This chapter focuses on presenting empirical findings from data analysis to provide sufficient interpretation so to draw reliable conclusions. To enhance and complement the textual contents of the study, tables and models are frequently utilized. Additionally, the discussion serves to contextualize the study’s findings and establish connections with the research work.

Result: The Table 13 depicts the distributive characteristics of the variables for the 90 respondents under the study.

Revelation: The pattern of distribution of the variable, Age (x1 ) depicts that the maximum age of the respondents is 70 and the minimum is 26. The mean value of age of the respondents is 42.98 with standard deviation of 10.77. The distribution of the variable is strongly consistent with a CV value of 25.06 per cent.

The pattern of distribution of the variable, Education (x2 ) depicts that the maximum education of the respondents is 5 and the minimum is 2. The mean value of education of the respondents is 4.09 with standard deviation of 0.82. The Coefficient of Variation is 19.96 per cent which indicates the distribution of variable is highly consistent.

7.1 Summary

Climate change is a complex global phenomenon that poses significant challenges for agriculture including uncertainty about future weather patterns, soil conditions, and other environmental factors. Farmers are facing the difficult task of interpreting and responding to these changes in order to maintain their livelihoods and ensure food security for growing populations. In this context, mental modelling has emerged as a useful approach for understanding how farmers interpret and respond to climate change. Mental models are cognitive representations of how individuals understand and make decisions about complex systems, and can be used to identify key drivers of behaviour and decision-making.

The present study has explored the social, ecological, and economic factors that influence farmers’ mental models on perception of climate change, level of uncertainty and trend of agricultural change in the selected study area. It argues that farmers’ mental models are shaped by a range of factors, including their own experiences and observations, social networks and interactions, access to information and resources, and broader economic and political contexts. The present study has also highlighted the role of uncertainty in shaping farmers’ mental models and decision-making. Uncertainty about the future impacts of climate change can create significant challenges for farmers, as they must make decisions based on incomplete information and uncertain predictions.

Despite the meticulous methodology and extensive analysis employed throughout this research project, it is crucial to acknowledge and address the limitations that arose during the study. By recognizing these limitations, we not only ensure a transparent evaluation of the research process but also shed light on potential areas for future investigations to refine and expand upon the findings.

In this section of the chapter, the limitations encountered in this thesis are highlighted as follows:

1. Due to the complexity of the concept, obtaining quantitative responses proved to be exceedingly challenging.

2. The development of improved scales for scoring could have enhanced the research if implemented.

3. The inclusion of a greater number of relevant variables could have provided a more comprehensive and in-depth understanding of the research topic.

4. Deploying mathematical optimization and simulations could have effectively integrated and co-integrated the seemingly disparate aspects of climate change perception, uncertainty perception, and perception of agricultural dynamics.

5. Presently, farmers are hesitant to engage in conversations and provide data to individuals outside their close social circles.

As the present study draws to a close, it opens up a myriad of possibilities for future research and exploration in the field of the present study. The insights gained and the findings presented in this thesis may provide a foundation for building upon and extending the existing body of knowledge. By identifying key areas that require additional attention and offering suggestions for future studies, this section of the chapter aims to inspire and guide researchers in their pursuit of enhancing our understanding and driving progress in this dynamic and evolving field.

The present study leaves behind the following domains which may be researched out in future:

1) The present research can be replicated with varied geo-spatial situations viz., in different locations and settings, with different respondents, with spatio-temporal expansion and methodological innovation.

2) PRA tools and materials can be deployed to collect more specific data to compensate for the inadequate information to go for the classical statistical approaches.

3) Multi-criteria decision analysis (MCDA) techniques can be integrated with mental modelling and fuzzy logic cognitive mapping so to enhance the decision-making process.

4) To realize the full potential of these frameworks, collaboration and knowledge sharing among researchers, practitioners, policymakers, and farmers are essential. Continued research and development efforts are needed to refine and validate the models, enhance user-friendliness, and adapt them to diverse agricultural systems and contexts.

Introduction

Regenerative agriculture is both a philosophy and a strategy for land management. It asks us to consider how all aspects of agriculture are connected through a web, which is a network of entities that grow, enhance exchange, and distribute, consumer products and services. Traditionally, supply chains have been organized linearly. It is about farming and ranching in a way that nourishes people as well as the earth, with specific practices differing from grower to grower and from region to region. This style of farming and ranching is also known as “permaculture.” Although there is no definitive rule book, the holistic principles that underpin the dynamic system of regenerative agriculture are designed to address issues of inequity, reestablish the health of soil and ecosystems, and ensure that our land, waters, and climate are in better condition for future generations. It is a novel concept of uplifting the natural world rather than imposing an invasive control, regenerative agriculture intriguing environmental approach to agricultural production that enables the natural systems to renew themselves. Regenerative agriculture refers to farming practices that, in comparison to industrial agriculture, cause significantly less damage to the surrounding environment and the soil. When we give more to nature than what we receive from it, we bring nature closer to us and bring ourselves closer to nature at the same time. Farming that is in harmony with both the surrounding environment and the community fosters a feeling of integrity and togetherness that helps to strengthen the connection between the two. The concept of harmonious farming can be summed up in a few words: It involves engaging in agricultural practices and cropping systems that increase soil fertility return nutrients to the soil, increase soil water retention, remove carbon dioxide (CO2 ) from the atmosphere, and, most importantly, adapt to the ecosystems in which they are grown.

The term “Integrated Farming System (IFS)” (or simply “integrated farming”) is a term used frequently and generically to describe a method of farming that is more integrated than monoculture methods. By virtue of the intensification of crops and related industries, an IFS offers the chance to enhance economic yield per unit area per unit time. This is accomplished through the system’s use of several crop rotations. It improves the efficiency of fertilizers and natural resources, improves the recycling of nutrients, and contributes to increased food security. The IFS technique promotes ecological intensification by lowering the amount of anthropogenic inputs while simultaneously enhancing the performance of ecosystem services such as the recycling of nutrients, the creation of soil, and the soil’s fertility. IFSs that are well- managed are seen to be safer than those that are not because they benefit from synergy among businesses, diversification of crops, and ecological stability (Behera et al., 2015). It requires paying attention to the minute details and constantly striving to improve in all facets of agricultural production through well- informed administrative practices. The IFS approach is widely regarded as the most important strategy for enhancing the nutritional status of families and ensuring the viability of their livelihoods. The research strategy that is centered on crops and cropping systems needs to give way to the research strategy that is based on agricultural systems, especially for smaller farms (Jha et al., 2011). Recycling of waste and increased effectiveness in terms of land utilization are two essential components of the IFS. The components and enterprises that make up the IFS might differ from region to region. This is because agroclimatic conditions, such as the kind of land, the availability of water, the socioeconomic standing of farmers, and market demand, all play a role. For effective holistic agricultural systems to be constructed, there must be strong connections and complementarities between the various components (Bell et al., 2014). Integrating land- based businesses into the biophysical and socio- economic contexts of farmers, such as aquaculture, poultry, duckeries, apiaries, as well as field and horticultural crops, is essential in the IFS to make farming highly profitable and reliable (Behera et al.,2004: Kashyap et al., 2015). Farmers who are limited in their access to resources can achieve sustainable output through the careful administration of agricultural residues and the distribution of limited resources

One possible way to lessen the effects of global warming on food production is to adopt the sustainable agriculture practice of striking a balance between environmental protection, social justice, and economic viability. The impacts of climate change on farming may be seen all across the world right now. Global food security is threatened by climate change, which is causing higher temperatures, changing precipitation patterns, and increasing extreme weather events. Greenhouse gas emissions, deforestation, and unsustainable farming practices all have a hand in accelerating global warming. Therefore, it is crucial to tackle the double problem of lowering agriculture’s environmental imprint while also adapting it to climate change. Sustainable agriculture works toward a compromise by encouraging robust and flexible agricultural practices. It aims to ensure the sustainability of food production in the future while reducing environmental effects by incorporating climate change adaptation and mitigation techniques into agricultural practices. Agricultural systems can be made more resistant to climate change by the use of sustainable farming practices such as agroforestry, crop diversification, soil conservation, and water management. Sustainable farming practices help communities adapt to climate change, boost local economies, and secure food supplies for future generations.

