has now shifted their focus on flavourings that are naturally available (Cohen et al., 2018). Such naturally occurring flavourings are referred to as bio-flavours (de Billerbeck 2015). The ease of producing bio active compounds from microbial culture extracts has played significant role in industrial food biotechnology (Verachtert et al., 2005).

The Hans Krebs cycle, known by different names like the citric acid or tricarboxylic acid cycle, was named after the scientist Hans Krebs. The citric acid cycle is named after the citrate product formed in the cycle. The name tricarboxylic acid is given as citrate itself, which is a tricarboxylic acid due to the presence of three carboxyl functional groups (-COOH).
The tricarboxylic acid (TCA) cycle occurs in aerobic conditions and within mitochondrial ETC, involving various sequential oxidoreductase reactions. The cycle requires Acetyl-CoA (C2 molecule) and oxaloacetate molecule (C4 molecule) to begin with the formation of a citrate molecule (C6 molecule). Acetyl-CoA is generated from the pyruvate (glycolysis), the fatty acids (lipolysis), or by oxidative deamination of specific amino acids. The complete oxidation of a glucose molecule results in the end products like energy (ATP), CO2 and H2O. A molecule of Acetyl-CoA has two carbons. It is formed from a three-carbon pyruvic acid molecule generated from the glycolysis of a sixcarbon glucose molecule. Acetyl-CoA is the substrate for this cycle and is an energy-rich molecule having a thioester bond between coenzyme A and acetic acid.
8.1 Formation of acetyl-CoA from Pyruvic Acid
Pyruvate is obtained by glycolysis or the oxidation of glucogenic amino acids. The pyruvic acid produced in the cytoplasm crosses the inner membrane of the mitochondrion to enter the mitochondrial matrix, where it undergoes oxidative decarboxylation, forming Acetyl-CoA. Acetyl-CoA carries (carrier molecule) the acetyl group to the citric acid cycle. It also links various metabolic products with the TCA cycle. Such products can be from the EMP cycle, beta-oxidation, and oxidative deamination of amino acids. Oxidative decarboxylation of pyruvic acid needs a complex enzyme system known as pyruvate dehydrogenase

The tropical root and tuber crops are comprised of crops covering several genera. They are staple foods in many parts of the tropics, being the source of most of the daily carbohydrate intake for large populations. These carbohydrates are mostly starches found in storage organs, which may be enlarged roots, corms, rhizomes, or tubers. Many root and tuber crops are grown as traditional foods or are adapted to unique ecosystems and are of little importance to world food production. Others such as cassava (Manihot esculenta Crantz) and white-fleshed sweet potato (Ipomoea batatas L.) are known worldwide. Several of these crops have been termed under-exploited and deserving of considerably more research input

Potato and Banana chips: Potato chips are one of the popular snack foods consumed throughout the country. Chips are mostly prepared by small scale artisans / confectioners, and sold in plastic film or paper packs. Recently, however some of the companies have installed plants having relatively large capacities and have started marketing chips in attractive packs. The fat content generally ranges between 25-40% and moisture content between 2-4%. Potatoes having high total solids, low reducing sugars and low amylase activity are most suitable for chips manufacture.

Banana chips are prepared from varieties rich in starch having very low levels of soluble sugars and polyphenol - oxidase activity. These are mostly processed and marketed in Southern States especially in Kerala. Previously, the chips were fried in coconut oil, but now other vegetable oils are also used in their processing. Relatively, banana chips are harder in texture than potato chips.

The machine parts, which have relative motion, rub against each other. The rubbing surfaces are never perfectly smooth, even when they are highly polished. If they are observed under a microscope, one sees small depressions and elevations which get interlocked when the machine parts are under pressure, and give rise to a resisting force called frictional force. Due to friction, the parts get worn out and become useless. Friction produces heat, which, in this case, represents loss of work. To reduce this wear and loss of power caused by friction, certain foreign substances are introduced between the rubbing surfaces which keep them apart. These substances are called lubricants; and lubrication is the science of reducing friction by the application of a suitable substance between the rubbing surfaces of bodies having relative motion.

Types of Bakery Products

Baked goods are produced from either doughs or batters which are a mixture of flour and water made by mixing, beating, kneading or folding. The processing method depends on the ingredients being used and the product being made. Bread is either leavened or unleavened. Leavened bread is made from a mixture of flour, yeast, salt and water. Unleavened bread does not contain yeast and therefore does not rise. It is flat bread that is quicker to make than yeast-bread.

