Introduction

Proteins are complex biomolecules composed of amino acids, playing crucial roles in virtually all biological processes. They serve as enzymes, hormones, structural components, signaling molecules etc.

Structure/ Levels of Organization of Proteins

Generally, Proteins are polymers of L-alpha amino acids, linked by peptide bonds. The sequence and structure of these amino acids determine the protein’s function. Protein structure is hierarchically organized into four levels:

1. Primary Structure

• The linear sequence of amino acids in a polypeptide chain, determined by the genetic code.

2. Secondary Structure

• Local folding of the polypeptide chain into structures such as α-helices and β-pleated sheets, stabilized by hydrogen bonds.

Introduction

Lipids may be regarded as organic substances relatively insoluble in water, soluble in organic solvents (chloroform, ether, benzene, ethanol and other non-polar solvents, etc.), actually or potentially related to fatty acids and utilized by living cells. Lipids are a diverse group of hydrophobic or amphipathic biomolecules that play crucial roles in energy storage, cell membrane structure, and signaling. Unlike carbohydrates and proteins, lipids are not defined by a specific polymer structure but rather by their solubility in non-polar solvents. Broadly lipids are classified based on their function as ‘Storage lipids’ and ‘Structural lipids’. Some of them are co-factors, hormones, electron carriers, light-absorbing pigments and hydrophobic anchors of proteins.

Fatty Acids: Fatty acids are straight aliphatic chains with a methyl group at one end and a carboxyl group (-COOH) at the other end. They are the simplest form of lipids.

• Saturated Fatty Acids have no double bonds between carbon atoms, leading to straight chains (e.g., palmitic acid).

• Unsaturated Fatty Acids contain one or more double bonds, causing kinks in the chain (e.g., oleic acid). Fatty acids containing more than one double bond (polyunsaturated fatty acids/PUFA) are found in relatively minor amounts.

Carbohydrates are essential organic biomolecules/macromolecules composed of carbon c., hydrogen (H), and oxygen (O) atoms, typically in a 1:2:1 ratio. They are one of the four major classes of biomolecules, alongside proteins, lipids, and nucleic acids. Carbohydrates play critical roles in energy storage, structural integrity and cell signaling. Carbohydrates with an aldehyde as their functional group are called Aldoses. Those with keto as a functional group are called Ketoses.

Classification of Carbohydrates: They are broadly classified into three major groups:

1. Monosaccharides: The simplest form of carbohydrates, consisting of a single sugar unit. They cannot be hydrolyzed to yield simpler forms of sugar. They can be subdivided into trioses, tetroses, pentoses, hexoses, heptoses and octoses etc., depending upon the number of carbon atoms they possess. Important monosaccharides are:

• Glycerose / Glyceraldehyde (Aldotriose): Glyceraldehyde 6-phosphate is an intermediate in Glycolysis.

• Ribose (Aldopentose): found in Nucleic acids.

• Glucose (Aldohexose): The primary energy source for cells, known as blood sugar.

• Fructose (Ketohexose): Found in fruits (fruit sugar), a major component of sucrose.

• Galactose (Aldohexose): Known as brain sugar. It is a part of lactose sugar.

Introduction

The smallest, most basic, most primitive organisms are known as prokaryotic (Greek: pro= primitive or prior, and karyon = nucleus). Likely, they originated around 3.5 billion years ago. Bacteria, such as Mycoplasma and Cyanobacteria, are known to contain these cells. A prokaryotic cell is a deeply organized single-enveloped system. Its constituent nuclear parts are encased in a plasma membrane, with the cytoplasmic ground substance encircling the center. Prokaryotic cells’ cytoplasm lacks distinct cytoplasmic organelles, the nuclear envelope, and any other cytoplasmic membrane.