The traditional methods of food production are coming under increasing strain due to the ever- increasing population of the world and the ever- decreasing availability of land and water resources. The good news is that there are currently concrete solutions to some of these difficulties that can be provided by technology. The fact that climate change is happening while the global population is growing poses the question of how we can ensure that the food we produce is reliable and of consistent quality. Is it possible to grow food in settings that can be regulated, allowing for production that is not dependent on the weather or temperature? It is just a computer enthusiast who is thrilled about how the industry is evolving, but maybe that makes it sound like a visionary. Over the course, gathering a significant amount of expertise working on initiatives that include the use of technology towards horticulture concerns is important. In order to keep newly emerging pests and diseases under control, horticultural crops require a higher degree of personal attention and plant monitoring than arable crops. The Importance of Adoption of Technologies

Recently, the necessity to improve production, earnings, and productivity was the primary driving force behind the choice of technology that was made available to farmers. The primary obstacles included a lack of access to cash, a lack of familiarity with how to use the technology, and exposure to market risks, the latter of which was mitigated by the policies of the government in many countries. In the past, “good policy practices” were therefore rather straightforward, relating primarily to increasing output, and the goal of agricultural and horticulture policies was to increase productivity in agriculture and horticulture. This was the case because in the past, “good policy practices” were related to increasing output. The simultaneous cultivation of two or more crops in the same field is known as intercropping. Sustainable agriculture aims to at least build agricultural systems that are inspired by nature. In nature, cooperation predominates over competition. Mutually beneficial interactions between species within groups are one example of cooperation. A few types of creatures may initially inhabit an abandoned agricultural field if it is left unattended and unplanted, but over several years, a complex community made up of numerous wild species emerges. When a community reaches a high level of variety, it becomes stable. Diverse communities are stable and have fewer swings in species abundance. Enterprise diversification, crop rotation, the use of windbreaks, the provision of more habitats for microorganisms, intercropping, and the integration of crop farming with animal production are methods that increase diversity and stability on the farm (Reddy and Willey,1981; Reddy et al., 1992). Each crop should have enough room while growing with another one or more to enhance cooperation and minimize competition. This is made possible by the elements of spatial layout, plant density, crop maturity dates, and plant architecture. The spatial arrangements are Row intercropping— Growing two or more crops together at the same time with at least one crop planted in rows; Strip intercropping— Growing two or more crops together in strips wide enough to separate crop production using machines, but close enough to interact; Mixed cropping— Growing two or more crops together in no distinct row arrangement; Relay intercropping— Plant a second crop into a standing crop at a time when the standing crop is at its reproductive stage but before harvesting.

Nowadays, horticultural fields are expanding due to the increasing demand for various utilizations. Increased production is leading to an increase in exports. The global horticulture market value of about 20.77 billion USD was estimated in 2021 and targeted to attain 40.24 billion USD by 2026 (Global Market Estimates, 2021). From the production of horticultural foods, various factors are involved in the hazardous environmental appearance. Uncontrolled use of pesticides and residual effects of various chemicals initiate the primary threats to nature by creating a negative impact on soil, water, humans, animals, and wildlife species too (Sanchez-Bayo, 2011). Alone agricultural practices are not responsible for the production of horticultural wastes some other aspects are also included such as horticultural food processing industries as well as urban and peri-urban wastes. Food processing is an important sector of agriculture. Being highly perishable horticultural food processing became more necessary to increase the self-life as well as the value addition of these crops. But at the same time, these food processing industries where several processes such as production, processing, and preparation of food take place again result in the generation of huge quantities of waste material causing health hazards due to environmental pollution. The huge waste of food is due to a lack of control and ignorance of higher officials in this agro-economic sector. Another important source where waste management is equally important is Urban and peri-urban areas. Being a sink for horticultural produce (raw or processed) creates huge waste at the same time. The urban population is increasing between 3% and 3.5% per annum and the yearly increase in waste generation is around 5% annually (Agarwal et al., 2005).

Preservation and promotion of a wide variety of plant and animal life on and around farms is an aim of both organic and regenerative farming practices. Pest and disease management, pollination, and ecosystem stability are all bolstered by biodiversity. Both methods encourage using less or no man- made chemicals, such as those found in pesticides, herbicides, and fertilizers. Organic matter, compost, and biological pest management are just some of the natural inputs that these farmers rely on. Water, energy, and material resource conservation are fundamental to both organic farming and regenerative agriculture. To lessen agriculture’s impact on the environment, they advocate for methods including water management, efficient irrigation systems, and the use of renewable energy. Strong relationships and collaborations between farmers, customers, and local communities are prioritized by these methods. All along the agricultural supply chain, they are committed to fair trade, social equity, and transparency. Farmers may grow healthy, resilient, and sustainable farming systems by applying the holistic concepts of regenerative agriculture to conventional organic farming practices. In addition to strengthening the bond between agriculture and its natural surroundings, these methods also improve soil quality, lessen negative effects on the environment, increase biodiversity, and result in more nutritionally dense food.

Introduction

Farmers have long used biorationals such as Indigenous Technical Knowledge (ITK) in both crop production and crop protection. This understanding is the result of many generations of interaction within natural and physical environments (Rajasekaran et al., 1991 and Kolawole, 2001). ITK is the unique and local traditional knowledge that is specific to a particular culture or community and is passed down from one generation to the next and every individual has inherited this information from their ancestors. Except for a handful, ITKs are passed down from generation to generation verbally, with no validated written documents. Many definitions of ITK systems have been proposed, but all of them are incomplete because the notion is still relatively young and changing. The ITKs are environmentally friendly and compatible with pest management approaches (Deka et al., 2006). ITK was defined by Wang (1988) as “the whole body of knowledge and practices based on people’s cumulative experiences in resolving situations and issues in a variety of spheres of life and such information and practices are unique to a specific culture.” According to Haverkort, 1995, ITK is the actual knowledge of a given population that reflects the experiences based on tradition and includes more recent experiences with modern technologies. The phrase “indigenous technological knowledge” is frequently disguised with the idea that it is connected to upcoming events and the innovations developed by farmers to address certain issues. Among the associated terms are (i) Indigenous Knowledge (IK), which refers to the participant’s understanding of their social and temporal context. The term “indigenous knowledge” as used here refers to any identifiable community’s knowledge, not just which of indigenous peoples; (ii) The Indigenous Knowledge System (IKS) describes a conceptual framework for theories and perceptions of nature and culture. The physical, ecological, social, economic, and ideational surroundings are thus included, together with their definitions, categorizations, and concepts. The cognitive and empirical levels both have a role in IKS dynamics. IKS is evident in institutions, artifacts, and technology on an empirical level. ITK is expressed through stories, songs, folklore, proverbs, dances, myths, cultural values, beliefs, rituals, community regulations, local language and taxonomy, agricultural techniques, tools, materials, plant species, and animal breeds. ITK is stored in people’s memories and activities, and is expressed in the form of stories; ITK is shared and communicated orally, by specific examples, and through culture. An African proverb says “When an old knowledgeable person dies, a whole library dies” indicating the importance of ITKs. It serves as the foundation for local decisions in rural communities on agriculture, health care, food preparation, education, and a variety of other activities. Indigenous knowledge serves as a society’s information hub and helps in decision making and communication. Indigenous information systems are dynamic and constantly affected by both internal innovation and experimentation and interactions with outside systems. The phrase “indigenous technological knowledge” is frequently disguised with the notion that it is connected to upcoming events and the innovations produced by farmers to address certain issues.

For many years, it has been acknowledged that “augmentative” or “inundative” biological management can decrease arthropod pests (DeBach, 1964). The goal of increased biocontrol is to bring about behavioral and numerical changes in the natural enemies that live within agricultural environments. This tactic calls for the large production of natural enemies as well as their periodic release, to maximize their potential for reproduction during the growing season. For instance, the use of augmentative releases may be required if naturally occurring populations of parasitoids or pests are either nonexistent, do not successfully colonize fields or orchards, or colonize at a point in the season that is too late to provide effective pest control. The increase is being carried out to establish a population of natural enemies capable of keeping the number of pests at economically hazardous levels until harvesting time.