Types of Confectionery

Confectionery also called sweets or candy is sweet food product. Confectionery is divided into two broad and somewhat overlapping categories, Bakers’ Confections and Sugar Confections. Confections are low in micronutrients and protein but high in calories. They may be fat-free foods, although some confections, especially fried doughs, are high-fat foods. Many confections are considered to give empty calories.

Baker’s Confectionery

Baker’s confectionery also called flour confections includes principally sweet pastries, cakes, and similar baked goods. In the Middle East and Asia, flour-based confections are more dominant. Baker’s confectionery includes sweet baked goods, especially those that are served for the dessert course. Baker’s confections are sweet foods that feature flour as a main ingredient and are baked. Major categories include cakes, sweet pastries, doughnuts, scones, and cookie.

Introduction and Relevance
UHT processing of milk was developed to produce a shelf-stable product with minimal chemical changes as encountered with sterilized milk. Aseptically packaged UHT milk can have a shelf life of 06 to 12 months and does not need to be stored in a refrigerator. Its role in the market varies by region. In countries like the India, USA and Canada, where UHT milk occupies a smaller segment of the liquid milk market, it is often used as a backup option, reserved for situations when pasteurized milk is unavailable. Conversely, in regions such as Europe, (particularly France, Belgium, Spain and Germany), Asia, (China, Japan, Thailand and Vietnam) Latin America and (Brazil) in UHT milk is the dominant form of milk available and is consumed regularly. In markets where it is a backup option, UHT milk requires long-term stability of 6–12 months, while in regions where it is used daily, a shorter shelf life of three months or less is often sufficient.
Ultra-High Temperature (UHT) treatment involves heating milk to very high temperatures, typically between 130°C and 150°C, for a brief period of few seconds. This process results in commercially sterile milk, meaning that while not all bacteria are destroyed, those capable of growing under ambient storage conditions are effectively eliminated. An essential component of UHT processing is aseptic packaging, where the sterilized milk is filled into presterilized containers in a sterile environment. This combination ensures that the milk remains free from contamination and maintains its extended shelf life without requiring refrigeration.
Standards for UHT processing vary globally. In the European Union, a minimum temperature of 135°C with an appropriate holding time is mandated. In the United States, the standard for ultra-pasteurization is 280°F (137.8°C) for 2 seconds. Many international standards recommend 135°C for at least 1 second, though some countries do not specify minimum conditions. However, this level of treatment is often regarded as insufficient for completely destroying the spores of spoilage organisms, highlighting the importance of precise temperature-time combinations and the critical role of aseptic packaging in ensuring product safety and stability.

Abstract

Application of ultrasound is one of the most promising uses of novel technologies in food processing. Use of ultrasound at various stages of drying has resulted in significant decrease in drying time and improvement in quality characteristics of dried products. The chapter discusses the principle behind use of ultrasound, general application in food industry with detailed description of various drying processes and equipments assisted by ultrasound.

The cold chain is the lifeline of the ice cream industry because the product must remain continuously frozen from factory hardening until consumption. This chapter examines the complete cold-chain network, beginning with hardening and factory cold storage and extending through CFA or distributor cold rooms, refrigerated transport, retail freezers, and final consumer handling. It explains temperature requirements, monitoring systems, equipment selection, cold-room management, stock rotation, and quality assurance. Particular attention is given to Indian challenges such as power interruptions, inadequate freezer maintenance, poor retailer practices, and temperature abuse. The chapter demonstrates how disciplined cold-chain management protects texture, flavour, shape, overrun, shelf life, and ultimately consumer confidence

The uronic acid pathway is a minor and alternative pathway for glucose oxidation and for the production of uronic acid sugar. Uronic acids of sugars are formed when the terminal carbon’s (C6) hydroxyl group (CH2OH) of sugar gets oxidized to a carboxylic acid. Uronic acid names depend on the name of the parent sugars. Uronic acid of glucose is glucuronic acid, hexose uronic acids are hexuronic acids, and pentose uronic acids are named penturonic acids. The uronic acid pathway is located in the cytosol of the liver and adipose tissue.
The glucuronic acid should not be confused with gluconic acid (aldonic acid) and glucaric acid (aldaric acid or dicarboxylic acid). Gluconic acid (aldonic acid or “onic” acid) is obtained from D-glucose by oxidizing its aldehyde group at C1 to a carboxyl group. Glucaric acid (aldaric or dicarboxylic acid) is formed upon oxidation of both the terminal hydroxyl groups (C1 and C6) and the aldehyde. They have two carboxylic acid groups. This pathway yields pentoses & ascorbic acid or vitamin C (not in primates & guinea pigs) from glucose.