Despite being incredibly thin and delicate structures, in Eukaryotes plasma membranes are essential to many of a cell’s most vital processes. Membranes are lipid-protein assemblages where non-covalent linkages hold the constituents in place in a thin sheet. Both integral protein and phospholipid lateral mobility are significantly impacted by the physical condition of the lipid bilayer. The plasma membrane is a selectively permeable barrier that permits solute passage via assisted diffusion, active transport, and simple diffusion via membrane channels or the lipid bilayer. A system of membrane organelles, such as the endoplasmic reticulum, Golgi complex, and lysosomes, is found in the cytoplasm of eukaryotic cells. These organelles are physically and functionally connected to the plasma membrane and one another. The cytoplasm is separated into a luminal region inside the ER membranes and a cytosolic space outside the membranes by the endoplasmic reticulum (ER), a network of tubules, cisternae, and vesicles. Additionally, the ER is where the majority of a cell’s membrane lipids are generated and transported to different locations. Proteins’ asparagine residues are added with sugars (a process known as glycosylation), which starts in the rough ER and continues in the Golgi complex.

pH is a quantitative measure of the concentration of hydrogen ions in a solution. The pH scale ranges from 0 to 14, with 7 being neutral. Lower pH indicates acidity, while higher pH indicated alkalinity. Pure water at 25°C has a pH of 7.0 as the concentrations of H3 O+ and OH-. pH is defined as potential of hydrogen and is a measure of how acidic or basic a substance or solution is. The concentration of Hydrogen (H+) in a solution determines its pH. Higher concentrations of H+ ions correspond to lower pH values (more acidic), and lower concentrations of H+ ions correspond to higher pH values (More alkaline/basic). pH is an important measurement concept in many fields, including chemistry, biology, and environmental science. pH of blood and other fluids of the body, secretions etc?, determine the health status of an individual. Likewise, pH determines the quality of soil for plant growth. In medical sciences, pH plays a crucial role in maintaining a healthy internal environment for various bodily functions. Internal environment of an animal or human is tightly regulated more so the pH of different body fluids like blood, saliva and stomach acid.

pH balance is a very prime aspect to maintain homeostasis inside a biological system. Speaking of which the first thing that comes to mind is the enzyme function. Many enzymes have a specific pH range for optimal activity. Any deviation from this specific pH range not only significantly disrupt their function but also hinder biochemical processes involved. Most vital operations of a cell are nutrition transport and waste disposal i?e?, excretion. pH of the fluids surrounding cells impacts these activities; hence, maintaining a proper pH is vital for healthy cell function. Different organs of the body need to maintain different pH ranges for functioning accordingly. Stomach operates at acidic pH to digest the food material ingested whereas, blood need alkaline pH for optimal oxygen transport by RBC.

“Bio” means pertaining to life and “Chemistry” is the Study of chemicals and chemical reactions. So Biochemistry may be defined as the study of chemical processes within and relating to, living organisms. It is a field of science that explores the chemical processes and substances within living beings. Biochemistry bridges the gap between biology and chemistry, aiming to understand how biological molecules give rise to the complex processes of life. In other words “Biochemistry is the science dealing with various molecules that are present in living cells and organisms and their chemical reactions.”

Scope of Biochemistry

As a life itself, the scope of biochemistry is vast and ever changing. It has its influence on vide range of fields playing a major role in understanding and influencing living organisms. Here some of the fields were put forth that have applications of biochemistry.

Human and Medical Healthcare

Nutrition and metabolism: As they say, we are what we eat; nutrition is a prime aspect of the being and knowing how the body process what it eats is a necessary knowledge that comes from biochemical aspect of nutrition. The dietary component of the beings converting into energy, protein and other essential bodily macro and micro molecules is a great knowledge imparted by biochemistry aspect of nutrition. Metabolism which broadly includes anabolism and catabolism deals with various activities of the physiological systems with in the body. Knowledge of biochemistry is very essential these days as the metabolic diseases are very much prevailing. Biochemistry imparts basic to complex know-hows about various metabolic processes and their derailments.

The aim of the breeder is to improve on various economic traits through selection. However, due to the lesser number of parents used, over a period of time, there creeps in the effect of inbreeding and loss of genetic variability which in turn causes depression and loss of fitness. This is more prominent in finite endangered populations and evaluation for genetic variability also forms an integral part of the animal breeding analysis.

Genetic variability is an integral part of domestic animal populations as fitness is directly related to the variability.