Biological Control

Controlling problems caused by things like mites, insects, weeds, and plant diseases with the help of other organisms is called biological control or biocontrol. It uses natural mechanisms like predation, parasitism, herbivory, and so on, but human intervention is almost always present.

Parasitoids

The term “parasitoid” refers to a group of insects, the larvae of which develop on or within the bodies of other insects, which the parasitoid kills eventually. The phenomenon is known as parasitism. The vast majority of parasitoids are classified as either wasps (Hymenoptera) or flies (Diptera). These insects are numerically quite abundant and play a significant role in almost every terrestrial environment.

One of the most important concerns confronting both developed and developing nations in this age of rapid industrialization is how to deal with the ever-increasing volume of solid waste. One aspect of the environmental crisis that occurs alongside the development of the global economy is the rapid increase in waste volume (Zibres et al., 2011). The threat that organic waste poses to both the natural world and people’s health in today’s modern society makes it a primary topic of investigation in the modern community. The annual production of agricultural waste across the globe is greater than hundreds of megatons, and a significant portion of this waste is either improperly disposed of or burned directly, which contributes further to the acceleration of both global warming and air pollution (Zhang et al., 2016).

The compost that is produced through the biological process of composting can be used as an organic fertilizer in agriculture, vegetable gardens, and other types of gardens. Composting is a biological process that acts on the transformation of the organic matter that is present in the residues into material that is humidified (Antunes et al., 2021). In addition to having low production costs, this method is also environmentally friendly, making it a viable option for the long-term processing of this waste (Lopez-Gonzalez et al., 2015). Composting processes that are mediated by invertebrates from the soil fauna, such as vermicomposting and milli-composting, are proposed along with a variety of mixtures, practices, and management to improve the efficiency of composting as well as the quality of the compost produced. Traditional composting relies heavily on the activity of microorganisms. Vermicomposting and milli-composting are two examples of composting processes that use invertebrates from the soil fauna. Milli-composting is a relatively unknown biotechnology that uses diplopods, more commonly known as millipedes, to increase the biotransformation of vegetable wastes into stable organic matter. It not only aids in climate change mitigation but also aids in carbon sequestration, so milli-composting can be used as an alternative to modern composting as it is crucial for preventing pollution and promoting lush plant life.

In the last two decades, sustainable agriculture has grown to become one of the most pressing issues in agriculture. Although there are numerous definitions of sustainable agriculture, most agree on three fundamental overlapping components: Ecological, economic, and social sustainability (Kaur 2013; Pilgeram 2013). Meanwhile, the prevalence of plant diseases is a major impediment to sustainable agricultural production systems worldwide, particularly in the tropics and subtropics. Farmers frequently apply agrochemicals (i.e., pesticides, insecticides, fungicides, herbicides, etc.) over their recommended dose to manage plant diseases, raising serious concerns about food safety, soil, environmental quality, and pesticide resistance (Dordas 2008; Kaur 2013).

Disease detection on plants is critical for sustainable agriculture, and manually monitoring plant diseases is difficult. It usually requires a significant amount of work, knowledge of plant diseases, and extended processing time. Early detection is the base for efficient plant disease prevention and control, and it is critical in agricultural production management and decision-making. Plant disease identification has become increasingly important in recent years (Li et al., 2021). Diseased plants usually have visible marks or lesions on their leaves, stems, flowers, or fruits. In general, each disease or pest condition has a distinct visible pattern that can be used to identify abnormalities. Typically, the leaves of plants are the primary source for identifying plant diseases, and most disease symptoms may first appear on the leaves (Ebrahimi et al., 2017). Rapid advances in biotechnology have resulted in the development of various molecular diagnostic tools over the last decade. These tools, which are based on the properties of nucleic acids (DNA or RNA) or proteins of the target agents, have improved the efficacy, accuracy, and speed of detecting and identifying disease-causing agents, as well as the characterization of a wide range of pathogens and pests. Various techniques are used in biotechnology for disease diagnosis and management, but molecular markers are used in particular for the detection, identification, quantification, and characterization of plant pathogens that cause diseases in plants (Kumar et al., 2015).

Regenerative agriculture is an approach to farming and land management that focuses on restoring and improving the health of ecosystems and the soil (Latacz and Nuppenau. 2019). It goes beyond sustainable agriculture by actively seeking to regenerate and rejuvenate the natural resources utilized in agricultural practices (Lal, 2018). The goal of regenerative agriculture is to create a resilient and sustainable farming system that enhances biodiversity, improves soil health, sequesters carbon, and promotes overall ecosystem health (Paustian, et.al., 2018). Regenerative agriculture encompasses a range of practices and principles that vary depending on the specific context and agricultural systems (Smith et. al., 2015).

Regenerative Agriculture often include:

• Conservation tillage: Minimizing soil disturbance to maintain soil structure, prevent erosion, and preserve beneficial microorganisms.

• Cover cropping: Planting cover crops in between cash crops to protect the soil from erosion, improve nutrient cycling, and enhance soil fertility.

• Crop rotation and diversification: Alternating crops and incorporating diverse plant species to improve soil health, break pest cycles, and optimize nutrient use.

• Composting and organic matter management: Adding organic materials to the soil to improve its fertility, water-holding capacity, and nutrient availability.

• Managed grazing: Employing rotational grazing strategies to mimic natural grazing patterns, improve soil health, and promote plant growth.

• Agroforestry: Integrating trees, shrubs, or other perennial plants into agricultural systems to provide additional ecosystem services, such as shade, windbreaks, and habitat for beneficial organisms.

The growing interest in agricultural production over the past few decades has resulted in more farming systems that rely on chemical pesticides or chemical fertilizers, which has seriously harmed the soil, water, air, and climate while paying little attention to quality or concern for the agricultural system’s negative effects on ecosystems. But as time went on, scientific research began to focus on sustainable agriculture, organic farming, clean agriculture, and other ideas that eventually evolved into agricultural systems that maintain the balance in all areas of the agricultural and production process, as well as consumer and environmental concerns. These agricultural approaches have attracted the attention of numerous farmers, and many large farms are now interested in organic farming in the context of sustainable agriculture. The idea of sustainable agriculture is simply the production of food, fiber, other plant products, or even animal products using agricultural techniques that protect the environment, public health, and human societies from the detrimental effects arising from carrying out that process using the standard format as well as technology that also takes into account animal welfare, such that this type of agriculture will enable us to produce healthy foods without compromise (Bahlai et al., 2010; Muller et al., 2017, and Palmgren et al., 2015).

The Future of Sustainable Agriculture

The adoption of sustainable agriculture was made to raise awareness of their significance and to support the governments of plantations and businesses that operate within the parameters of sustainability, in particular organic agriculture, which is clean and recently devoted a lot of studies and research as well as major scientific conferences on sustainable agriculture, as were a lot of major exhibitions for organic goods and food made from organic ingredients around the world. Specifically, the potential role that organic agriculture could play in reducing the adverse effects of climate change, reducing deforestation rates, maintaining biological risk, and also arranging the steady increase in the world’s population, attracted attention to the level of international policy.

Introduction

The soil is an invaluable resource that sustains life in innumerable ways. It provides a dynamic foundation for agriculture, supporting growing crops, and serving as a habitat for organisms. It plays a crucial role in maintaining biodiversity. Additionally, soil acts as a filter, purifying and storing water, and it reserves essential plant nutrients. however, soil resources face various threats that endanger their health and productivity. Soil erosion, the removal of the top layer of soil through natural or human-induced processes, poses a significant challenge. It not only leads to a loss of fertile soil but also causes widespread environmental degradation, and reduced agricultural productivity in response to this difficulty, soil conservation emerges as a principal priority. Soil conservation incorporates a range of practices and strategies to prevent soil erosion, and promote sustainable land use. By adopting and implementing effective soil conservation measures, we can safeguard this irreplaceable resource for future peers and ensure food security and environmental sustainability.

The main highlight of the study is to shed light on the causes and consequences of soil erosion, exploring different conservation practices and techniques; also to understand the vital processes and factors contributing to soil erosion, including wind and water erosion. Additionally, to study its impact on soil and crop production and strategies for different conservation practices to mitigate erosion, preserve soil fertility, and ensure the long-term sustainability of our agricultural systems and ecosystems.