Abstract

The constant improvement in automation of bakery industry has placed higher demands on wheat. As a consequence bakery industry requires wheat possessing specific quality attributes. In view of this, there is need to produce good quality wheat not only for good chapattis but also for better bakery products viz., bread, biscuit, noodles, pasta products. New technologies are emerging which could be exploited for determining the Indian wheats for their suitability for various value added products. In the current investigation, 50 genotypes possessing low protein content (<10.5%) selected from a vast amount of breeding material were analysed for their biscuit making attributes. These lines were screened by actual baking test and alkaline water retention capacity (AWRC) test keeping the standard prescribed for biscuit making qualities viz., low protein content, low sedimentation value, low AWRC and larger diameter with larger spread factor of biscuit. Further with the help of Stable Micro Systems TA.HDi Texture Analyser the biscuits were analysed for hardness and crispness. Correlation coefficient among 8 quality traits suggested highly significant and positive correlation between sedimentation value and AWRC, biscuit diameter and spread factor, and biscuit hardness and fracturability, and significant negative correlation between biscuit diameter and its thickness. On the basis of findings, genotypes identified for biscuit making qualities may be used as donor for developing varieties suitable for biscuit making.

Different basic as well as advanced laboratory equipment need to be used in a laboratory while conducting experiments. Many of them like pH meter, laminar air-flow hood, viscometer, thermometer, magnetic stirrer, microwave ovens, incubators, sand-bath, water-bath, oil-bath, etc. are the basic and simple equipment which require to be used routinely while conducting experiments in a laboratory. However, basic knowledge of their operation, handling procedure, when to be used, how to be used, and more importantly what not to be done with it, etc. are essential for keeping them in serviceable condition as well as for the accuracy/precision in their operation. In this chapter, we will discuss about some of the routinely used laboratory equipment along, while some other sophisticated equipment will be discussed in other chapters.

pH Meter

A pH meter measures exactly the H+ concentration of a solution; thus indicates acidity or alkalinity of a solution with greater precision. Actually, it measures the difference in electrical potential between a pH electrode and a reference electrode, and display the results converted into the corresponding pH value. Electrode is the key part (rod-like structure, usually made of glass) of a pH meter, having a bulb at the bottom containing a sensor. The pH meters range from simple and inexpensive pen-like devices (portable/pocket pH meter) to complex and expensive laboratory instruments with computer interface (Figure 5.1).

Frequent calibration of a pH meter using the solutions of known pH (standard solution of pH 4.01, 7.00, 10.01, Figure 5.2), generally before each use, ensures the accuracy of the instrument. For very precise work the pH meter should be calibrated before each measurement. For normal use calibration should be performed at the beginning of each day. This is necessary because the glass electrode does not give a reproducible measurement over a longer period of time.

Introduction

To accomplish experimentations in laboratories, sometimes we need microscope to test/validate our ideas/findings. A microscope is needed to examine objects which are very small which cannot be visualised by naked eyes. However, microscopic study requires some expertise in operating the microscope, particularly the sophisticated microscopes, as well as to prepare (peeling, sections, cutting, fixation, staining, dehydration, mounting, etc.) the sample, so that it can be appropriately examined for the anticipated details.

According to the scientific requirements different types of microscopes are used in the laboratories for research purpose. In this chapter, our aim is to acquaint you with the basics of using and handling of microscopes, particularly a compound microscope that is commonly used by the graduate and post graduate students for basic research.

Microscopy is often designated as the scientific investigation that is employed to review and analyse minute structures and objects using a microscope. The term “microscope” has been derived from ancient Greek words “micros” meaning “small” and “skopeîn” meaning “to look at or see,”.

Microscope

A microscope is a laboratory instrument used to examine objects (biological specimens) which are too small to be seen by the naked eye. The microscope must accomplish three tasks:

• Magnification- to provide a magnified image of the specimen.

• Resolution- to separate the details in the image.