Milk yield is one of the most important economic traits and modelling the lactation curve is important with respect to the unique shape with ascending, persistent and descending phases. The non-linear relationship of daily milk yield with days in milk has led to extensive research in modelling the lactation curve. The nature of the lactation curve, different models, identifying the best model of fit, factors affecting the curve parameters and orthogonal polynomials to model the curve are discussed in this chapter.

Traits such as growth, lactation test day yield, semen evaluation are longitudinal traits measured repeatedly on the animal and random regression models (RRM) have been found to be suitable for modeling these traits, The methodology used, data requirement, assumptions, validity, software available and application of RRM are explained in this chapter.

Growth, test-day milk yield, and semen traits are examples of longitudinal traits measured repeatedly on the animal and random regression models (RRM) have been found suitable for modeling such traits. RRM is based on a covariance function which could be defined as “a continuous function to give the variance and covariance of traits measured at different points on a trajectory” (van der Werf, 2001). Kirkpatrick et al. (1990) showed that variance components for longitudinal data can be modelled through covariance functions. Random regression models (RRM) and the resulting covariance function have been recognized as ideally suited for the analysis of longitudinal data (Hill, 1998; Meyer, 1998).

Linear models have been extensively used in animal breeding. This chapter explains the basics of linear models including description; fixed, random and mixed models; model diagnostics and covariates. The concept of linear models in animal breeding is explained with representation and actual examples of use in different types of livestock.

Introduction to Linear Models

A model can be defined as a physical, mathematical or otherwise logical representation of a system, entity, phenomenon, or process. Linear models are a way of describing response variables in terms of a linear combination of the predictor variable. They provide a framework for a large set of models whose common goal is to explain or predict a quantitative dependent variable by a set of independent variables that can be categorical or quantitative.

Estimation of variance is primary for any breeding program as all the genetic parameters are dependent on these estimates. The properties of an estimate, their assumptions, type of data, availability of data, and computational requirement make it important to understand the method of estimation of variance components so as to make a wise choice that would suit the breeder’s requirement. This chapter explains the methods of estimation of variance with merits and demerits of each and the reader could use these as a guide to choose the right method of estimation.

The genetic parameters are the basis for any animal breeding program and the important parameters such as heritability, repeatability and genetic correlation are ratios of (co)variances and their reliability depends on the accurate estimation of variance components. Estimates of variance components are used extensively in animal breeding.

The key genetic parameters available with the animal breeder to attain improvement are heritability, repeatability and genetic correlation. This chapter will mainly deal with the theoretical considerations of these parameters.

The goal of an animal breeder is to bring about permanent improvement in different economic traits, either individually (univariate) or simultaneously in multiple traits (multitrait). The phenotypic value available from the animal is a combination of genotypic value and environmental deviation. By permanent improvement in a trait, we mean the improvement in genotypic value, which could be carried over to future generations. The environmental deviations are spatially and temporally specific, and thus will not be the same over generations. The genotypic value, itself is made up of additive effect, dominance deviation and interaction deviation. The additive effect is a value of genes (the sum of the average effect of genes) and hence is more important for the breeder as only the genes are passed on to the next generation.

It is known that the phenotype of an individual is a combination of genotype and environment. Animal breeding analysis is mainly done to partition the genetic part from the environmental part so as to assess the actual genetic worth of the animal. In this process correction for the known environmental part is integral to any animal breeding analysis. In this chapter, a list of common factors affecting important economic traits are described. The analytical part for these effects will be explained in later chapters.

Correction or standardization of data is very important in any animal breeding analysis. Unless the candidates under selection are placed on a common platform, various factors affecting the trait will lead to bias in assessing the performance of the individual with respect to a particular trait. For eg. a lamb born during a favorable season, a lactating cow in its third parity, first lactation yield of a cow with longer age at first calving.

Correlations and regression are important components of animal breeding analysis. The genetic correlation is an important genetic parameter that is useful in the planning of breeding programs involving multiple traits. In this chapter, the basic concepts of correlation and regression including method of estimation, assumptions and test of significance are presented. An example using SPSS software is also given at the end for better understanding.