Introduction

According to research released not too long ago by the United Nations, the population of the entire planet is expected to reach around 8 billion by the end of the year 2022. As a consequence, the future demand for the production and distribution of food must be satisfied to provide sustenance for the expanding population of the world and to alleviate human suffering. However, in countries like India, food is wasted at an alarmingly high rate every year because there are insufficient facilities for cooling and storing food once it has been harvested. The majority of the losses began on the farm level since the fresh produce, which requires fast refrigeration after harvest in large quantities, was unable to be given, which led to the spoilage of the food. It is especially vital to shorten the amount of time between harvesting and first cooling to slow down the rate at which newly harvested fruits and vegetables breathe out carbon dioxide and exhale it. When keeping fresh fruits and vegetables, the two most important controlling parameters are temperature and relative humidity. These two factors work together to minimize the amount of water that is lost and to keep the product quality from deteriorating. Following their preferred ranges of temperature and relative humidity, fruits and vegetables can be divided into the following categories: (Table 15.1). For the objectives of on-farm cooling, the majority of fresh fruits and vegetables that are classified as belonging to groups 1 and 2 need to be concentrated, which necessitates maintaining an ideal temperature of less than 10°C and a relative humidity of more than 90%.

Landscaping is an expanse of scenery, a great way to add beauty, and curbs appeal to the place. The urban landscaping designs the outdoor and indoor places marveling the beauty of nature, i.e., the warmth, breeze, sunrays, shade, and the cool climate. Sustainable Landscaping can be defined as the designing of a garden considering the environmental issues at hand and landscaping in such a way that the designs, the planting materials, and the management are in balance with the locality with minimal resources. According to Loehrein (2013), organic gardening practices are integral to the design, construction, and maintenance of sustainable residential and commercial gardens.

Precision sustainable gardening, which involves applying specific amounts of care and resources to individual plants at precisely the right time and in the right place, is a form of precision farming. The local community, the environment, and the health of birds, bees, and other wildlife can all benefit immediately from a sustainably landscaped yard. Xeriscaping, grasscycling, erosion control, water conservation, carbon sequestration, habitat creation, mulching, and composting are all practices that contribute to more sustainable and climate-appropriate landscapes (Kamakshi, 2022).

Seed technology improves seed genetics and physiology. Variety creation, release, seed production, processing, storage, and certification are included. (Feistritzer, 1975). Organic farming began in the early 20th century due to constantly changing farming practices. Over 50 million hectares of certified organic agriculture are in Australia. “That discipline of study having to do with seed production, maintenance, quality and preservation” is seed technology. Organic seeds are fully produced with organic matter practices made with proven organic function and natural seed production, including seeding plants, with a set of guidelines prohibiting the use of manufactured products or chemicals to be sold as such. They must be produced by an accredited institution by an authorized person government agency. Organic seed is created by a planted crop and organically raised at least one generation for annual crops and two generations for annual and perennial plants. Organic farming is sustainable and eco-friendly. Farming needs better norms and standards to achieve biological goals. It is usually cold and people need lots of vegetables but have few organic, clean supplies. Organic farming in India needs a certain space and high-yield vegetable crops in national marketplaces to produce high-quality products. Indian agriculture uses chemical fertilizers, which can be minimized. Experiments have shown that it works well in conjunction. Inorganic and biological fertilizers boost yields, soil quality, and productivity.

The current science and technology is highly developed and everything is at our fingertips. The drudgery that was involved earlier while doing any kind of activity is so much reduced at present. The use of technology is so important nowadays that it has become unavoidable in all fields. With the tools and techniques, most of the problems can be solved and so technology has become a part of our lives thereby making our lives better. Some of the trending technologies are Artificial Intelligence (AI), robotic process automation, 5G, Information and Communication Technology (ICT), Machine Learning (ML), Virtual Reality (VR) etc. With the application of all these technologies, every discipline whether it is infrastructure, medical science, transport, communication, etc. has become stronger than ever. Even the field of agriculture has become so advanced with these technologies. The enhancement of agricultural activities has become so important with the depletion of natural resources, less human labor, and the changing climatic conditions. All over the world agricultural technologies are adopted, disseminated, and practiced to increase production and productivity, improve livelihood and alleviate poverty (Kilima et al., 2013). Similarly, the main reason for the growing demand for ICT in agriculture is for the farmers to get up-to-date knowledge and apply the tools and techniques to improve their livelihood. However, the ultimate aim is to adopt sustainability in agriculture. Through ICT, farmers can get updated information on the market prices, weather forecast, emergence of pest and diseases and their management practices, planting materials, varieties, etc. This knowledge not only gives the farmer an efficient method of farming for more production but also empowers them with modern agricultural ways. The use of ICT in combination with other techniques enables the farmer to save time, use resources efficiently, and produce better quality products.

Introduction

Water retention is an important soil hydraulic property that affects how soil operates in an ecosystem and how it is managed. Hydrology, agronomy, meteorology, ecology, environmental protection, and many other soil-related professions employ soil water retention data. Some soil survey programs measure water retention. A soil survey can estimate water retention from other soil properties. However, these measurements are problematic during project design and in large-scale implementations (Rawls et al., 1991; Wo sten et al., 2001) since agricultural land needs rise periodically. Agriculture is using dry land. Food self-sufficiency and becoming a world food barn by 2045 necessitate agricultural land expansion (Mulyani and Agus, 2017). Water concerns afflict farming in this agroecosystem (Syam et al., 1996; Mulyani and Agus, 2017). Because of soil composition, much water stays in the earth. Tang et al. (2022).

For plant growth, soil water retention variables indicate how well the soil holds water. The dirt provides most of the plant’s water. Water in root layer dirt pores is absorbed by plants. Thus, soil water retention is the key factor affecting plant development and productivity. Plants can take in the soil pores’ water between the field capacity and the point where they always wilt. Field capacity occurs when the matrix potential and gravitational potential match. The most significant component for plant growth is soil water (Tang et al., 2022, Deng et al., 2016, Su et al., 2020). The soil’s physical features determine plant water availability. The amount of water available depends on clay minerals, organic matter, absorbed cations, soil texture and structure, and water availability. Thus, to prevent plants from experiencing water or groundwater stress, soil physical attributes must be investigated to determine groundwater availability (Kirkham 2023). Infiltration is crucial to groundwater availability. The amount of water that can penetrate the soil depends on its physical features, such as soil texture, organic matter, bulk density, porosity, aggregate stability/stability, and water content. Thus, soil water availability depends on its physical qualities, particularly its ability to absorb and transfer water. The physical properties of saturated soil affect how water travels through it, which affects runoff and infiltration (Sauer and Logsdon 2002).

The changing climate and its variability are a big concern for the human being. The recurrent droughts and floods, which are the results of this changing climate seriously hamper the livelihood of billions of people who depend on land for most of their needs. According to the United Nations Framework Convention on Climate Change (UNFCCC) (UNFCC; 1992), “climate change is the change that can be attributed directly or indirectly to human activity that alters the composition of the global atmosphere and which is in addition to natural climate variability observed over comparable time periods” The Intergovernmental Panel on Climate Change (IPCC report, 2022) issued a cautionary statement regarding the imminent attainment of the 1.5oC threshold by the global climate system within the forthcoming 20-year period. It further emphasized that only implementing exceedingly stringent reductions in carbon emissions, commencing at present, would serve as a viable measure to avert an impending ecological catastrophe.

Droughts: A time in which circumstances are significantly drier than average is referred to as a drought.

Floods: A flood is an abnormally large outpouring of water (or, on very rare occasions, other fluids) that covers normally dry ground.

Livelihoods: The term “livelihood” refers to a collection of activities that are carried out during one’s lifetime and are considered necessary to one’s day-to day existence. Obtaining necessities such as water, food, fodder, medication, shelter, and clothing are examples of such activities.

This chapter delves into the profound significance of integrating livestock with cropping systems and grasslands in modern agriculture, exploring how this synergistic combination enhances resource utilization, promotes resilience, and fosters a thriving agricultural sector.

