The fruit is seen covered with a mealy coating due to colonies of small reddish-white mealy bugs. They also infest leaves, shoots and buds. Due to sucking of sap by nymphs and adult female mealy bugs, fruits remain small in size and become shrivelled. Large number of mealy bugs is seen in the tip of the petiole abd also on the grooves of fruits epicarp. The fruit also becomes blackish due to the development of sooty mould.

34.2. Theory

Cut out examination of canned products is conducted to evaluate the product whether confirming to FSSAI standards or not. Canned fruit and vegetables are evaluated for presence of vacuum or pressure in the can, drained weight, total soluble solids, presence or absence of preservative, internal condition of can product contents and presence or absence of fermentation for ensuring microbiological safety.

ABSTRACT

Cultivation of mat grass (Cyperus pangorei Rottb.) is valuable and expensive products can plays an important role in rural areas such as Sabong and Pingla in Paschim Medinipur of West Bengal in India. Mat grass plays an act to fill the empty employment field to the resource of poor farming community to support and secured their livelihoods and maintain the economical balance of their area. This grass usually cultivated by the poor and marginal farmers. The mat sticks are used to make various household utility items, such as floor and bed mats and other attractive goods like bags, purse, TV covers. Now a days the products prepared from this grass is exported to different countries. Mat sticks are attractive durable and is eco-friendly substances which easily gets decomposed by organisms, does not create any environmental hazards and gets more profit as compared to growing rice per annum. Even an old man or woman in rural areas can earn a net income of about Rs.120 to Rs150 per day. Major focus of the study is to establish the scientific principles behind processing, separation of clum strand, texture and strength properties of the mat fibers and increase mat production which leads to improve the economic condition of rural people.

Abstract

Dactylorhiza hatagirea (D.Don) Soo, a plant species native to Indian Himalayas (Badola and Aitken, 2003; Ved et al., 2003), is one of 110 orchids known for their medicinal properties (Jain 1991; Lal et al., 2004). Known earlier as Orchis latifolia auct. non Linn., D. hatagirea has traditionally been used in indigenous system of medicine in India as ‘Hatajari’ or ‘Salam Panja’. The plant has therapeutic, ornamental and ecological significance (Gupta et al., 1998; Vij et al., 1992; Kala, 2000a). Due to over extraction from nature for medicinal use, the species has become threatened and categorized as critically endangered species (Ved and Tandon, 1998; Kala, 2000b; Ved et al., 2003; Bhattarai et al., 2014). This contribution provides insight into the botanical background, distribution, medicinal importance, threat status, cultivation and commercial aspects, and strategies for its conservation and sustainable utilization.

Introduction
Among flowers, the rose enjoys the greatest adorable position over the world. In real sense, rose almost hypnotised the man most by its colour and fragrance. Apart from its aesthetic value, the roses are of high economic benefits, and religious significant. Besides, it finds it place in art and culture from times immemorial. The rose thus merits pride of place as the ‘Queen of flowers’.
Origin
The damask rose is originated in ancient Persia (now Iran), and later on spreading over Europe and Northern Africa. Further, widespread farming of R. damascena for rose oil manufacturing began in Turkey and Bulgaria. Many countries of the world grow scented roses for extraction of the volatile essential oil. The genus Rosa grows wild in many parts of the temperate and sub-temperate regions. The major growing countries are Bulgaria, France, Italy, Turkey, Russia, China and India. Bulgaria is the principle European country for supply of rose oil to the world market. Among different species, Rosa centifolia Linn. is largely grown in France and Italy, whereas Rosa
damascena Mill. in Turkey, Bulgaria, Russia and Persia. Rosa damascena Mill. was first introduced successfully in middle east countries during 1680, resulting in the establishment of entire area as ‘Valley of flowers’. It belongs to family rosaceae.

The term dandruff includes several different conditions of the scalp. There is scaling in which the entire scalp may be covered with loose dry scales. When the hair is combed or brushed, or when the scalp is scratched, these scales fall like snow on the shoulders. Often there is itching, as well, and the scalp may be red from scratching.

What is at first thought to be dandruff may later prove to be some other conditions, such as. seborrhoeic dermatitis, or worse still psoriasis and even neurodermatitis.

Neurodermatitis : It is a chronic type of itching and inflammation of the skin. The itching area may be in some localized spot or it may extend over large areas of the body. The condition may even develop in infants. It may disappear after the age of two and return in the early teens of later. The condition is more often seen in those who suffer from fever, hives and asthma. True cause is not known for most of the people are allergic to many things. Many of them are tense and restless and are troubled with a dry skin. They are hypersensitive and are often given to itching, especially in the bend of elbow or behind the knee. The constant scratching leaves deep scratch marks and causes thickening and furrowing of the skin.