Basic knowledge of statistics is important to understand animal breeding analysis. In this chapter, the hypothesis testing, principles of experimental design, types of data and probability distribution are described with examples using appropriate software.

Meta-analyses has been used extensively to combine the results of existing studies and obtain powerful estimates of a parameter. In animal breeding, attempts have been made to obtain meta-analysis estimates for genetic parameters. This tool is more pertinent to animal breeding on account of heterogeneity in terms of location, sample size and other factors in the estimation of genetic parameters. This chapter explains the background, methodology, precautions and implications of meta-analysis with respect to genetic parameters.

A meta-analysis is a powerful tool of statistics that could combine several estimates from different studies for a parameter and the resulting weighted estimate could be used as a representative value for the particular parameter. However, not all the studies are similar with respect to location, sample size, and methodology used and thus various precautions are to be taken right from the selection of studies to incorporating of between-study heterogeneity in the models. In animal breeding, met-analysis is very much useful for obtaining meaningful estimates of genetic parameters and this chapter explains the methodology of meta-analysis in animal breeding.

Animal breeding data is unique in terms of volume, statistical analysis, type of information required etc. The large volume of data extended spatially and temporally warrants additional tools in the form of databases to handle data and enable accuracy, retrieval, efficient utilization and adaptability to software. This chapter explains the nature of animal breeding data and the importance of databases with examples and explanations.

Animals are raised for some utility including meat, milk, hide, wool etc., and efforts are on to continuously improve the quality and quantity of these products. Some of the important economic traits in this regard were already discussed in earlier chapters. Record maintenance is primary for decision-making with respect to the selection of animals. The details of record maintenance including the format of registers are already explained in chapter 2. Apart from the actual information required for economic traits, pedigree information forms an important part of animal breeding data.

Maintaining the required type of information in the form of records is primary for any type of animal breeding analysis and this chapter explains the importance of records and ways to maintain them in the form of registers or computer databases. The formats for different types of registers to be maintained are also given as a template in appendix. Any breeding plan is formulated to improve the economic traits depending on the utility of species reared and the chapter lists important economic traits of livestock and poultry with definitions and formulae, where ever required.

Basic record maintenance is a must for any animal breeding program. Every farm or breeding program is expected to maintain records in the form of registers or computerized databases. The goal of animal breeding is to select mates with the objective to improve economically important traits. In this chapter, the most common economic traits in different species of livestock are described. Even though the list is not exhaustive the basic economic traits that have been worked for improvement are explained. Breeders also explore new composite traits which are a combination of these base traits.

This chapter describes various multivariate techniques which are useful in animal breeding. The genetic correlation with multiple response variables is already explained in the chapters on Correlation and Genetic Parameters. The other techniques including MANOVA, Principal Component Analysis, Factor Analysis, Cluster Analysis and Discriminant Analysis are explained.

Multivariate analysis is referred to a set of data with multiple response variables in general and such data is encountered often in animal breeding analysis. The simplest form of multivariate analysis is the genetic correlation where two or more traits are analysed together as response variables with factors affecting them on the right-hand side (RHS) and details of these are already explained in earlier chapters.

Domestic livestock plays a major role in meeting the food requirement of human beings. Especially with dwindling resources, improvement in production efficiency is primary to meet the ever-increasing demand for animal protein. The role of an animal breeder is to bring about a sustained improvement in the quality and quantity of such products. Over the years, there has been an enormous improvement in the production ability of livestock and statistical analysis has played a predominant role in the application of principles of animal breeding to improve the productivity of animals. The combination of genetics and statistics developed into a separate branch of science called biometry.

The large volume, type and nature of animal breeding data and the complex nature of analytical requirement warrants additional tools for handling, storage, retrieval and solving complex equations. Accuracy, speed, computational requirement, applicability, etc., has resulted in the development of numerous software exclusive for animal breeding data analysis. The database management system is already explained in the chapter on ‘Nature of animal breeding data and database management’. This chapter gives a note on the commonly used animal breeding software.