The roots of integrated farming practices can be traced back to ancient agricultural traditions, where farmers instinctively recognized the advantages of combining crop cultivation and animal husbandry (Devendra and Thomas, 2002). Over time, this integrated approach evolved and adapted to different cultural and geographical contexts. In traditional mixed farming systems, livestock provided essential services such as draft power (Gomiero et al., 2011), manure for fertilization, and protein-rich food. As agriculture progressed with technological advancements, there was a shift toward specialized monoculture and intensive livestock farming (Herrero et al., 2013). However, the unintended consequences of these systems, including soil degradation, environmental pollution, and declining rural livelihoods, paved the way for a resurgence of interest in integrated farming methods (FAO, 2020).

The process of recycling involves the conversion of waste materials into reusable products or materials and aims to reduce the consumption of fresh raw materials, minimize energy use, and mitigate trash creation. The primary objective of recycling is to effectively preserve finite resources, safeguard the natural environment, and mitigate the volume of garbage deposited in landfills or subjected to incineration (Lienig et al., 2017). Recycling refers to the systematic transformation of discarded materials into fresh resources and tangible products. The aforementioned term often encompasses the process of extracting energy from waste materials. The recyclability of a material is contingent upon its capacity to regain the characteristics it has in its initial form. There exists an alternative approach to garbage disposal that has the potential to save resources and mitigate the release of greenhouse gases. Additionally, it can mitigate the waste of potentially valuable resources and diminish the use of pristine raw materials, therefore reducing energy consumption, air pollution resulting from incineration, and water pollution stemming from landfill practices. The promotion of environmental sustainability is achieved by the removal of raw material input and the redirection of waste output within the economic system. The increasing volumes of trash have given rise to several environmental challenges, including air pollution, water pollution, soil contamination, and biodiversity loss. The emission of diverse greenhouse gases from solid waste landfills also contributes to the phenomenon of global warming. At now, the predominant techniques for solid waste disposal are open dumping and landfilling. Various alternative techniques, such as pyrolysis, gasification, anaerobic digestion, composting, and bio-methylation, are used to extract valuable resources from solid waste materials (Griffin et al., 2018).

Reviving the agricultural economy is a crucial endeavor with far-reaching implications for food security, economic prosperity, and sustainable development. Several key strategies can be adopted to achieve this goal. Promoting sustainable agricultural practices is vital to ensure long-term viability. Encouraging the adoption of eco-friendly methods such as organic farming, crop rotation, and water-efficient irrigation can enhance environmental resilience while safeguarding natural resources for future generations. Empowering farmers through training and education is essential. Equipping them with the latest knowledge and techniques enhances their capacity to adapt to changing conditions and make informed decisions about crop selection, pest control, and market opportunities. Furthermore, enhancing market linkages is critical to connect farmers with consumers and ensure fair prices for agricultural produce (African Development Bank, 2022). Developing efficient supply chains and reducing post-harvest losses can improve income prospects for farmers and attract more youth to the sector. Lastly, encouraging private sector involvement and investment in agriculture can accelerate growth and innovation. Public-private partnerships can create opportunities for technological advancements, value addition, and market expansion (Liu & Lee, 2018). Reviving the agricultural economy requires a collaborative effort involving governments, farmers, the private sector, and civil society. By addressing challenges, investing in modernization, and promoting sustainable practices, nations can create a thriving agricultural sector that contributes to food security, economic development, and environmental sustainability (Brown, 2021).

The term Information and Communication Technologies (ICT) encompasses a broad spectrum of electronic tools that converge to facilitate information processing, communication, transmission, and display. Scholars like Michiels and Van Crowder (2001) define ICTs as a constellation of electronic technologies capable of transforming organizations and reshaping social relations. In the context of agriculture, the integration of ICT holds significant promise, offering a myriad of advantages ranging from accelerating agricultural growth to empowering marginalized farming communities. This review examines the need, catalytic role, advantages, scope, and limitations of ICT in agricultural extension, elucidating its potential and challenges in promoting sustainable rural development.

Need for ICT in Agriculture Extension

1. Accelerating Agricultural Growth: ICTs play a pivotal role in catalyzing agricultural development by providing timely access to critical information, knowledge, and resources.

2. Expanding Knowledge Resources: ICTs serve as repositories of agricultural knowledge, enabling extension workers and farmers to access comprehensive information on best practices, innovations, and market trends.

3. Facilitating Information Services: ICTs enhance the dissemination of agricultural information and advisory services, enabling farmers to make informed decisions on crop management, pest control, and market opportunities.

For proper and efficient experiment to be conducted, experimenters need so many chemicals to deal with. This includes different acids, base, salt, different kind of indicators and so on. Some of these chemicals are safe for health, but some of them are dangerous to health when come in close proximity to our body while some of the chemicals cause health issues on inhaling. These may cause improper functioning of some of our physiological activities. Acids and bases are very dangerous for our tissues when come in our contact. These may cause serious respiratory problem in addition to injuries on coming to our contact. Some chemicals are exploratory yet they may cause even death of the person working with them in laboratory. Therefore, mere use of the chemicals in laboratory is not only the objective of the experimenter. The person working in laboratory should also be concerned about the proper use and handing of the chemical while working with it. Thus, every researcher of an institution who has to work with chemicals in a laboratory should be made aware about the chemicals to be used in the laboratory and the consequences if any mishandling of a laboratory chemical occurs. Not only these, suitable precautionary measures should also be known to the researcher along with ensuring the availability of the required safety equipment/installations. Hence, a proper understanding of the ways of handling of chemicals in a laboratory is of prime importance for the personals like you who are working or have to work in laboratory.

Safe Handling of Chemicals in Laboratory

Safe use/handling of chemicals in laboratory include minimized exposure to chemicals, maintaining safety data sheets, proper labelling, storage, segregation, and transport of chemicals for personal hygiene.

Introduction

While experimenting with chemicals, sometimes we need to dry the chemical/ solution (solid, liquid or gas) using a process like evaporation or distillation, mainly to remove water or different solvents. Drying is the process of evaporation or distillation to remove water/solvent from a solution, solid or even from gas. Generally, a drying agent (usually an anhydrous inorganic salt) is used for this process, which reacts with the water (present in the chemical/ solvent to be dried) to form a hydrate. For example, anhydrous MgSO4 is one of the drying agents. The drying agents are recognized by their capacity (the rate at which they absorb water) and their intensity (the amount of water left behind in the solvent at equilibrium). No chemical reaction or change in phase occurs during the drying process.

Type of Solvents

Apart from water (a universal solvent), there are three families of organic solvents used in laboratories.

1) Oxygenated solvents: Alcohols, Ketones, Glycol ethers, Glycol esters, etc.

2) Hydrocarbon solvents: Aliphatic and Aromatic hydrocarbons.

3) Halogenated solvents: Chlorinated hydrocarbons.

Introduction

A variety of glassware, plasticware, and other apparatus are used for experimental purposes in a laboratory. They need to be washed, cleaned, dried, and sometimes sterilized before they can be used in the next set of experiments. Glassware needs special care of handling during washing, drying, sterilization, etc. because even a minor mistake in handling of glassware may result in its cracking/breakage and it may become unserviceable. All of these may result in a delay in experimentation if spare piece of the glassware is not available or it may affect serviceability of the associated apparatus.

A large number of glassware such as beaker, flask, measuring cylinder, burette, test tube, etc. are regularly used in a laboratory, which requires careful handling during the different stages of experiment as well as after completion of the experiment while preparing them for another set of experimentation.

Glass being a complex silicate, its properties depends on the type of silicate anions in the structure and the cations formed. Thermal properties of glass changes by the addition of boron oxide (B2 O3 ); hence, borosilicate glass is extensively used in the laboratories. With this brief background information, let us discuss about the necessity, procedures, and precautions during washing, cleaning, drying and sterilization of laboratory-wares. Figure 3.1 shows various advantages of using borosilicate glass in a laboratory.

Good laboratory practices require clean glassware, because an experiment may give erroneous result if dirty glassware is used at any step of the experiment. Hence, the glassware must be physically and chemically clean; and in many cases, it must be microbiologically clean or sterile. The commonly used glassware in a laboratory include test tube, measuring cylinder, pipettes, beakers, flasks, microscopic slides, etc.