• Contrast- to render the details clearly visible to the eyes.

Although we will tnroduce to you the different types of microscopes used for different speciments, our main focus is to discuss about compound microscope, which is commonly used in the laboratories. Compound microscope was developed around the beginning of 17th century.

A variety of glassware like burettes, pipettes, measuring cylinders, flasks, separating funnel etc. are used for experimental purposes in a laboratory. All of these need to be used and handled correctly, otherwise the chances of experimental error and damage to the glassware increase significantly. Being made of glass, they need special care while washing and using in experiments because even a minor mistake in handling of glassware may result in breakage of the apparatus. With this precautionary note, let us discuss about some of the specific glassware used in the laboratories.

Burette

Burette is a long graduated glass tube with a stopcock at one end for delivering a known/desired volume of a liquid, especially in titration experiments. It contains a stopcock at the lower end and a tapered capillary tube as the stopcock’s outlet. The flow rate of liquid from the tub stopcock’s outlet can be controlled with the help of stopcock valve. Burette is used for measuring the volume of a solution/chemical added in a quantitative (titrimetric) estimation experiment.

Procedure of using a Burette

The burette reading must be noted before and after delivering the chemical/ solution from the burette. The difference in these two readings is the volume of the chemical/solution released out of the burette. The solution should be released slowly/drop-by-drop to avoid any experimental error/mistake. If the solution/chemical is allowed to pour out too fast, the wall of the burette will not drain properly and some liquid may remain sticking to the surface of the walls. Moreover, excessive amount of chemical may be released/dropped during the experiment, particularly during the titration experiment wherein we need to know the exact amount of acid/base needed to get neutralization point. Thus, any minor mistake may lead to get a faulty reading. The measuring capacity of a burette usually used in a laboratory is 50 mL.

Since mouth pipetting of liquid reagents in a laboratory is not advisable, different tools are needed for safe, easier, and precise dispensing of liquid reagents. This chapter is devoted to make you aware of the tools like vacupette and micropipette which can be used for the handling of liquid reagents in a safe and precise way.

Vacupette

Glass pipettes have been traditionally used by sucking liquid solvent/solution directly using mouth. For safety reasons, mouth pipetting is no longer permitted in a laboratory. Hence, to fill a pipette vacupette is used by placing it on the top of glass or plastic pipette. It acts as a device to create vacuum for filling reagent in the pipette as well as for releasing the reagent with mechanical regulation on the flow out of the liquid. Vacupette can be of different types.

1. Rubber Bulb Vacupette

Rubber bulb vacupette is often used to transfer (pump up and discharge) liquid using a pipette when a larger quantity of chemicals is to be transferred. Small/tiny rubber bulb is attached to glass/plastic pipette for drawing little amount of reagents. Small vacupette is used to precisely discharge smaller volume/ drops.

Handling of Vacupette

• Insert the top of a pipette in the bottom of the rubber bulb vacupette (Figure 6.1).

• Remove air from the pipette by squeezing valve “A” at the top of the vacupette while simultaneously squeezing the rubber bulb. The amount of air released depends on the size of the rubber bulb.

Introduction

It became clear from the discussions of previous chapters that probiotics perform metabolic functions such as fermenting indigestible dietary residues and endogenous mucus, production of vitamins, and absorption of minerals. They also play key role in intestinal epithelial cell proliferation and differentiation, and the homeostasis of the immune system. Further, probiotics are also known to involve in modifying gut pH, antagonizing pathogens through the production of antimicrobial compounds, competing with pathogens for recept or sites as well as for available nutrients and growth factors, stimulating immunomodulatory cells of the host, and produce a wide range of enzymes such as lactase, amylase, lipase, protease, phytase and so on. The molecular mechanisms of probiotics are largely unknown and opens avenue for research in probiotics. Moreover, current research in probiotics and health consciousness in the public lead to an increased interest towards probiotics from the public, researchers, governmental organizations (such as the WHO/FAO) and medicine and food companies. In this chapter, application of probiotics in treating various diseases and disorders are discussed.

Abstract

Drying is an age old practice of food preservation. Removal of moisture from the food materials is usually carried out at atmospheric pressure and elevated temperature. In the presence of oxygen, drying can cause oxidation of some compounds leading to quality deterioration. Drying at higher temperature adds to the losses and can be responsible for enzymatic and non enzymatic browning, loss of aroma, nutrients, antioxidants, pigments etc. On the other hand, drying under sub atmospheric pressure helps in retaining the themosensitive compounds present in natural foods as drying can be accomplished in oxygen free environment. Under vacuum, evaporation of moisture can be possible at lower temperatures.