Morphological characteristics

The plant is a monoecious undershrub that grows up to 3.5 metres tall and has serrated leaves and stiff branches that emerge from the root. Lower branches feature big, typically broad, elliptical, three- to five-lobed leaves with rounded bases, while above branches have small, lanceolate leaves. Petioles range in length from 5 to 15 cm.

Floral characteristics

The flowers are unisexual. The calyx of male flowers is globose, 2.5 mm long, four to five partite, glabrous or slightly pubescent, membranous, finely mottled with a disc of six glands, and glabrous or slightly pubescent. There are approximately 20 stamens in each flower. Sepals are ovate-lanceolate and glabrous, and the disc is roughly 2.5 mm in diameter in female flowers. Fruit is a three-lobed capsule that is around 8–13 mm long and hairy. Seeds contain an oily endosperm and are speckled and smooth. Flowers bloom in January–February, followed by fruit that mature a month later.

Distribution

The species grows in tropical and subtropical places with rainfall of around 1000 mm, such as the Himalayan foothills, Kashmir to the Khasi hills, and especially in the Vindhyas southward. In North and East Bengal, Chhota Nagpur, and peninsular India, it is fairly frequent.

Date-palm (Phoenix dactylifera L.). belongs to family Palmae and is the most common tree of desert and dry areas. Date palm is one of the oldest trees known to man kind. Date-palm is believed to have originated in the countries around Persian Gulf. It is extensively grown in Iraq, Iran, UAE, Saudi Arabia, Algeria, Egypt, Libya, Yemen, Mauritiana, Morrocco, Tunisia, Baharin, Sudan, USA, Spain, Australia, Pakistan, South Africa, Somalia, Oman and some parts of the South America. Date is a staple food of the people of the Western Iraq and North Africa. In India, it has been believed to be introduced by soldiers of Alexander the Great in 4^th century B.C. In India it is commercially cultivated in Rajasthan, Punjab, Hariyana and Gujarat.

Cultivars: Deglet Noor, Halawy, Khadhrawi, Hayani, Majhul, Zahidi, Khalaeh.

Uses

Date palms produce many products that are useful to human beings. The primary product is the date fruit, which can be eaten fresh, dried, or in various processed forms. In North Africa and the Middle East, some dates are harvested and consumed during the Khalal stage, when the fruit are still very astringent with a high tannin content (Dowson and Aten, 1962 ; Glasner et al., 2002; Kader,1992). Dates can be used in cereal, pudding, bread, pressed cakes, cookies, candy bars, ice cream, and date shakes (a California specialty). Date fruit also can be made into juice, vinegar, wine, beer, sugar, syrup, honey, chutney, pickle, paste, dip, and food flavoring (Barreveld, 1993; Glasner et al., 2002).

It is said that there are as many uses of dates as there are days in a year. The trunk and wood of date palms can be used as timber, wood, or fuel. Fiber from the trunk and leaves can be made into bags, baskets, camel saddles, cords, crates, fans, food covers, furniture, mats, paper, ropes, trays, and twine. Dried bundles of leaves (‘‘Barusti’’) can be made into shades, roofs, separating walls, and enclosures. Ribs of the leaves can be used to build boats (small fishing boats called ‘‘Shasha’’) or fishing traps. The base of the leaves and fruit stalks can be used for fuel. The pith of the palms can be made into date palm flour. Terminal buds (palm heart) can be eaten as a salad or as a cooked vegetable. Date seeds can be used as feed for livestock or strung as beads for decoration. Oil from date seeds can be manufactured into soap (Chao and Krueger, 2007). Dates are considered a balanced and healthy diet and have several medicinal uses. Leaves are source of fibre used for making ropes baskets and cordage. Walking sticks are made from leafstalk.

Introduction
Davana (Artemisia pallens Wall.) is one of the important aromatic herb found abundantly in humid habitats in the plains all over India. It is popularly known as sage brush, worm wood and mug word. The davana (Artemisia pallens) is a small to medium height; annual and herbaceous aromatic plant. This herb is erect, and grow up to 60 cm height. Its leaves are highly divided and bears small size yellow coloured flower. Its leaves and stem found protected with greyish-white tomentum. The leaves are lobed, alternate, and petiolate.
Origin & Distribution
It is native to southern India, where it is traditionally grown in Karnataka, Maharashtra, Kerala, T.N., and A.P, growing widely in tropical to subtropical
plains as Rabi season crop. It also found growing in wild form in the Himalayan region in the Simla, Kashmir valley, and Nainital hills. It is widely used by Ayurvedic healers to balance the three energies. It is an erect growing
plant up to 60 cm height. It does not grow wild, but grown as a garden plant in South India. It is a crop of irrigated conditions; and do well when planted in November-December, yielding significantly higher oil yield in comparison to crop planting in July-August. The crop should be harvested 2-3 inches above ground level during bright sunny days at 50 percent flowering stage to have high oil content and oil yield.