A variety of glassware like burettes, pipettes, measuring cylinders, flasks, separating funnel etc. are used for experimental purposes in a laboratory. All of these need to be used and handled correctly, otherwise the chances of experimental error and damage to the glassware increase significantly. Being made of glass, they need special care while washing and using in experiments because even a minor mistake in handling of glassware may result in breakage of the apparatus. With this precautionary note, let us discuss about some of the specific glassware used in the laboratories.

Burette

Burette is a long graduated glass tube with a stopcock at one end for delivering a known/desired volume of a liquid, especially in titration experiments. It contains a stopcock at the lower end and a tapered capillary tube as the stopcock’s outlet. The flow rate of liquid from the tub stopcock’s outlet can be controlled with the help of stopcock valve. Burette is used for measuring the volume of a solution/chemical added in a quantitative (titrimetric) estimation experiment.

Procedure of using a Burette

The burette reading must be noted before and after delivering the chemical/ solution from the burette. The difference in these two readings is the volume of the chemical/solution released out of the burette. The solution should be released slowly/drop-by-drop to avoid any experimental error/mistake. If the solution/chemical is allowed to pour out too fast, the wall of the burette will not drain properly and some liquid may remain sticking to the surface of the walls. Moreover, excessive amount of chemical may be released/dropped during the experiment, particularly during the titration experiment wherein we need to know the exact amount of acid/base needed to get neutralization point. Thus, any minor mistake may lead to get a faulty reading. The measuring capacity of a burette usually used in a laboratory is 50 mL.

Different basic as well as advanced laboratory equipment need to be used in a laboratory while conducting experiments. Many of them like pH meter, laminar air-flow hood, viscometer, thermometer, magnetic stirrer, microwave ovens, incubators, sand-bath, water-bath, oil-bath, etc. are the basic and simple equipment which require to be used routinely while conducting experiments in a laboratory. However, basic knowledge of their operation, handling procedure, when to be used, how to be used, and more importantly what not to be done with it, etc. are essential for keeping them in serviceable condition as well as for the accuracy/precision in their operation. In this chapter, we will discuss about some of the routinely used laboratory equipment along, while some other sophisticated equipment will be discussed in other chapters.

pH Meter

A pH meter measures exactly the H+ concentration of a solution; thus indicates acidity or alkalinity of a solution with greater precision. Actually, it measures the difference in electrical potential between a pH electrode and a reference electrode, and display the results converted into the corresponding pH value. Electrode is the key part (rod-like structure, usually made of glass) of a pH meter, having a bulb at the bottom containing a sensor. The pH meters range from simple and inexpensive pen-like devices (portable/pocket pH meter) to complex and expensive laboratory instruments with computer interface (Figure 5.1).

Frequent calibration of a pH meter using the solutions of known pH (standard solution of pH 4.01, 7.00, 10.01, Figure 5.2), generally before each use, ensures the accuracy of the instrument. For very precise work the pH meter should be calibrated before each measurement. For normal use calibration should be performed at the beginning of each day. This is necessary because the glass electrode does not give a reproducible measurement over a longer period of time.

Since mouth pipetting of liquid reagents in a laboratory is not advisable, different tools are needed for safe, easier, and precise dispensing of liquid reagents. This chapter is devoted to make you aware of the tools like vacupette and micropipette which can be used for the handling of liquid reagents in a safe and precise way.

Vacupette

Glass pipettes have been traditionally used by sucking liquid solvent/solution directly using mouth. For safety reasons, mouth pipetting is no longer permitted in a laboratory. Hence, to fill a pipette vacupette is used by placing it on the top of glass or plastic pipette. It acts as a device to create vacuum for filling reagent in the pipette as well as for releasing the reagent with mechanical regulation on the flow out of the liquid. Vacupette can be of different types.

1. Rubber Bulb Vacupette

Rubber bulb vacupette is often used to transfer (pump up and discharge) liquid using a pipette when a larger quantity of chemicals is to be transferred. Small/tiny rubber bulb is attached to glass/plastic pipette for drawing little amount of reagents. Small vacupette is used to precisely discharge smaller volume/ drops.

Handling of Vacupette

• Insert the top of a pipette in the bottom of the rubber bulb vacupette (Figure 6.1).

• Remove air from the pipette by squeezing valve “A” at the top of the vacupette while simultaneously squeezing the rubber bulb. The amount of air released depends on the size of the rubber bulb.

Introduction

To accomplish experimentations in laboratories, sometimes we need microscope to test/validate our ideas/findings. A microscope is needed to examine objects which are very small which cannot be visualised by naked eyes. However, microscopic study requires some expertise in operating the microscope, particularly the sophisticated microscopes, as well as to prepare (peeling, sections, cutting, fixation, staining, dehydration, mounting, etc.) the sample, so that it can be appropriately examined for the anticipated details.

According to the scientific requirements different types of microscopes are used in the laboratories for research purpose. In this chapter, our aim is to acquaint you with the basics of using and handling of microscopes, particularly a compound microscope that is commonly used by the graduate and post graduate students for basic research.

Microscopy is often designated as the scientific investigation that is employed to review and analyse minute structures and objects using a microscope. The term “microscope” has been derived from ancient Greek words “micros” meaning “small” and “skopeîn” meaning “to look at or see,”.

Microscope

A microscope is a laboratory instrument used to examine objects (biological specimens) which are too small to be seen by the naked eye. The microscope must accomplish three tasks:

• Magnification- to provide a magnified image of the specimen.

• Resolution- to separate the details in the image.

• Contrast- to render the details clearly visible to the eyes.

Although we will tnroduce to you the different types of microscopes used for different speciments, our main focus is to discuss about compound microscope, which is commonly used in the laboratories. Compound microscope was developed around the beginning of 17th century.

Introduction

In a research laboratory, we frequently need weigh chemicals and prepare different solutions of varying strength, as per the requirements. Accuracy in weighing and preparation of solution, with respect to weight/volume and strength, determines the success of experiment. In turn, the accuracy depends on weighing of solid chemical and/or measuring the volume of liquid chemical/making up the volume. In this Chapter, weighing of chemicals using appropriate weighing balance, preparation of solutions of different strength, their dilution, and precautions while preparing will be discuss in brief.

Weighing of Chemical

The primary method to determine the amount of chemicals to be used is weighing the chemical by using a balance.

In general, there are two types of balances to weigh chemicals in a laboratory:

1. Standard laboratory balance

Standard Laboratory Balance

Generally, a standard laboratory balance is used for weighing a chemical when an error of ±0.02 g is acceptable. It is also used for pre-weighing, where approximate mass needs to be determined. Precise mass of the chemical is then determined using an analytical balance. Following are the steps in using a weighing balance in a laboratory.

Introduction

For agricultural applications as well as for research work, often we need to prepare solutions of different agrochemicals of varying strength, as per the requirements. Agrochemicals are used to help crop plants grow, produce, and protect them from biotic/abiotic stresses. Many of the agrochemicals (insecticides, herbicides, fungicides, rodenticides, molluscicides, nematicides, etc.) are used to protect the crop from insect pests and diseases. However, some of them (fertilizers and soil conditioners) are used to augment crop yield. While in the previous chapter (Chapter 8) we discussed about preparation of laboratory solutions, in this chapter we will briefly discuss about the preparation of different agrochemicals.

Agrochemical/agrichemical is a generalized term used for the chemicals applied in agriculture. In general, it also includes the feed-supplements used in f isheries and animal husbandry.

Agrochemicals

Fertilizers

Fertilizers are used to augment growth of crop plants and to mitigate nutrient deficiency in the soil. Fertilisers can be organic (containing natural substances, prepared through natural processes) and inorganic fertilisers (synthetic fertilizers, manufactured artificially using chemical processes using natural deposits, by chemical alterations).

Soil Conditioners

Soil conditioners include manure, livestock and leaf compost, peat that help making the soil in good condition. They are laid on the top soil and then mixed properly. They not only increase nutrient contents in the soil but also improve aeration and water holding capacity.

Introduction

Measurement of pH of a solution and preparation of different types/strength of buffers are common practices in a laboratory, particularly in the Biochemistry and Molecular Biology laboratories. Therefore, in this chapter you will be acquainted with the procedures of measuring pH of a solution as well as preparation of some of the commonly used buffers in a laboratory.