Introduction

Animal behaviour is the study of how animals interact with one another, other living things, and their environment. Animals display a variety of behaviours in response to both external stimulus and their own pathophysiological state. Almost all domestic animals express a variety of behavioural responses throughout their lifespan, like feeding, sexual behaviours, etc. But every species responds to the same stimuli in a different way. This is why; the study of animal behaviour helps in efficient breeding, feeding, and reproductive management. It also serves as a reliable indicator of overall health and well-being.

Introduction

Growth is the irreversible gain in body weight, size, length, and circumference. It is accomplished by two fundamental processes: hypertrophy (increase in tissue, cell volume and progressive accumulation of intercellular substances) and hyperplasia (growth as a result of cellular multiplication by mitotic division). Animal growth is influenced by both genetic and environmental factors.

Animal body growth and weight gain play a critical role in reproductive performance. It also acts as one of the traits to evaluate the breeding value, optimize management & feeding practices, and prepare the correct dose of therapeutic pharmaceuticals to treat animal diseases. As a result, measurement of body growth is essential for the selection of superior germplasm, feeding & housing management, health care management, and optimizing other environmental conditions that have an impact on animal growth for an economically viable animal industry.

Climate change induces significant and long-lasting shifts in the statistical distribution of weather patterns over periods ranging from decades to million years. It is believed to be the cause of long-term temperature changes, changes in rainfall patterns and volume, prolonged summer seasons, increasing sea levels, and increased frequency and intensity of floods, droughts, and heat waves, etc.

Animals are intimately and inseparately associated with their physiological and chemical environments from conception until death. As a result, climate change has an impact on animals both directly and indirectly. Since, animals have had to alter their behavioural and physiological mechanisms in order to maintain homeostasis.

Climate change has emerged as a serious threat to the livestock industry. It reduces animal productivity by lowering growth, milk yield, meat production, egg production, wool production, reproductive performance, and health. About 20 to 30 percent of plant and animal species would likely be at risk of extinction with an increase of 1.5 to 2.5°C, which will result in severe consequences for food and nutritional security in developing countries. This is why; measurements of meteorological variables are needed for effective scientific management of livestock and poultry to cope with the negative impacts of climate change while enhancing growth and productivity.

Reproduction is the biological process that results in the formation of new progeny or individual organisms. Male and female gametes unite to form a zygote, which develops or produces offspring.

Endocrinology is the study of the endocrine system, which includes the glands and their secretions known as hormones. Hormones are essential for animal behaviour, growth and development, metabolism, lactation, reproduction, excretion, and reabsorption of inorganic ions. Therefore, if a certain hormone concentration in blood or another body fluid is detected, it provides the pathophysiology of the representative endocrine gland.

Endocrine glands, the cells or tissues they govern, and the secretory substances (hormones) they produce are investigated using a number of methodologies. They are as follows:

1. Surgical methods (e.g., hypophysectomy, gonadectomy, etc).
2. Chemical ablation or impairment (e.g., alloxan or streptozotocin agents that selectively destroy the insulin-secreting beta-cell, etc).
3. Hormone replacement therapy (e.g., estrogen during menopause to reduce the risk of bone fracture related to osteoporosis, etc).
4. Immunological neutralization of hormone activity (e.g., contraception both male and female, injection of antibodies against NGF resulting in failure of sympathetic nervous system development, etc).
5. Tissue extraction and purification (e.g., progestin is a synthetic hormone that mimics progesterone, etc.).
6. Histological-cytological studies (e.g., employment of a battery of histological stains to characterize each pituitary cell, etc).
7. Bioassays (e.g., oxytocin-induced contraction of rat uterine myometrial tissue strips, etc).

Introduction

The kidneys play a vital role in maintaining water and electrolyte balance (such as chloride, potassium, calcium, hydrogen, magnesium, and phosphate ions). It also regulates the acid-base balance of the blood, arterial blood pressure, and osmotic pressure of the nd water-soluble waste products, which are removed from the bloodstream by the kidneys and excreted as urine. Normally, the kidneys receive about 22% of the cardiac output. According to Arthur Cushny (1866-1926), there are three steps involved in urine formation in the nephrons: glomerular filtration, tubular reabsorption, and tubular secretion. When the kidneys are injured, their regulatory activities are disrupted, resulting in a change in the quality and quantity of urine. Urine is composed of the following components:

This is the reason why a urine analysis is a common test to evaluate the pathophysiology of the urinary system in general and the kidneys in particular, as well as to aid in the differential diagnosis of certain diseases.