In monetary terms, value-added’ per unit of the product is the margin between its selling price and cost of production. However, in the dairy and food industry, value addition refers to, extra, features of a product that are beyond the expectations of a consumer at little or no extra cost. In this context, the addition of value to milk can involve monetary value added to a liter of milk by turning it into milk products, enhancing the convenience of quality, shelf life, or health characteristics using processing and packaging interventions.

4.1. Conversion of Milk into Conventional Products

It is an age-old tradition to add value to milk by processing it into milk products. The primary aim was to increase the shelf life of milk that would otherwise be highly perishable. With inadequate facilities to preserve liquid milk fresh in the rural areas’ conversion of milk into a variety of milk products would be an ideal option to effectively preserve the huge quantities of milk procured during the flush season’ In the processing of milk products such as cream, butter, cheese and ghee, by-products such as skim milk, buttermilk, whey, and ghee residue are obtained’ Many of the dairy plants, particularly in India’ are usually confronted with the problem of by-products utilization, especially that of whey and ghee residue in an economical manner. Membrane separation processes have presented new possibilities for the production of newer intermediate products from dairy by-products that can be used for value addition in dairy and food industries.

Abstract

30 lines were selected from 864 germplasm lines for good biscuit making attributes on the basis of low protein content (<10.5%) and low sedimentation value (<40cc). These lines were further screened by actual baking test and alkaline water retention capacity (AWRC) test keeping the standard prescribed for biscuit making qualities viz., low protein content, low sedimentation value, low AWRC and larger diameter with larger spread ratio of biscuit. AWRC had significant negative correlation with biscuit spread ratio. Positive significant correlation was found between biscuit spread ratio and biscuit diameter. On the basis of above findings, it may be recommended that low protein content, low sedimentation value, low AWRC may be used as criteria for better biscuit making i.e. increase biscuit spread ratio and biscuit diameter. So wheat-breeding programme has opened up new opportunities for the end use quality product and wheat based food industry. Hence there is an urgent need for qualitative analysis of desired traits in wheat towards enhanced value addition and thereby an attempted has been made to identify suitable genotypes possessing good biscuit making attributes

Milk, as secreted in the udder of a healthy animal is practically sterile, but during its transit to the processing plant, becomes heavily contaminated with a variety of micro-organisms. These may be responsible for spoilage of milk or for causing diseases in animals and human beings. It thus becomes imperative that adequate heat treatment is given to milk to destroy these micro-organisms, thus increasing the shelf-life of milk as well as making it safe. Varied heat treatments offer the potential of producing different kinds of processed milk to satisfy the needs of the consumer. To cater to the ever-increasing demand of consumers, several varieties of processed milk are available in the market, some of which are discussed below.

5.1. Pasteurised Milk

Pasteurisation was named after the renowned French scientist Louis Pasteur, who discovered that spoilage organisms could be inactivated in wine by applying heat at temperatures below its boiling point. The process was later applied to milk and remains the most important operation in the processing of milk.

It is the process by which every particle of milk is heated to such temperature-time combinations (usually to 63°C for not less than 30 minutes or to 72°C for not less than 15 seconds) and promptly cooled to 4°C or below so as to destroy any harmful bacteria that may be present without affecting flavour and food value. Pasteurisation does not involve the use of any additives. Apart from making milk safe to drink, pasteurisation increases the length of time it can be kept before it spoils. The nutrient losses due to pasteurisation are so small, that they are considered insignificant.

1.3 Dextrose

Sweets which include dextrose have different eating qualities to those made with sucrose and glucose syrup alone. Recipes should be balanced so that the sucrose to dextrose ratio achieves the required product texture.