Potential or Power of Hydrogen Ion (pH)

Measurement of pH of a solution is measuring the molar concentration of hydrogen ion [H+] in the solution, which is an indication of the acidity or basicity of the solution. The symbol pH stands for “power of hydrogen” and numerical value for pH is just the negative logarithm of the molar concentration of hydrogen ions [H+]. The ionic product of water (Kw ) is the basis for the pH scale. The total hydrogen ion concentration from all the sources and it can be experimentally measured.

The pH scale designates the concentration of H+ (and thus of OH−) in an aqueous solution, which ranges between 1.0 M H+ and 1.0 M OH−. The term pH is defined by the expression

Thus, the ionic product of water makes it possible to calculate the concentration of H+, with given concentration of OH−, and vice versa. The value of 7 for pH of a neutral solution is not an arbitrarily chosen figure; it is derived from the absolute value of the ionic product of water. A solution having pH greater than 7 is basic, and the concentration of OH− is greater than that of H+. A solution having a pH less than 7 is acidic.

Introduction

Use of acid/base is common in laboratories for experiments like titration and proper safety in their handling laboratory is necessary. In this chapter we will briefly discuss about the safety measures necessary to be adopted while using acids/bases in laboratory for any experiment.

Handling of Strong Acid/Base

There are four major points that need to be taken care of while working with strong acids or bases that might be quite harmful if any accident happens.

1. Protect Yourself

Wear a lab-coat and acid-resistant gloves: Always use lab-coat/apron and make it sure that the sleeves properly cover your arms. More importantly, use gloves made of acid-resistant materials and also wear shoes to protect your feet while wearing long-pants/jeans. Female with long hair, must tie her hair properly (with rubber band) to avoid any disturbance caused by appearance in between.

Wear safety goggles: It is important to wear safety goggles to protect your eyes using larger and fitting goggles that cover front as well as sides of your eyes. Always use properly fitting and adjustable goggles because smaller/ loose goggles may not protect the areas around your eyes against accidental acid spills and it may fall down from your face, if it is too loose.

Locate emergency wash area: There must be an emergency washing area nearby the work place or laboratory containing lab spill management kits. All the workers in the laboratory must be aware of using the kits and emergency plans.

Neutralize acid: Don’t panic even if an acid or chemical spill happens. Stay calm and act as quickly as possible. In case an acid spills, neutralize it by using sodium bicarbonate so that they can be cleaned and disposed safely. If acid comes in contact of skin, wash it off under running tap water but do not use/ apply sodium bicarbonate on skin.

Different types of culture media are used for culturing microbes and plant tissues. Preparing as well as sterilizing them properly following appropriate method with safety for the desired purpose are important parts of research work. In this chapter, we will discuss briefly about different types of media, their composition, preparation and sterilization in laboratory for microbial or plant tissue culture.

Culture Medium

Ingredients mixed to prepare solid, liquid or semi-solid substance to support the growth of microorganism or plant tissue in vitro (outside the body) is called culture medium. Frequently, it becomes important to grow microorganism or plant/animal tissue in vitro for any or all of the following purposes:

• To study characteristic features or properties of microorganisms.

• To identify the causal organism(s) of infection in a clinical sample, so that appropriate remedy can be determined.

• To culture/multiply microorganism or plant/animal tissues in vitro for research or commercial purposes.

• To prepare/produce biological molecules, vaccines, antigens, proteins, metabolites, etc. in vitro for research or industrial purposes.

Introduction

Tissue culture refers to the technique of growing cells, tissues, or organs under controlled, artificial conditions. This allows propagation of tissues from a small sample of cells/tissues under sterile conditions. Plant tissue culture is widely used for various purposes, including the production of disease-free plants, rapid multiplication of plants with desirable traits, genetic modification, conservation of rare or endangered species, etc. Tissue culture is a valuable technique because it enables consistent and large-scale production of plants, ensures genetic uniformity, and allows manipulation of plant biology at cellular level. In this chapter, we will discuss about the major types of tissue culture techniques and their advantages, disadvantages, as well as applications in brief.

Different Types of Tissue Culture

Callus culture involves induction and growing of unorganized, undifferentiated cell masses from an explant on a nutrient/culture medium. The part of plant used to induce callus is called explant which can be tissue, organ or part of a plant.

Organ culture is used to maintain or grow an entire organ or a part of organ in vitro. It preserves the structure and function of the organ for studies or transplantation.

Embryo culture involves isolating and growing embryos from seeds or ovules in a nutrient medium. It is used to bypass seed dormancy or rescue embryos from hybrid crosses that may not develop naturally.

Cell suspension culture involves growing dispersed cells or small cell clusters in a liquid medium under continuous shaking. This is used for large-scale production of cells, secondary metabolites, and studying cell biology..

Introduction

Seed is a living entity (mature ovule) consisting of a tiny plant in embryonic stage along with the stored food protected by an outer layer known as seed coat. Seed is an important biological input in agriculture, which is economically important for successful crop husbandry. Cotyledons serve as the source of nutrients (proteins, carbohydrates, and oils) that not only nurture the plant at the early developmental (seedling) stages of plant but also the vegetarian (heterotroph) organisms. Being one of the important inputs in agriculture, viability of seed needs to be assessed to know the germination percentage of seeds used to raise a crop. Since viability of seeds may deteriorate on storage depending on the storage conditions. Therefore, often it becomes important to test viability/germinability and vigour of seeds before being used for raising a crop or conducting an experiment. In this chapter, we briefly discuss about the different methods which can be used for testing the viability of seeds.

Seed Viability

Seed viability can be defined as the capability of the seed to germinate and produce a normal seedling within a specified period. In other words, seed viability indicates how many seeds in a lot are alive and could develop into plants under appropriate field conditions. It is very important that most of the stored seeds are capable of germinating when sown in a field. They must possess higher viability at the beginning of storage and maintain it over the storage. A seed with high initial viability survives longer in storage; however, the viability deteriorates slowly at first and then rapidly as the storage prolongs. It is important to know the duration of storage after which a considerable decline in viability occurs under given storage conditions so that to take necessary action to revive the seeds. Excessive deterioration in vianility of seeds may lead to loss of the seed/material/ germplasm.

Introduction

Test of pollen viability (extent of pollen viability) is important in pollination experiments to ensure fertility/seed setting. It is also important in monitoring state of pollens during storage in ecological/ taxonomic studies as well as during research on pollen biochemistry, genetics, stigma interactions, and incompatibility studies. Pollen viability refers to the ability of pollen to accomplish post-pollination events to achieve fertilization. Pollen viability test has gained importance particularly in plant breeding and hybrid seed production experiments/ practices. In this chapter, you will be acquainted with the basics of pollen viability test. However, for more professional knowledge on the topic readers are suggested to refer specialized books.

Pollen Viability Test

It is important to know the extent of viability of pollens used in pollination experiment, in finding fertility of a plant of interest, to monitor state of pollen on storage, in ecological/taxonomic studies, as well as in research on biochemistry, genetics, stigma interaction, incompatibility assay, etc. of pollen. Several pollen viability tests have been developed time-to-time. It is recommended to initially use different tests for a given pollen type to find out the most appropriate method that provides reliable result on pollen viability. It is important to note that every procedure requires standardization of protocol according to the sample used. Following different tests are used for assessing pollen viability.

1. In-vitro Pollen Germination Test

It is a simple, speedy, and quantitative test for assessing pollen viability. In this method, a collection/sample of pollen is germinated in vitro and observed under microscope. The percentage of germinating pollens (producing pollen tube) after appropriate treatment and a given period of time is determined. The percentage provides an index of viability of the pollen sample.

Flowering plants (Angiosperms) are identified by their morphological characteristics like flower, fruit and seeds that help their taxonomic characterization into different species, genera, families and orders. The floral characters which are used for taxonomic characterization of plants include the type of inflorescences, perianth structure, floral symmetry, arrangement of floral leaves, type of androecium, stamens, gynoecium, carpel, ovule, etc. The type of fruit and seed also play important roles in taxonomic classification. In this chapter we will briefly discuss about various morphological features of flowering plants (in botanical terms) that are useful in taxonomy.