Semilooper: Thysanoplusia(Plusia) orichalcea : Noctuidae

Bluish green or grey coloured eggs are laid singly on lower surface of leaves.The larvae are semi loopers and feed voraciously on leaves and cause defoliation of the plants.

48.2. Theory

Drying generally refers to the method of removal of moisture from the food under natural condition such as sunlight and wind for example open sun drying, shade drying etc. Dehydration refers to a process of removal of moisture by application of artificial heat under controlled conditions of temperatures humidity and air flow. It also represents drying of agricultural commodity to dry state. For drying, single thin layer of fruit and vegetables, either whole or sliced after primary pre-treatments is spread on the trays which are then placed inside the dehydrator or in the open sun for drying. In the dehydrator initial temperature is generally kept at 43oC which is then gradually increased to 66-71oC for fruits.

To perform practical on drying/dehydration of different vegetables.

49.2. Theory

Drying refers to the method of removal of moisture content from the food to a level at which the activities of food spoilage and food poisoning micro-organism are inhibited. Therefore, reduction in water activity of the food is the main principal of preservation by drying. In the dehydrator initial temperature is generally kept at 43oC which is then gradually increased to 60-66oC for vegetables. The product shall be prepared from wholesome vegetables free from blight, insect infestation and fungal discoloration. Only edible portion of the vegetables shall be used and it shall be free from stalks, peels, stems, and extraneous matter. The dried/dehydrated vegetable may contain permitted preservative. The finished product shall be of good edible quality and shall reasonably reconstitute to its original shape and quality on boiling for 15 minutes to an hour. The finished product shall be free from visible mould, insect or larvae. Kind of dry vegetable shall be declared on the label. Acid insoluble ash shall not be more than 0.5%.

This chapter provides an understanding on demand-driven vegetable breeding, particularly opportunities, and challenges.

Sustainable development and efficient management of water is an increasingly complex challenge in India. Increasing population, growing urbanization and rapid industrialization combined with the need for raising agricultural production generates competing claims for water. Ground water has an important role in meeting the water requirements of agriculture, industrial and domestic sectors in India. About 85 percent of India’s rural domestic water requirements, 50 percent of its urban water requirements and more than 50 percent of its irrigation requirements are being met from ground water resources. Ground water is annually replenishable resource but its availability is non-uniform in space and time. Technically, the dynamic ground water refers to the quantity of ground water available in the zone of water level fluctuation, which is replenished annually. Hence, the sustainable development of ground water resources warrants precise quantitative assessment based on reasonably valid scientific principles. National Water Policy, 2012 has laid emphasis on periodic assessment of ground water resources on scientific basis. The trends in water availability due to various factors including climate change must also be assessed and accounted for during water resources planning. To meet the increasing demands of water, it advocates direct use of rainfall, desalination and avoidance of inadvertent evapo-transpiration for augmenting utilizable water resources. The National Water Policy, 2012 also states that safe water for drinking and sanitation should be considered as pre-emptive needs followed by high priority allocation for other domestic needs (including needs of animals), achieving food security, supporting sustenance agriculture and minimum eco-system needs. Available water, after meeting the above needs should be allocated in a manner to promote its conservation and efficient use.

1. Introduction

The Indian economy is heavily dependent on agriculture (employing 53 % population). Around 84 % of the holdings are less than 2 ha while approximately 67% of the farms have less than 1 ha of operational farmland (Government of India, 2013; Pandey and Suganthi, 2015). A large fraction of the farming population is mostly reliant on monsoon rains for irrigation. The condition of the farmers is found to be deteriorating year after year with the revelation that 27% of farmers did not consider farming as a profitable enterprise and 40% were ready to migrate to other avenues for employment (NSSO, 2005; Pandey and Suganthi, 2015). 

Individuals suffering from the more pronounced types of mental disorder are sometimes said to be insane, but the term insanity is a legal one, not a medical term. An insane person is one who cannot distinguish right from wrong or assume responsibilities for his own actions. In the case of a psychotic patient the whole personality is involved. And he is completely out of touch with real world around. Not so the neurotic patient. Only a small part of his personality is involved. Psychotic patients often require hospitalization; neurotic patients rarely do, unless they have some additional physical problem.