Introduction

The main function of the respiratory system is to exchange O₂ and CO₂ between the atmosphere, blood, and cells, which are necessary for animal survival. The circulatory and respiratory systems work together to provide oxygen to cells while eliminating CO₂. O₂ consumption and CO₂ production are determined by cell metabolic activity.

Respiratory volumes or lung volumes refer to the volumes of gas in the lungs at any given point during the respiratory cycle. These volumes vary according to the animals’ physiological state, ambient factors, and health status. This is why; measurements of lung volumes are helpful in understanding the pathophysiology of the respiratory system.

The digestive system and its auxiliary organs main functions include digestion and the absorption of nutrients required for cell metabolic processes. Digestion is the mechanical and chemical or enzymatic breakdown of large nutritional macromolecules into simpler molecules that cells can absorb. It takes place in the aqueous milieu of the digestive system and is secreted by the salivary glands, stomach and mucosal glands, pancreas, and liver.

Digestive enzymes are secreted almost throughout the digestive tract. Salivary enzymes such as lingual lipase and α-amylase initiate the breakdown of fat and carbohydrates in the mouths of young animals (e.g., calves) and omnivores (e.g., rats, pigs, and humans).The optimum pH for oral digestion is 6.7 (6.2-7.6).

The gastric juice contains pepsinogen, HCI, and intrinsic factors. The pepsinogen enzyme is secreted by the stomach’s chief cell. Pepsinogen is converted to pepsin at acidic pH ranges of 2.0 to 3.5 and initiates the digestion of proteins in the stomach of monogastric animals. Also, the stomach’s parietal cells secrete HCI and intrinsic factors. Cyanocobalamin, is a synthetic form of vitamin B₁₂ requires an intrinsic factor in ruminants to be absorbed. The abomasum of young ruminants secretes proteolytic enzymes such as rennin or chymosin. Rennin acts on milk at a pH of 6.5.

The cardiovascular system is comprised of the heart and blood vessels (arteries, capillaries, and veins). The heart pumps blood to blood vessels which allow blood to flow throughout the body. Both heart and blood vessels are work together to keep the blood circulating, ensuring the interchange of O₂, CO₂, electrolytes, and nutrients between the blood and the tissue fluids & cells, and removing waste products from the body via the excretory system.

The purpose of the study of the cardiovascular system is aimed at understand the pathophysiology of the heart and vascular system. A physical examination, arterial blood pressure, and electrocardiography are routinely employed to examine the cardiovascular system. The physical examination of the heart includes palpation, percussion, and auscultation.

Hematology is the study of blood and its constituents. Blood is a specialized fluid connective tissue, which consists of plasma (55-70%) and the formed elements (30-45%) that flow all over the body in the vessels of the cardiovascular system. Plasma comprises water (90-92%), proteins (6-7%), enzymes, hormones, lipids, and organic & inorganic constituents. The three major classes of plasma proteins include albumin, globulin, and fibrinogen. The organic elements include non-protein nitrogen (NPN) compounds (like urea, uric acid, creatine, creatinine, amino acids, glutathione, and xanthine), glucose, neutral fats, phospholipids, cholesterol and others. The inorganic constituents include calcium, phosphorus, magnesium, potassium, sodium, chlorine, sulfur, iodine, iron, copper, cobalt, manganese, zinc, selenium, and molybdenum; they contribute about 1 percent of the plasma.

Blood performs various functions by its constituents. The constituents of the blood are highly dynamic, and they inter shift from their reference values very quickly in any adverse condition. This is why; studies of blood cells and complete blood count (CBC) are important for accessing general health, diagnosing various diseases, and providing prognostic information when coupled with a thorough physical examination. The blood examination is performed routinely to diagnose hematological diseases, including certain hereditary diseases of the blood, and to determine the body’s ability to respond to a hematological insult. The key parameters evaluated in the hemogram are ESR, PCV, Hb, TEC, MCV, MCHC, TLC, and DLC.