The incorporation of dextrose will increase the likelihood that crystallization will occur either on store or during manufacture. Replacement usage levels for dextrose normally lie between 5% and 15% of the Sucrose content

Venture Capital

Venture capital refers to financing that comes from companies or individuals in the business of investing in young, privately held businesses. They provide capital to young businesses in exchange for an ownership share of the business. Venture capital firms usually don’t want to participate in the initial financing of a business unless the company has management with a proven track record. Generally, they prefer to invest in companies that have received significant equity investments from the founders and are already profitable. Venture capital investors also prefer businesses that have a competitive advantage or a strong value proposition in the form of a patent, a proven demand for the product, or a very special (and protectable) idea. Startups with innovative ideas and technologies that have a great potential for wealth creation, but a high risk of failure might receive funding from venture capital. Typically, venture financing comes in the form of equity shares or a claim on equity that may arise in the future, such convertible debt, allowing the venture capital firm to acquire a portion of the company. New businesses are often the focus of venture capital, and Georges Doriot, a professor at Harvard Business School, is widely regarded as the “Father of Venture Capital.”

Vitamins are naturally occurring, vital nutrients that are needed in small amounts. They are important for bone and tissue health, wound healing, growth and development, immune system function, and other biological processes. These vital organic substances serve a variety of biochemical purposes. Vitamins, like minerals, are not biosynthesized by the body. Consequently, in order to maintain our health, we must obtain them from the food we eat or, in rare circumstances, supplements.

Vitamins in Milk

Together with water- soluble vitamins like vitamin C and B complex and provitamins, milk also contains a variety of fat- soluble vitamins like A, D, E, and K. Some of these vitamins do, however, have very low concentrations.

Vitamin A (Retinol)

Plant foods contain carotenoids, which are the precursors of vitamin A, whereas animal foods provide preformed vitamin A in the form of retinyl esters.

Introduction

The basic components of diet include carbohydrate, protein, fat, vitamins and minerals. Vitamins are the essential nutrients which are not formed in our body and required in minute amount from the diet. Vitamins are crucial for the optimal functions of several enzymes; thus important for optimal growth and development. Vitamins perform various functions that help in regulation of metabolism, prevent chronic diseases, maintain natural appetite, mental health and immunity.

In 1912, Polish biochemist Kazimierz Funk coined the term ‘vitamine’;a substance that was present in rice polishing and foods that cured polyneuritis in birds and beriberi in humans. The name is from ‘vit’ for vital nutrient and ‘amine’ for its similarity as an amine. Vital signify that these factors are indispensable for life. Earlier they are identified and thought to contain amino groups, thus the word amine was used, which was true for thiamine but later work revealed that most of the vitamins do not have amino groups. Thus the word was shortened to vitamin in English. It should be noted that the term vitamin does not denote to some othern utrients such as essential amino acids, essential fatty acids, dietary minerals or other nutrients that contribute towards good health.

Introduction

A variety of glassware, plasticware, and other apparatus are used for experimental purposes in a laboratory. They need to be washed, cleaned, dried, and sometimes sterilized before they can be used in the next set of experiments. Glassware needs special care of handling during washing, drying, sterilization, etc. because even a minor mistake in handling of glassware may result in its cracking/breakage and it may become unserviceable. All of these may result in a delay in experimentation if spare piece of the glassware is not available or it may affect serviceability of the associated apparatus.

A large number of glassware such as beaker, flask, measuring cylinder, burette, test tube, etc. are regularly used in a laboratory, which requires careful handling during the different stages of experiment as well as after completion of the experiment while preparing them for another set of experimentation.

Glass being a complex silicate, its properties depends on the type of silicate anions in the structure and the cations formed. Thermal properties of glass changes by the addition of boron oxide (B2 O3 ); hence, borosilicate glass is extensively used in the laboratories. With this brief background information, let us discuss about the necessity, procedures, and precautions during washing, cleaning, drying and sterilization of laboratory-wares. Figure 3.1 shows various advantages of using borosilicate glass in a laboratory.

Good laboratory practices require clean glassware, because an experiment may give erroneous result if dirty glassware is used at any step of the experiment. Hence, the glassware must be physically and chemically clean; and in many cases, it must be microbiologically clean or sterile. The commonly used glassware in a laboratory include test tube, measuring cylinder, pipettes, beakers, flasks, microscopic slides, etc.

Abstract

Pulses being nutritionally vital, is an important constituent of diet for a very large number of peoples. A part from being a relatively cheaper source of dietary protein, it is also an important source of calories, certain essential amino acid, minerals and vitamins. Post harvest scenario of pulse shows that 75 per cent of production was milled in pulses with as dehusked split i.e. dal. Traditionally green gram and black gram dal is dehulled manually after soaking in water. Present investigation was carried out to develop the wet type dehuller and study was carried out on water absorption characteristics of green gram and black gram. The result reveals that the dehulling efficiency of abrasion type mechanism was about 95 per cent and 92 per cent in case of shearing type mechanism. It has been observed that rate of water absorption was initially high and then gradually decreases i.e. 0.54 to 0.18 and 0.48 to 0.18 for black and green gram, respectively. 