Another distinction between these two major groups is that his friends usually bring the psychotic patient to the doctor, while the neurotic comes of his own accord. He knows he is sick, and he usually wants help. Most often he is suffering from a nervous depression.

Introduction

Flowering plants (Angiosperms) are identified by their morphological characteristics like flower, fruit and seeds that help their taxonomic characterization into different species, genera, families and orders. The floral characters which are used for taxonomic characterization of plants include the type of inflorescences, perianth structure, floral symmetry, arrangement of floral leaves, type of androecium, stamens, gynoecium, carpel, ovule, etc. The type of fruit and seed also play important roles in taxonomic classification. In this chapter we will briefly discuss about various morphological features of flowering plants (in botanical terms) that are useful in taxonomy.

Nomenclature of Plant

Nomenclature of plant is the formal and scientific naming of plants. It is related to but distinct from taxonomy for grouping and classifying plants and provide botanical name.

The scientific/botanical nomenclature started with Species Plantarum by Carolus Linnaeus in 1753. Botanical nomenclature is governed by the International Code of Nomenclature for algae, fungi, and plants (ICN), which replaces the International Code of Botanical Nomenclature (ICBN).

ICBN and ICN

International Code of Botanical Nomenclature, (ICBN) was adopted by the VIIth International Botanical Congress held in Stockholm, July1950. While the International Code of Nomenclature for algae, fungi, and plants (ICN) was adopted by the XVIIIth International Botanical Congress held in Melbourne, Australia in July 2011.

Protocol

Collect 5 ml of blood from the animal in 1 ml (20 IU/ml of blood) of diluted heparin and add 2 ml of PBS. Layer over the diluted blood on 5 ml of histopaque-1077 in 15 ml centrifuge tube and centrifuge at 1500 rpm for 40 min. Collect the buffy coat at the interface of plasma and histopaque in separate centrifuge tubes. Wash the cells thrice with PBS and suspended in 2 ml of RPMI-1640 media supplemented with 10% fetal calf serum. Determine the percentage cell viability by 0.1% trypan blue exclusion test, using hemocytometer. Finally dilute the cell suspension to 106 cells/ml in RPMI 1640 medium.

Injudicious and indiscriminate use of several agrochemicals viz. pesticides, herbicides, insecticides, dyes and medicaments in the field of agriculture and veterinary practices threatens the human and animal population for the toxicity of these environmental deterrents. These agrochemicals have been associated with several episodes of accidental poisoning and mortality among human and animal population. The episode of Mass ethion poisoning in Gujarat is one of the commonest example of inclusion of pesticides or agrochemicals in the environmental pollutants. There are modern tools to evaluate and analyze these chemicals in the environmental pollutants of human and animal use. The sensitivity of these modern highly sophisticated tools has now immensely changed the analytical environmental pollutants industry. Gas chromatography and Mass Spectrophotometry i.e. (GC-MS) technique is employed to detect the volatile residues from environmental pollutants. Whereas, High performance liquid chromatography i.e. HPLC is employed to analyse and detect non volatile drug residues. Gas Chromatography/Mass Spectrometry (GC/MS) instrument separates chemical mixtures (the GC component) and identifies the components at a molecular level (the MS component). It is one of the most accurate tools for analyzing environmental samples.

ABSTRACT

Genetic relationships were estimated among twenty five ashgourd (Benincasa hispida (Thunb.) Cogn.) landraces of different types using PCR-based random amplified polymorphic DNA (RAPD) markers. The molecular survey of the ashgourd germplasm by RAPD using three random primers resulted in twenty scorable bands (average of 6.66 bands per primer) of which two were monomorphic and rest, 18 were polymorphic (90.0%). Pair-wise genetic similarities among the landraces determined using Jaccard’s coefficient ranged from 0.14 to 1.00. The dendrogram based on clustering matrix separated the landraces with morphologically distinct smooth textured fruits from waxy textured fruits and the former group showed less divergence than the latter. With in the group of waxy textured fruits, limited variation was detected among landraces with small and medium sized fruits. Further, morphologically similar landraces with large fruits form distinct clusters with in the major clusters. The study demonstrated the usefulness of the RAPD marker analysis in effectively assessing the genetic relationship present among the ashgourd landraces examined.