An accumulation of cerebro Spinal fluid (CSF) within the brains ventricles, leading to the enhancement of intra cranial pressure (ICP). This condition can happen at any age and may result from many underlying causes. Hydrocephalus can be congenital or acquired. Symptoms of Hydrocephalus include headache, nausea, vomiting, Gait disturbances, enlargement of the head in infants. Diagnosis is based on medical history, review, physical examination, Diagnostic tests and imaging studies namely MRI or CT scans. Differential Diagnosis include normal pressure Hydrocephalus, intea cranial hemorrhage and idiopathic intra cranial hypertension. Treatment is dependent upon ventricular shunt and endoscopic third ventriculostomy. Finally it is concluded that Hydrocephalus is a complex neurological condition along with diverse causes as well as presentations.

Rachel Carson written Silent Spring 50 years ago, she expressed her concerns about the use of systemic insecticides like neonicotinoids. We are currently seeing f irst-hand how her forecasts come true

“The world of systemic insecticides is a weird world, surpassing the imaginings of the brothers Grimm… It is a world where the enchanted forest of the fairy tales has become the poisonous forest in which an insect that chews a leaf or sucks the sap of a plant is doomed. It is a world where a flea bites a dog, and dies because the dog’s blood has been made poisonous, where an insect may die from vapors emanating from a plant it has never touched, where a bee may carry poisonous nectar back to its hive and presently produce poisonous honey (Rachel Caron, Silent Spring)”.

Chemicals are commonly employed to manage plant diseases, pests, and weeds, but they are only effective if administered promptly once an infestation has been discovered. These chemicals must be sprayed, dusted, or misted over the ground and plants. Since the chemicals are expensive, it is important to have tools for applying them evenly and effectively. Crop Protection Products (CPP) like herbicides, pesticides, and fungicides work best when they are used at the right time during the crop’s most productive times. This problem is getting worse because there aren’t enough people to do the work. In this situation, the only choice that makes sense is to automate the application process. Most of the time, poisons are put on with dusters or sprayers. Dusting is the easier way to apply chemicals. It works best with movable equipment and usually only needs simple tools, but it is less effective than spraying because the dust doesn’t stick around as long. High volume spraying is usually efficient and reliable, but it is more expensive than low volume spraying, which fixes some of the problems with the first two methods while keeping the things that work well about them.

Crops are grown with the use of plant protection chemicals to boost production, enhance quality, and lengthen shelf life. Wherein the enormous use of pesticides which are having the longer half life period led to leave the residues in the different bodies of the ecosystem. Pesticide residues refer to the residual presence of pesticide active ingredients, their metabolites, or breakdown products in various environmental components subsequent to their application, accidental release, or disposal.

Analysis of residues gives insight on the type and extent of chemical contamination in an area, as well as how long it has been there. Before governments will approve the use of pesticides, they require comprehensive testing of their efficacy, environmental impact, and toxicology. A possible risk to human health arises from the presence of chemical residues and/or breakdown products in agricultural goods. This leads to more effective pest control and reduces the need for higher pesticide concentrations.

2. Induced Resistance: Silicon has been shown to induce plant resistance against pests. When plants take up silicon from the soil and deposit it in their cell walls, it can create physical barriers that make it more difficult for pests to penetrate plant tissues. Additionally, silicon can trigger the production of defensive compounds in plants, making them less susceptible to pest damage.

3. pH Buffering: Silicon compounds can act as pH buffers in pesticide formulations, helping to maintain the stability and efficacy of the active ingredients over a broader pH range. This can be particularly useful when mixing pesticides with other chemicals or using them in different environmental conditions.

The role of toxicology in the evaluation of new agrochemicals

The widespread use of agrochemicals for crop protection may expose humans, animals, and the environment to toxic chemicals. Humans may be exposed during manufacture or application, and to a lesser extent as consumers of food products containing trace amounts of residues.