Introduction

Water is not only a vital environmental factor to all forms of life, but it has also a great role to play in socio-economic development of human population. Since antiquity humans have valued the importance of clean water and have used various techniques to improve its quality. In modern times, the safe drinking water became first a public health issue and then a human rights issue. The access to safe drinkin gwater now considered as a basic human right which is an essential component of effective policy for health protection. Improving access to safe drinking-water can result in tangible as well as intangible benefits to health. It has been shown that investments in water supply and sanitation can yield a net economic benefit, because the reductions in adverse health effects and healthcare costs outweigh the costs of undertaking the interventions (WHO, 2017). Past experiences have also proved that interventions in improving access to safe water favour the poor in particular, whether in rural or urban areas, and can be an effective part of poverty alleviation strategies. The disparity in water availability, reliability, and quality between the developed world and developing countries is enormous. Unfortunately, in many developing countries people lack access to safe water and therefore waterborne diseases are still a major cause of death, especially among children under age of 5. According to a joint report by World Health Organization (WHO) and United Nations International Children’s Emergency Fund (UNICEF) (WHO/UNICEF report 2017), around 2.1 billion people worldwide lack access to safe and readily available water at home, and around 4.5 billion, lack safely managed sanitation facilities. Moreover, many of these countries that have set drinking water standards often lack the resources to implement or enforce them.

Water Requirements

Dairy plant requires large quantities of water on a daily basis for processing and cleaning purposes. Normally the specific water consumption ranges from 1000-5000 litres per 1000 litres of milk processed. The actual requirement can vary between dairies and is a function of 
    a)    Manufacturing process or manufacturing profile.
    b)    Auxiliary processes such as steam generation, design of the refrigeration processes and extent of social services.
    c)    Multiple use/reuse.
Water is a very valuable basic natural resource  and needs careful consumption. The water supply system in a dairy plant should be designed in such a way that consumption for each manufacturing department and auxiliary processes can be determined. This makes it possible to influence water consumption directly, and major losses can be determined very rapidly.

Introduction

In a research laboratory, we frequently need weigh chemicals and prepare different solutions of varying strength, as per the requirements. Accuracy in weighing and preparation of solution, with respect to weight/volume and strength, determines the success of experiment. In turn, the accuracy depends on weighing of solid chemical and/or measuring the volume of liquid chemical/making up the volume. In this Chapter, weighing of chemicals using appropriate weighing balance, preparation of solutions of different strength, their dilution, and precautions while preparing will be discuss in brief.

Weighing of Chemical

The primary method to determine the amount of chemicals to be used is weighing the chemical by using a balance.

In general, there are two types of balances to weigh chemicals in a laboratory:

1. Standard laboratory balance

2. Analytical balance

Standard Laboratory Balance

Generally, a standard laboratory balance is used for weighing a chemical when an error of ±0.02 g is acceptable. It is also used for pre-weighing, where approximate mass needs to be determined. Precise mass of the chemical is then determined using an analytical balance. Following are the steps in using a weighing balance in a laboratory.

Wheat is perhaps best known for its different end-use baking properties as determined by grain texture, protein content and quality. Wheat is generally traded by and classified into “hard” and “soft” market classes on the basis of grain hardness or texture of the wheat kernel (caryopsis). Soft wheats have softer endosperm texture that fractures easily, therefore requiring less energy to mill, yields smaller particles, and has less starch damage after milling when compared to hard wheats. Hard and soft are used with slightly different connotations in different parts of the world. In Western Europe, hard is applied primarily to durum wheats and the soft applied to bread wheat. Elsewhere the term is applied to different cultivars within the species Triticum aestivum. In general, hard wheats are used for bread baking, while soft wheats are used for cookies and cake making. Hard wheat requires more force to crack the kernel, maintains a larger particle size, passes through sieves more easily and has more damaged starch MacRitchie (1980). Higher starch damage is required for good bread making quality Tipples et al. (1994) but is detrimental to the biscuit making quality.