ABSTRACT

Genetic variability was assessed among twenty five distinct drumstick (Moringa oleifera Lam.) landraces using PCR based random amplified polymorphic DNA (RAPD) markers. Genomic DNA extracted from tender leaves was used for RAPD analysis. Thirty five primers were screened and based on the number of bands three primers were selected. The primers produced 34 bands, of which 58 % (20 bands) were polymorphic. The data were analysed using NTSYS pc 2.1 software. The similarity matrix constructed using Jaccard’s coefficient method was subjected to cluster analysis using UPGMA (Unweighted Pair Group Method with Arithmetic Mean) and dendrogram was generated. The pair wise coefficient values in the genetic similarity matrix among the landraces varied between 0.28 and 1.00. The dendrogram based on the matrix separated the landraces with distinct morphological characters and it was noted that the cluster formation was not fully in agreement with the geographical location. The study showed the use of RAPD markers in detecting genetic variability present among the drumstick landraces studied.

Fibres can be classified into four categories like crude fibre (cellulose); acid detergent fibre (cellulose + lignin); neutral detergent fibre (cellulose + lignin + hemicelluloses) and total dietary fibre.

55.3. Procedure for estimation of dietary fibre

55.3.1. Acid detergent fibre (ADF)

Reagents: Cetyl tri-methyl ammonium bromide (CTAB) reagent and Acetone

Apparatus: Digestion apparatus vacuum pump, oven, desiccator and sintered glass crucible.

Purpose: The organic acids present in fruits and vegetables influence the flavour, colour, keeping quality and maturity. The maturity standards are determined by the acid and sugar ratio. The total acidity (citric acid) is determined by titrating the juice against standard alkali solution.

Equipments and chemicals: 10 ml., pipette, 25ml. Burette, small glass funnel, dropper, juice extractor, distilled water, 100ml. volumetric flask, burette stand, beaker, N/10 sodium hydroxide, filter paper, phenolphthalein indicator (one gram of phenolphthalein in 100ml. of 95% ethyl alcohol)

High-performance liquid chromatography or high-pressure liquid chromato graphy (HPLC) is a chromatographic method that is used to separate a mixture of compounds in analytical chemistry and biochemistry so as to identify, quantify or purify the individual components of the mixture. It involves the injection of a small volume of liquid sample into a tube packed with tiny particles (1.8- to 5-micron (μm) particle size called the stationary phase) where individual components of the sample are moved down the packed tube (column) with a liquid (mobile phase) forced through the column by high pressure delivered by a pump. These components are separated from one another by the column packing that involves various chemical and/or physical interactions between their molecules and the packing particles. These separated components are detected at the exit of this tube (column) by a flow-through device (detector) that measures their amount. An output from this detector is called a “liquid chromatogram”.

Aflatoxins are toxic and carcinogenic compounds produced as metabolites by many Aspergillus species. This fungus invades habitats such as soil, grain, nuts, or decaying vegetation when- ever suitable conditions such as high moisture and temperature are met. B1 is the most toxic of at least 14 different types of naturally occurring aflatoxins. Therefore, many countries and regulatory agencies impose strict limits on B1 as well as other common aflatoxins such as B2, G1, and G2 despite exhibiting lower toxicity. The U.S. FDA set the action level to20 μg/kg (ppb) for human food and to 300 μg/kg (ppb) for animal feed. Aflatoxin B1 is extremely widespread and is present in food and feed products such as cottonseed, corn, and peanuts. It is highly toxic and has been classified as a group 1 carcinogen by the WHO. The aflatoxins B2 , G1 , and G2 are usually found accompanying B1 , in lower concentrations in the contaminated samples. The order of toxicity is AFB1 > AFG1 > AFB2> AFG2.

Principle: Sample extract in oxalic acid is titrated against standard sodium 2, 6 dichlorophenolindophenol dye to a faint pink colour which persists for 5-10 seconds.

(Indophenols Method)

Purpose: The ash content of plants represents the deposition of minerals elements in the plant or in its parts. The value of ash content in the plant tissue indicates the health of the plant. Plant ash is also used both in qualitative and quantitative estimation of mineral elements.

Material and Equipments: Crucible, oven, chemical balance and weight box.

To determine crude fibre in given samples.

54.2. Theory

Fibres can be classified into four categories like crude fibre (cellulose); acid detergent fibre (cellulose + lignin); neutral detergent fibre (cellulose + lignin + hemicelluloses) and total dietary fibre (Cellulose + lignin + hemicelluloses + pectic substances). Crude fibre is the organic residue of a sample. A fat free sample is treated with boiling sulphuric acid and subsequently with sodium hydroxide. The residue after subtraction of ash is regarded as crude fibre.

Fructose, a keto-hexose (called as fruit sugar), is usually accompanied by sucrose in fruits and estimated by the methods of Ashwell (1957).

Materials

Resourcinol reagent: Dissolve 1g resorcinol and 0.25 g thiourea in 10 ml glacial acetic acid. This is stable in the dark.

Dilute HCl: Mix five part of conc. HCl with one part of distilled water.