Strategies of toxicity testing

No-Observable-Adverse- Effect-Level (NOAEL)

A level in which the total diet that causes no adverse effect in treated animals when compared to untreated animals maintained under identical conditions. This NOAEL is expressed on a mg/kg of body weight/day basis. Chronic and sub chronic studies are need to be carried out in different species to determine NOAEL values.

It depends on how much of the substance an organism is exposed to, but all chemicals are toxic to some degree. Toxicology studies try to figure out the exact types and amounts of harmful effects, as well as the lowest doses that won’t have any negative effects (called the “No Observed Adverse Effect Level” or NOAEL). Most data come from studies with lab animals and cells that have been grown in a lab. Using these numbers and extrapolation or uncertainty factors, it is possible to figure out an Acceptable Daily Intake (ADI). This is the maximum amount of a substance that a person can take in every day without putting themselves at a very high risk of getting sick. Farmers still rely heavily on insecticides because of their efficiency, portability, and speedy returns on investment. Although synthetic organic insecticides are highly efficient, their widespread usage has led to toxicity to natural enemies, toxic residues in plants and the environment, insect resistance, and a resurgence in insect populations. Ripper (1956) first time recognized the problem of resurgence in plant protection.

Metcalf (1986) distinguished between two types of resurgence: primary insect resurgence, in which populations of target insects that had been suppressed by insecticide application quickly recover to excessive levels (Fig. 1.A), and secondary insect outbreak, in which non-target species develop into serious insect s after insecticide application (Fig. 1.B).

Primary insect resurgence

Primary insect resurgence occurs when the target insect population increases to at least as high (Hajek, 2004) or higher (Hardin et al., 1995) than in an untreated control or higher than before the treatment (Pedigo and Rice, 2006).

Secondary insect resurgence/ secondary insect outbreak (Type II resurgence)

The phenomenon known as replacement of a primary insect with a secondary insect, or secondary insect outbreak, occurs when the population of a non-target insect, which is harmful to the crop, experiences a surge following the application of a insecticide aimed at controlling the population of the primary insect. A second insect outbreak is also called resurgence. A secondary insect outbreak is the rise of a non-target species after an insecticide has been used.

Insecticide resistance is a growing concern for those who need insecticides for medical, veterinary, and agricultural control of insect pests. In response to the introduction of DDT resistance in 1947, the incidence of resistance has increased annually by an alarming amount. Resistance is “the inherited ability of a strain of some organism to survive doses of a toxicant that would kill the majority of individuals in a normal population of the same species” (WHO, 1957).

As a result of continued insecticide application, the proportion of resistant insects increases compared to that of susceptible and the population becomes increasingly difficult to control

An adaptation to a pesticide leads to reduced susceptibility to it among the pest population targeted by that pesticide. In nature, pests develop resistance to chemicals through natural selection. In the most resistant organisms, their genetic traits are passed on to their descendants. Since 1945, at least 500 species of pests have developed resistance to pesticides (Anonymous, 2007), but other sources estimate as many as 1000 species (Miller, 2004).

Synergists are chemicals that significantly increase the toxicity of an insecticide while being nearly non-toxic on their own. Thus, non-toxic chemicals that contribute to the increased toxicity of insecticides are referred to as synergists. Many chemical substances have the potential to synergize with poisons. Some enzyme inhibitors work well synergistically by inhibiting enzymes that metabolize pesticides, particularly in insecticide-resistant strains. When mixed with other insecticides, some insecticides increase the toxicity of the latter at non-toxic dosages. When a mixture is more harmful than expected based on the sum of its separate efficacies when administered alone, the interaction between substances is known as synergy. Antagonistic interactions among components reduce a mixture’s efficacy.

Synergism can also develop between two or more poisons. Several statistical methods have been proposed to quantify the synergy of both non-toxic and hazardous substances. Toxin interactions are assessed using bioassays, which involve mixing toxicants (two or more) in a specific ratio. The combination is then used to prepare serial dilutions. Alternatively, the poisons can be supplied sequentially in the same amounts as in the mixture. Bioassays are also used to determine the dose-mortality response of each toxin on the same insect strain. The data are subjected to probit analysis, and the toxin interactions can be evaluated using the statistical methods listed below.