Nomenclature of Plant

Nomenclature of plant is the formal and scientific naming of plants. It is related to but distinct from taxonomy for grouping and classifying plants and provide botanical name.

The scientific/botanical nomenclature started with Species Plantarum by Carolus Linnaeus in 1753. Botanical nomenclature is governed by the International Code of Nomenclature for algae, fungi, and plants (ICN), which replaces the International Code of Botanical Nomenclature (ICBN).

ICBN and ICN

International Code of Botanical Nomenclature, (ICBN) was adopted by the VIIth International Botanical Congress held in Stockholm, July1950. While the International Code of Nomenclature for algae, fungi, and plants (ICN) was adopted by the XVIIIth International Botanical Congress held in Melbourne, Australia in July 2011.

Introduction

We come across a variety of chemicals every day in a laboratory. While some of them are relatively safer, others are extremely hazardous. Thus, it is very important to know the potential hazards that can be caused by the chemicals present in the laboratory and the safety measures needed to be followed to avoid the hazards. Additionally, we should also be aware of the steps necessary to be taken on such hazardous situations. According to IUPAC, a hazardous chemical mean the chemical which has the potential to cause damage in various ways. In a laboratory, there are chemicals that might be irritant, carcinogen, respiratory fibrogen, systemic poison, asphyxiant, flammable, etc. which have the potential to cause hazard to the workers or environment. Therefore, it is important to use appropriate procedure to reduce the risk and protect the health/safety of laboratory personnel, the public and the environment.

Types of Hazards Associated with Chemicals

Chemical hazards: The hazards caused by chemicals can be broadly classified into three categories.

1. Health hazard: This is also known as toxic effects of chemical. The risk associated with a chemical in laboratory depends on the extent of exposure and the inherent toxicity of the chemical. Many chemical reactions form the product that might be much more toxic than the reacting chemicals. For example, inadvertent mixing of formaldehyde (a common tissue fixative) and hydrogen chloride results in the generation of bis(chloromethyl) ether, a potent human carcinogen. For most of the chemicals, the route of exposure (through skin, eyes, gastrointestinal tract, or respiratory tract) is more important (Figure 17.1) that must be considered during risk assessment.

Proper electrical wiring and earthing are crucial for laboratory safety and functionality. A laboratory houses several installations/equipment that demand stable/continuous power supply. Inappropriate or faulty wiring and earthing may lead to significant risks, including electrical shocks, damage to equipment, and even incidence of fire. Therefore, understanding the requirements of electrical wiring and earthing is essential for designing, construction, and maintenance of laboratory. This chapter aims to provide a basic knowledge of electrical wiring and earthing necessary for a laboratory. Importance of earthing in mitigating electrical hazards and ensuring integrity of electrical installations are also described.

Basics of Electrical Wiring

Basic understanding of electrical wiring is necessary for laboratory safety and efficient electrical functioning. The potential difference between two electrical points (negative and positive) is known as Voltage, which drives the flow of electric current. This is measured as volt (V). The rate of flow of electricity in a circuit is known as Current (I), which is measured as ampere (A). Current can be either direct (DC) or alternating (AC). Opposition/resistance experienced in the flow of current in circuit is known as Resistance (R) and it is measured in ohms (Ω). Higher the resistance means lesser the current flows at a given voltage. The rate at which electrical energy is transferred/utilized in an electric circuit is known as Power (P), which is measured in watt (W). The Power can be calculated as the product of voltage and current (P = V × I).

For a researcher, laboratory is another home where he/she spends much time every day. As we take care of cleanliness, safety, and design of our home, we must take care on these aspects of laboratory too. It is the responsibility of the laboratory personnel to ensure cleanliness/safety of the laboratory. Since every laboratory chemical and equipment have the potential to cause harms if not used with appropriate precautions. Hence, as a laboratory user, it is our responsibility to follow the basic safety/biosafety rules and regulations set up by the Principal Investigator of the laboratory, Institutional Biosafety Committee, as well as by other regulatory authorities, if any. We must be aware of the general/personal protective measures, cleanliness and laboratory waste disposal prior to joining a research laboratory. In this chapter we will briefly discuss about personal safety, chemical safety, cleanliness, waste disposal, etc.

A. Personal Safety

1. Personal Protective Equipment (PPE)

Personal protective equipment (PPE) is special arrangement to protect the worker from specific hazards because of using a hazardous substance. PPE does not reduce or eliminate the hazard, but protects the wearer from exposure to hazardous material. PPE should not be reused or brought outside the laboratory as these might increase the risk of spread hazardous material or contamination. Used PPE must be kept inside the laboratory in a designated waste storage area adjacent to the work area.

Introduction

Several hazardous chemicals, microbes, and techniques like recombinant DNA technology are used for research and development work in research Institutes and universities. Therefore, the researchers, technical, helping and supporting staffs as well as the laboratory outputs must be kept free from contaminations. Thus, biosafety is necessary to prevent unintentional exposure/ release of hazardous chemicals, microbes/pathogens, and toxins to the laboratory staff and environment. Following the biosafety guidelines and ethical considerations are essential to maintain safety standards for the researchers as well as the environment. This chapter deals with various biosafety measures and ethical issues to be strictly followed while working in a research laboratory. The required safety level needs to be determined as per the chemical/biological materials are used in the laboratory to keep the researcher/environment safe for sustainable research and developmental activities.

Biosafety

The term biosafety is used to describe the procedure and policies needed to ensure environmental and personal safety. Biosafety refers to the containment principles, technologies and practices that are implemented to prevent unintentional exposure to pathogens, toxins, or their accidental release into the environment. A fundamental objective of any biosafety program is the containment of potentially harmful chemical/biological agents as precautionary safety measures.

Why Biosafety?

With the increasing number of countries adopting molecular biology tools and techniques in life science research and development activities, especially in the areas of agriculture and medicine, the biosafety issues are gaining importance to ensure biological safety for the public and the environment. Conducting research in safe and sustainable manner is not only a personal requirement but essential collective efforts to ensure biological safety for a clean and safe environment. This requires rules, regulations, monitoring bodies, and awareness among the workers as well as the public. Therefore, biosafety per se is an integral part of laboratory research and requires awareness among the researchers so that biological safety can be well taken care from the grassroot level.

A laboratory is essential for research work in science and development. Most of the research institutes have dedicated laboratories for researchers and students to conduct experiments, perform research work, as well as to provide hands-on learning/training to students. There are certain rules and regulations laid down by the laboratory in-charge which need to be followed strictly for safety purposes. Some of the laboratories keep ‘safety manual’ for researchers to read before entering into the laboratory to conduct experiments. Whenever safety is compromised in a laboratory, there is increase in risk of unwanted incidents in the laboratory. Storing food/beverages in the same refrigerator where laboratory chemicals are kept may lead to disastrous effects on health. This chapter aims to provide instructions in brief on the things to be performed (Do’s) before and after entering onto a laboratory as well as the things never to be done (Don’ts) in a laboratory for the safety reasons.

Good Laboratory Practices (Do’s)

• P Wear apron/labcoat while working in the laboratory.
• P Use gloves/respiratory mask/fumehood while handling toxic/volatile/inflammable chemicals/reagents.
• P Use pippet or pippetor for safety in transfer of solutions.
• P Protect eyes, face and naked parts of the body from UV radiations. Use safety goggles or face mask/apron.
• P Keep your eyes away from Liquid Nitrogen and use cryogloves while handling it.
• P Arrange all the necessary materials/solutions before starting the experiment.
• P Periodically fumigate/ozonize culture and inoculation rooms.
• P Autoclave all contaminated materials before discarding.
• P Pre-treat gel or solution containing Ethidium Bromide (EtBr) before discarding into laboratory waste-disposal bin, and use gloves while handling them.
• P Burn all the transgenic plant materials in an incinerator after taking necessary observations.
• P Make entry in the designated log-book and clean the instrument after using it properly.
• P Switch-off the instrument after use or when not in use.
• P Wash hands with soap and remove the labcoat before leaving/going out of the laboratory7.
• P Ensure that switches for fans/lights, and water taps are off while leaving the laboratory.
• P Follow the biosafety guidelines suggested for handling and disposal of the hazardous chemical.