Standard fructose solution: Dissolve 50mg of fructose in 50 ml water. Dilute 5 ml of this stock soln. to 50 ml for a working standard.

Preparation of the material: Common minute material for analysis in Waring blender for 1 to 2 minutes. Do not over heat. Pass it through No.60 sieve and put is in airtight container.

Procedure: Take 2 to 3 g sample in duplicate in pre-dried and pre-weighed 7 cm diameter aluminium dishes. Remove the dish cover and put them in vacuum oven and dry for 6 hours at 88°C and 26-22 inch vacuum. During this process, admit to oven a slow current of air (2 bubbles per second) dried by passing H₂SO₄ replace the cover, cool it in desiccators and weigh again.

Principle: Addition of calcium chloride results in the precipitation of pectin as calcium pectate from an acid solution. Calcium pectate is washed with water to make free from chloride and is dried and weighed.

26.1. Aim

To determine the pH of food samples.

26.2. Theory

The hydrogen-ion concentration of a food is a controlling factor in many chemicals and microbiological reactions. Pure water has equal concentration of H+ and OH- ions, each having the value of 1×107g/litre at room temperature that is why water is regarded as neutral. The pH of pure water is 7.0, the solution having pH below 7.0 is regarded as acidic while solution with pH above 7.0 is alkaline. Most fruits and fruit products have pH below 4.0 while vegetable; milk and meat products have pH more than 4.0. The pH value of some fruits and fruit products is given in Table 6.1. Thus, with the increase in acidity in the solution, the pH value decrease and vice versa. However, presence of buffer salts helps in maintenance of pH. Estimation of pH of the food is important as the processing conditions for different food products are categorized on the basis of their pH.

Procedure: Standardize the pH meter with a buffer solution of pH. Take the sample solution in a 100 ml beaker. Put the electrodes in the solution and read on pH meter. After each observation, wipe the electrodes with a piece of cotton soaked in distilled water and rinse the electrodes with water with the help of a wash bottle and dry with filter paper.

Principle: The copper in the Fehling solution is reduced to red, insoluble cuprous oxide in the presence of invert sugars like glucose and fructose. The quantity of sugars solution liquids for complete neutralization of a known quantity of Fehling mixture is determined by titration using methylene blue as the indicator.

(Lane and Eynon Method)

Principle: A weighed sample is dried in an oven at 60-70⁰C up to a period of time when weighing made at 2 hour intervals do not differ by more than 2-3 mg.

23.1. Aim

To determine the total soluble solids in fresh and processed fruit and vegetable products.

23.2. Theory and principle

Total soluble solids (TSS) are determined by means of brix which measures the specific gravity or by using hand refractometer which measures refractive index. Brix is a measure of soluble solids only in case of pure sucrose solutions. Generally, the fruit juices contain more sugar than any other soluble constituent. Hence, brix provides a useful guide of soluble solids or sugar contents. Brix hydrometer is used to determine the percentage of brix in syrup while Abbe refractometer gives refractive index as well as degree brix. The Refractive index of the test solution is measured at 20°C using a refractometer and the soluble solid content is determined by the use of tables correlating refractive index with soluble solids. Generally, hand refractometer is used for determining TSS in food samples in fruit processing units (Table 3.1).

Purpose: The estimation of total soluble constituents gives the approximate amount of water soluble substance present in the sample. Among the various soluble substances the amount of sugar is 80-85 percent. The TSS value is roughly considered to be the amount of sugar and soluble minerals present in fruits and vegetables. The total soluble solid content can be determined by hand refrectometer.

Material and equipments: Hand refrectometer, juice, muslin cloth or tissue paper and glass rod.

Jackfruit is a fruit crop that is under appreciated and neglected. The jackfruit (Artocarpus heterophyllus Lam.) is a member of the Moraceae family. India is the world’s greatest cultivator and producer of jackfruit, also known as the poor man’s fruit because it grows more in rural than metropolitan areas. It is a pretty huge tree that bears the world’s largest edible fruit. The jackfruit is a tropical fruit native to Southeast Asia. It is thought to have originated in India’s rainforests, in the Western Ghats, and is now widely farmed throughout the region’s lowlands. The leading jackfruit producers in the world include Bangladesh, India, Myanmar, Nepal, Thailand, Vietnam, China, the Philippines, Indonesia, Malaysia, and Sri Lanka. Jackfruit can be found in East Africa, Brazil, and Caribbean nations such as Jamaica. Bangladesh’s national fruits are the jackfruit, mango, and banana.

Owing to paucity of available genetic / molecular markers, onion breeding still relies heavily on phenotypic selection methods. DNA markers can provide plant breeders genetic information for indirect selection of plants. All the DNA based procedures require high quality plant genomic DNA. Abundance of polysaccharides and phenolics present in plant tissue often impede PCR amplification. The polysaccharide contaminants can also produce downstream problems by inhibiting many of the enzymes that are employed in subsequent analysis and cloning. RFLP based markers in Allium species pose certain problems due to relatively large size of the genome compared to most other herbaceous crops. As an alternative, RAPD would prove advantageous in genetic analysis of Allium. Here we present our results on optimization of DNA extraction in onion to get amplifiable quality of DNA for further RAPD analysis. Leaf samples were collected from the onion germplasm maintained at Indian Institute of Horticultural Research (IIHR), Bangalore. Dried leaf protocol was suitably modified to obtain fair quality of DNA. After varying each of the ingredients of the extraction buffer we arrived at the optimal extraction buffer, which resulted in good quality DNA. Quality was analyzed by subjecting the DNA to agarose gel electrophoresis and restriction digestion. Extracted genomic DNA was quantified with ‘Hoefer’s Dynaquant’ (Pharmacia, USA). The DNA quantity ranged between 160-2400mg/gm of dried powder. The standardized 25ml PCR mixture contained template DNA (25ng), 5 pmol of primer (Operon Tech. Inc., Alameda), 2mM MgCl₂, I unit of Taq polymerase (Bangalore Genei), with 10x buffer along with 200 mM each of dNTPs. Amplifications were performed in thermal cycler (MJ Research, PTC 100) for 45 cycles after an initial denaturation of 95°C for 3 minutes. In each cycle denaturation for one minute at 95°C, annealing for one minute at 35°C and extension for two minute at 72°C was programmed with final extension step at 72°C for 5 minutes. This fine tuned protocol resulted in clear, informative RAPD profile.

Development of fungicide resistance in a fungal disease pathogen is a major concern for some of the important diseases in their management. This can be overcome by designing of fungicide use strategies to manage the threat of fungicide resistance.

Tomato (Solanum lycopersicum L.) is one of the most important vegetable crops cultivated in the world. It is used as raw in salads and also used as cooked vegetable in the preparation of curries. Processed items such as tomato puree, ketchup, pickle, chutney, whole peeled tomatoes and tomato powder are also consumed considerably. It is a rich source of Vitamins A & C. Lycopene and other flavonoids in tomato serve as good source of antioxidants. Tomato occupies 5 million hectares with a productivity of 34t/ha. in the world. In India it is cultivated in an area of 0.88 million hectares with productivity of 21 t/ha.

ABSTRACT

Cassava has developed importance mainly as an industrial crop due to its ability to store large quantities of starch (74% to 85% of dry weight) within secondary root structures. Cassava starch is exploited as a source of human food, feed for livestock and as raw material for the food processing and chemical industries. Research accomplishments during the last four decades helped India to have the top position among cassava growing countries with an average yield of 24 t ha-1. However several traits like starch quality, high protein , post harvest deterioration etc can be altered only through the development of transgenics. The present study summarizes the results of the experiments undertaken to optimize the protocol for the development of transgenic cassava through Agrobacterium mediated gene transfer. Friable Embryogenic Callus (FEC) was developed in the cassava var. TMS 60444 us ing Murashige and Skoog (MS) media with picloram (50μM). Experiments were conducted to study the efficiency of different Agrobacterium stains viz, AGL 8, GV3101, C58C1, LBA 4404 and EH105 in transforming FEC. The strains differed in their growth rate. The strains EH105 and AGL were slower in growth as compared to other strains and the protocol was modified accordingly. Among all the strains, EH105 resulted in maximum recovery of transgenic tissues. The paper also summarizes the present status of development of transgenic cassava along with future thrusts.

Morphological characteristics

Abroma is a shrub or small tree that grows to a height of around 3-5 metres. The plant has horizontal and velvety branches that are 10-30 cm long, 6-18 cm wide, oblong or lanceolate, more or less cordate, delicately acuminate, membranous, whole, and three to five lobed with a petiole that is 1-12 cm long. The dorsal surface of the leaves is glabrous, while the ventral surface is hairy.

Floral characteristics

The plant has beautiful and intricate scarlet like flowers (5 cm in diameter) bearing purple colour. The inflorescence is a cyme. The sepals are persistent and lanceolate, measuring about 2.5 cm long. The petals are imbricate and will shortly fall off. Stamens are situated on a short staminal tube, and there are five staminodes. Each cell in the ovary is five lobed and pyramidal, with numerous ovules. The apex, the capsules are obpyramidal, membranous, finely pubescent, and truncate at the apex. Behind each carpel is a triangular wing. December and January are the fowering and fruiting months.

ABSTRACT

Diabetes is a chronic endocrinologic disorder characterized by high blood levels of glucose due to insufficient secretion of insulin by the pancreas or improper utilization of insulin by target cells. More than 120 million people from all over the globe are estimated to suffer from this malady. Many plants and their constituents have become chief ingredients of a number of pharmaceutical preparations used against diabetes. An intensive survey undertaken for over 4 years in the tribal belts of Orissa to identify the plants used in folk medicine and the methodologies employed by the herbal healers have revealed that 35 species are being employed to cure diabetes. Out of 313 patients (185 males and 128 females) interviewed, 211 (67.5%) use Gymnema sylvestre, Momordica charantia, Trigonella foenum-graecum, Tinospora cordifolia, Pterocarpus marsupium, Andrographis paniculata and Rauwolfia serpentina in one form or the other regularly. Multi-plant preparations gave 80% results whereas single plant administration gave less than 50% results. Polyherbal treatment is advocated as multi-pronged attack to achieve desired results.

Introduction

Fiber is that part of plant food which is not digested when eaten. Since it is not absorbed into the body, dietary fiber is not a nutrient. Nevertheless, fiber is a very important component of a healthy diet. Fiber, also called “roughage” helps prevent constipation, hemorrhoids and diverticular disease. Some types of fiber have a cholesterol-lowering effect, which could lead to reduced risk of heart disease.

Dietary fiber has been defined as plant cell wall material resistant to the intestinal secretions of the host. This definition gives the appearance of precision and allows gravimetric and chemical measurements. However, a proportion of other non-fibre dietary constituents (e.g, retrograded starch, protein, and fat) pass along the gut, without absorption or metabolism, to the caecum. Chemical analysis or quantitative measurements of the fibre content of specific diets do not allow prediction of their biological action. The physiological effects of dietary fibre depend predominantly on physical properties unrelated in any simple or direct way to chemical composition. Quantitative measurement of dietary fibre does not allow for diverse effects on nutrient absorption, sterol metabolism, fermentation in the colon, and stool weight. We still do not know whether to restrict the definition of “dietary fibre” to non-starch polysaccharides or to include related materials such as resistant starch, lignin, and the complex assemblies typical of plant tissues, where polysaccharides occur in close association with other molecules. An alternative to defining fibre in terms of chemical composition would be to develop measurements of physical properties relevant to physiological action in the intestine. This approach could be extended to other polymeric constituents, in particular proteins (ingested and secreted) and mucous glycoproteins.

Polysaccharides are the main constituents of dietary fibre and show a wide spectrum of physical, properties that reflect the nature and extent of intermolecular association. Dietary fibre can be likened to a water-laden hydrated mass passing along the gut. Physical properties that influence function are a combination of the rheological and colligative properties of the water-soluble fibre components, the surface characteristics of the water-insoluble components, and the network properties of the swollen hydrated components. Such features include viscosity, water holding, cation exchange, organic acid adsorption, gel filtration, and particle size distribution.

1. Introduction

Among three major functions identification, classification and nomenclature of taxonomy (Fig. 1), identification is a core activity and one of the major objectives of taxonomy, which determines whether other two elements are the same or different. Classification and nomenclature are two separate methods in taxonomy, however, both are totally dependent on the correct identification of the objects. The identification of an unidentified plant with a correctly identified specimen also involves classification. So, both identification and classification, involve comparison and judgment of similarities and differences. Therefore, the identification is a primary function in classification involving nomenclature which performs an essential role as a means of communication. According to Blackwelder (1967) “identification enables us to retrieve the appropriate facts from the system (classification) to be associated with some specimen at hand” and is “better described as the recovery side of taxonomy.” The identification of plants is very important for proper assessment and characterization of biodiversity on which the whole life of human beings is dependent starting from birth to death. The use of biodiversity for various purposes like food, nutrition, pharmacognosy etc. requires accurate and reliable identification and naming of plants (Nesbitt et al. 2010).

Digital technologies are transforming horticultural production by enabling data-driven crop management and precise decision-making. This chapter introduces agricultural informatics, sensors, Internet of Things, remote sensing, drones, artificial intelligence, machine learning, geographic information systems, robotics, and precision technologies. These tools can support crop monitoring, disease detection, irrigation scheduling, nutrient management, yield prediction, phenotyping, and resource optimization. AI-based image analysis can identify symptoms and assist in early crop diagnosis, while sensors provide continuous information about soil and plant conditions. The chapter emphasizes the integration of digital data with practical horticultural knowledge to improve productivity, profitability, sustainability, and resilience while reducing unnecessary inputs and improving decision-making.
